Untersuchung Nanopartikel-katalysierter organischer Synthesen im Mikrodurchflussprozess
ï»żThe development of heterogeneous catalysts for fine chemicals synthesis
has become a major area of research recently, because the potential
advantages of these materials (their simplified recovery and reusability,
their potential for insertion in continuous microreactors) compared to
homogeneous systems can have a major impact on the environmental
performance of an organic synthesis[a2]. In this context, the development
of a strategy based on the deposition of gold nanoparticles on alumina by a
colloidal coating process without calcination and their implementation as
heterogeneous catalyst in multistep syntheses of propargylamines,
pyridines, and polypyridines was the main objective of this thesis. A
modular continuous flow micro reaction assembly consisting of two
subsequent packed-bed capillary reactors (PBCR) fed with educt solutions
was developed to study the following different multi-compound reactions
(figure A1). The two-step reaction sequence known as A3-reaction[a3] for
the formation of propargylamine derivatives (3d) was investigated. The
reaction sequence consisted of an initial condensation reaction of
aldehydes (3a) with secondary amines (3b) yielding enamine intermediates.
Following addition of phenylacetylene (3c) led to the desired
propargylamine derivatives (3d) under appropriate catalytic activation.
Different heterogeneous catalysts were investigated to force the overall
reaction performance. About 50 different propargylamine derivatives of type
3d were synthesized using Montmorillonite K10 clay for PBCR1 and alumina
supported gold nanoparticles in PBCR2.The same catalyst combination was
used to optimize the formation of pyridines (1c) in the modular continuous
flow micro reaction system. The pyridine forming reaction sequence was
studied with different methylketones 1a under varying reaction conditions.
In case of bivalent methylketones 2a, the ratio between the acetylpyridine
and the bipyridine product building can be adjusted by tuning of the
stoichiometric ratio of the starting materials 2a and 2b. About 85 %
turnover in case of terpyridine was achieved. Furthermore, the introduced
reaction enables the formation of different polypyridine ligands by easy
optimization of the reaction conditions for various bivalent ketones[a4].In
summary, alumina supported gold nanoparticles possess an excellent
catalytic performance in the investigated reactions. In addition,
significant process intensification was achieved in terms of a reaction
time reduction from 24 h down to about 30 min. This demonstrates the great
potential of the combination of continuous flow processes with the
heterogeneous catalysis for the synthesis of fine chemicals by optimizing
multi-component reactions using separately improved connected elementary
reaction steps.In der letzten Zeit ist die Entwicklung heterogener Katalysatoren fĂŒr
die Feinchemikaliensynthese ein Hauptgebiet der Forschung geworden.
Potenzielle Vorteile dieser Materialien sind ihre vereinfachte
RĂŒckgewinnung, Wiederverwendbarkeit und ihr möglicher Einsatz in
Mikrodurchflussreaktoren im Vergleich zu homogenen Systemen. Diese Vorteile
können drastische Auswirkungen auf eine organische Synthese haben[a2]. In
diesem Kontext war das Hauptziel dieser Dissertation die Entwicklung einer
Strategie zur Immobilisierung kolloidaler Gold Nanopartikel ohne
Kalzinierung auf Aluminiumoxid sowie deren möglicher Einsatz als
heterogener Katalysator in mehrstufigen Synthesen von Propargylamin-,
Pyridin- und Polypyridin Derivaten.Ein modularer kontinuierlicher
Mikrodurchflussaufbau, welcher aus zwei hintereinander geschalteten
Festbettkapillarreaktoren (PBCR) bestand und durch den die Eduktlösungen
gefördert worden sind, wurde entwickelt, um folgende
Mehrkomponentenreaktionen untersuchen zu können (Abbildung A1). ZunÀchst
wurde die als A3-Reaktion[a3] bekannte Zweistufenreaktion zur Synthese von
Propargylamin-Derivaten (3d) untersucht. Die Reaktionsreihenfolge besteht
im ersten Teilschritt aus einer Kondensationsreaktion eines Aldehyds (3a)
mit sekundĂ€ren Aminen (3b), welche zu Enamin Intermediaten fĂŒhrt. Eine
nachgeschaltete Addition des Phenylacetylens (3c) fĂŒhrt unter geeigneter
katalytischer Aktivierung zu den gewĂŒnschten Propargylamin-Derivaten (3d).
Verschiedene heterogene Katalysatoren wurden bei der Prozessoptimierung
untersucht. Ăber 50 Propargylamin-Derivate vom Typ 3d wurden durch den
Einsatz des Montmorillonit K10 im PBCR1 und von auf Aluminiumoxid
immobilisierten Gold-Nanopartikel im PBCR2 synthetisiert. Die gleiche
Katalysatorenkombination wurde auch benutzt, um die Bildung von Pyridinen
(1c) im Mikrodurchflussprozess optimieren zu können. Die Pyridinreaktion
wurde mit verschiedenen Methylketonen 1a unter unterschiedlichen
Reaktionsbedingungen untersucht. Im Fall der bis-Methylketone, wie
beispielsweise Verbindung 2c, konnte das VerhÀltnis zwischen der
Acetylpyridin- und Bipyridinproduktbildung durch eine gezielte Ănderung
des stöchiometrischen VerhÀltnisses der eingesetzten Materialien 2a und
2b eingestellt werden. Ăber 85 % Reaktionsumsatz wurden im Fall der
Terpyridinliganden erreicht. AuĂerdem ermöglichte diese Reaktion, durch
eine leichte Optimierung der Reaktionsbedingungen fĂŒr verschiedene
bis-Ketone, die Bildung verschiedener Pyridinliganden[a4].Auf Aluminiumoxid
immobilisierte Gold-Nanopartikel waren in der Lage, alle untersuchten
Reaktionen zu katalysieren. Zudem wurde eine signifikante
Prozessintensivierung erreicht. So konnten die Reaktionszeiten von 24
Stunden auf etwa 30 min verringert werden. Dieses demonstrierte den Erfolg
eines kombinierten Einsatzes von Mikrodurchflussprozessen mit der
heterogenen Katalyse fĂŒr die Feinchemikaliensynthese, der auch durch eine
Optimierung von Multikomponenten-Reaktionen erfolgte, in dem diese in
elementare, gekoppelte Reaktionsschritte zerlegt wurden
Rapid WolffâKishner reductions in a silicon carbide microreactor
WolffâKishner reductions are performed in a novel silicon carbide microreactor. Greatly reduced reaction times and safer operation are achieved, giving high yields without requiring a large excess of hydrazine. The corrosion resistance of silicon carbide avoids the problematic reactor compatibility issues that arise when WolffâKishner reductions are done in glass or stainless steel reactors. Kinetic and Scale-up Investigations of a Michael Addition in Microreactors
Microreactors are an efficient tool
for process development and
intensification. However, the scale-up from lab studies to small-scale
commercial production is challenging, since a change in the channel
dimensions requires good knowledge of heat and mass transfer phenomena.
In this work, complete process development for an exothermic Michael
addition is presented. In a systematic scale-up approach, kinetic
studies and experimental characterization of the employed reactors
provide key parameters for detailed reactor modelling. The residence
time distribution, reactant mixing, and removal of reaction heat are
taken into account. It is exemplarily shown how preliminary experiments
can be the basis for the prediction of scale-up effects and the development
of a continuous production process. Plug flow behavior and short mixing
times could be confirmed for all investigated flow reactors. Furthermore,
interactions of reaction kinetics and the formation of hot spots in
the reactor channel were investigated. For the examined reaction,
the simulations predicted the product yield under production conditions
in good accuracy
