    Profiliranje uporabnikov postaja vse bolj pomembna tema pri razvoju spletnih strani, saj omogoča zagotavljanje boljše uporabniške izkušnje z ugotavljanjem uporabnikovih interesov. V tem delu se ukvarjamo z dinamično izbiro metod profiliranja uporabnikov. Cilj je uporabiti metode strojnega učenja in zgraditi učni model, ki bo znal kar najbolje kombinirati metode profiliranja in tako ustvariti kombinirano metodo profiliranja, uspešnejšo od vsake posamezne metode, ki smo jih uporabili pri učenju. Pokazali smo, da je kombiniranje profilirnih algoritmov z uporabo strojnega učenja lahko močno orodje pri izboljšavi uspešnosti profiliranja. Pokazali smo tudi, da je uporaba dinamične izbire metod smiselna v primeru, ko so razlike med posameznimi algoritmi profiliranja večje in so tako tudi možnosti za izboljšave večje.User profiling is becoming an increasingly important subject in the field of web development as it enables improving the user experience through learning the users interests. In this study we examine dynamic selection of web user profiling methods. Our goal is to use machine learning methods to build a learning model that predicts the most successful combined profiling method, which is expected to be significantly better from each individual method. We have shown that combining of profiling methods using machine learning can be a powerful tool when looking for a way of improving the accuracy of web user profiles. We have also shown that dynamic selection is most effective when differences between profiling methods are relatively large and therefore providing room for improvement

    Une enquête empirique (par entretiens et par scénarios) auprès de couples des classes populaires a permis de saisir comment s’organisent les mobili­sations domestiques de l’argent, à la fois en situation ordinaire où leur agencement est apaisé et en situation de tension où il s’agit de sortir de di­lemmes. Les résultats montrent une organisation plutôt minimale des dé­penses, réglées par deux ou trois moyens de paiement principaux, et assu­rées selon une répartition qui semble être explicite même si elles sont surtout surveillées sinon contrôlées par les femmes. Les questions d’argent ne sont pas de l’ordre du non-dit et ne sont pas source de tensions, ce qui semble autoriser la présence d’un sentiment de plaisir de la dépense. Ces com­portements reposent sur un triple fondement d’attitudes : autonomie de la personne et du couple, rationalisation concertée des choix domestiques et prudence consumériste. On peut imaginer que ces potentialités sont à un tournant de la modernité : hybridations fragiles préfigurant des relations « liquides » ou ébauche d’une nouvelle culture populaire ?An empirical survey (using interviews and scenarios) concerning couples of popular classes brings informations on the organization of domestic mobile­zations of money, both in ordinary situations where its management is appeased, and in situations of stress where it is necessary to escape from dilemmas. The outcomes are : a rather minimal organization of expen­ditures, payed by the way of two or three means of payment, explicitly dispatched in the couple, while women are mainly supervising and some­times controlling them. Money issues are not of unvoiced comments and not a source of tension, so seeming to let the expenditure being felt as a pleasure. These behaviours lay on a threefold grounding: a claimed au­tonomy of the person and of the couple, a jointly arranged rationalization of the domestic choices and a prudent consumerism. One can imagine that these potentialities are at the cross-road of modernity: weak hybridations prefiguring « liquid » relationships or outline of a new popular culture

    Les livres de fête sont parmi les plus spectaculaires des nombreux documents rassemblés par Jacques Doucet pour sa bibliothèque d’Art et d’Archéologie, rattachée depuis 2003 à l’Institut national d’histoire de l’art. Comme les fêtes destinées à solenniser chaque acte de la vie publique des souverains, ces livres étaient le produit de la contribution de tous les grands artistes du temps. Baptêmes, mariages, funérailles, couronnements, victoires ou entrées solennelles dans les « bonnes » villes du royaume donnaient lieu à des déploiements de fastes dont les livres se veulent les traces tout aussi fastueuses. L’exposition sur les livres de fête du XVIIe au XVIIIe siècle organisée à l’INHA (galerie Colbert à Paris IIe, du 15 septembre au 15 décembre 2010) présente des livres de cette période illustrant des fêtes versaillaises et parisiennes. Ce catalogue accompagne et éclaire la visité et l’étude de ces ouvrages.     Epilepsy with migrating focal seizures

    To report new sporadic cases and 1 family with epilepsy of infancy with migrating focal seizures (EIMFSs) due to KCNT1 gain-of-function and to assess therapies' efficacy including quinidine. We reviewed the clinical, EEG, and molecular data of 17 new patients with EIMFS and KCNT1 mutations, in collaboration with the network of the French reference center for rare epilepsies. The mean seizure onset age was 1 month (range: 1 hour to 4 months), and all children had focal motor seizures with autonomic signs and migrating ictal pattern on EEG. Three children also had infantile spasms and hypsarrhythmia. The identified KCNT1 variants clustered as "hot spots" on the C-terminal domain, and all mutations occurred de novo except the p.R398Q mutation inherited from the father with nocturnal frontal lobe epilepsy, present in 2 paternal uncles, one being asymptomatic and the other with single tonic-clonic seizure. In 1 patient with EIMFS, we identified the p.R1106Q mutation associated with Brugada syndrome and saw no abnormality in cardiac rhythm. Quinidine was well tolerated when administered to 2 and 4-year-old patients but did not reduce seizure frequency. The majority of the KCNT1 mutations appear to cluster in hot spots essential for the channel activity. A same mutation can be linked to a spectrum of conditions ranging from EMFSI to asymptomatic carrier, even in the same family. None of the antiepileptic therapies displayed clinical efficacy, including quinidine in 2 patients

    Amphiphilic block copolymers enhance the cellular uptake of DNA molecules through a facilitated plasma membrane transport

    Amphiphilic block copolymers have been developed recently for their efficient, in vivo transfection activities in various tissues. Surprisingly, we observed that amphiphilic block copolymers such as Lutrol® do not allow the transfection of cultured cells in vitro, suggesting that the cell environment is strongly involved in their mechanism of action. In an in vitro model mimicking the in vivo situation we showed that pre-treatment of cells with Lutrol®, prior to their incubation with DNA molecules in the presence of cationic lipid, resulted in higher levels of reporter gene expression. We also showed that this improvement in transfection efficiency associated with the presence of Lutrol® was observed irrespective of the plasmid promoter. Considering the various steps that could be improved by Lutrol®, we concluded that the nucleic acids molecule internalization step is the most important barrier affected by Lutrol®. Microscopic examination of transfected cells pre-treated with Lutrol® confirmed that more plasmid DNA copies were internalized. Absence of cationic lipid did not impair Lutrol®-mediated DNA internalization, but critically impaired endosomal escape. Our results strongly suggest that in vivo, Lutrol® improves transfection by a physicochemical mechanism, leading to cellular uptake enhancement through a direct delivery into the cytoplasm, and not via endosomal pathways

    Resistance to Gemcitabine in Pancreatic Cancer Is Connected to Methylglyoxal Stress and Heat Shock Response

    peer reviewedPancreatic ductal adenocarcinoma (PDAC) is a fatal disease with poor prognosis. Gemcitabine is the first-line therapy for PDAC, but gemcitabine resistance is a major impediment to achieving satisfactory clinical outcomes. This study investigated whether methylglyoxal (MG), an oncometabolite spontaneously formed as a by-product of glycolysis, notably favors PDAC resistance to gemcitabine. We observed that human PDAC tumors expressing elevated levels of glycolytic enzymes together with high levels of glyoxalase 1 (GLO1), the major MG-detoxifying enzyme, present with a poor prognosis. Next, we showed that glycolysis and subsequent MG stress are triggered in PDAC cells rendered resistant to gemcitabine when compared with parental cells. In fact, acquired resistance, following short and long-term gemcitabine challenges, correlated with the upregulation of GLUT1, LDHA, GLO1, and the accumulation of MG protein adducts. We showed that MG-mediated activation of heat shock response is, at least in part, the molecular mechanism underlying survival in gemcitabine-treated PDAC cells. This novel adverse effect of gemcitabine, i.e., induction of MG stress and HSR activation, is efficiently reversed using potent MG scavengers such as metformin and aminoguanidine. We propose that the MG blockade could be exploited to resensitize resistant PDAC tumors and to improve patient outcomes using gemcitabine therapy
