    Fishery biology of the swordfish (Xiphias gladius, Linnaeus 1758) caught by surface longliners based in Itajaí, southern Brazil

    No presente estudo foram analisados os dados obtidos para Xiphias gladius (Linnaeus, 1758) através de observadores, mapas de bordo e fichas de desembarque de carcaças, coletadas pela frota de espinhel-de-superfície sediada em Itajaí (SC). Modelos de análise de covariância (ANCOVA) foram aplicados aos dados de captura (peso em kg) do espadarte, tendo como efeitos: (1) anos de 1997, 1998, 2001, 2002 e 2007 (2) fases lunares e (3) épocas do ano. A covariável foi o esforço de pesca (n° de anzóis). As maiores médias de captura ocorreram durante as fases de lua crescente e no inverno. O procedimento estatístico de Mantel-Haenszel foi utilizado para comparar as capturas do espadarte no anzol tipo "Jota" (J 9/0 10º offset), comumente utilizado pela frota, com o anzol tipo "circular" (18/0 10º offset), este último oferecido como alternativa mitigadora à captura de tartarugas marinhas. O teste M-H mostrou que o anzol "Jota" apresentou 1,2 mais chances de capturar Xiphias gladius que no "circular". A conversão dos pesos individuais das carcaças (kg) em comprimentos totais (cm), referentes aos anos de 2000 a 2002, indicou que 66% das capturas foram de sub-adultos ao longo de todo o ano.The Xiphias gladius (Linnaeus, 1758) data used in the present study were obtained from commercial fishing cruises of the pelagic longline fleet based in Itajaí, operating in the Southwest Atlantic. For the Covariance models, fishing effort (number of hooks) and catches (kg) were pooled and categorized into the following groups: (1) year (1997, 1998, 2001, 2002 and 2007); (2) lunar phase; and (3) seasons, fishing effort being the covariate. The highest mean swordfish catch (kg) occurred during the waxing crescent moon and during wintertime. The Mantel-Haenszel procedure was applied to compare the efficiency of the two hooks tested, and showed that the "J" hook type (J 9/0 10º offset), traditionally used by the fleet, catches 1.2-fold more Xiphias gladius than the circle hook (18/0 10º offset), used as an optional device to reduce sea turtle bycatch. The conversion of individual dressed weight (kg) into total length (cm) from 2000 to 2002, showed that sub-adults represented 66% of the total catch

    Properties of Intra-group Stars and Galaxies in Galaxy Groups: "Normal" versus "Fossil" Groups

    Cosmological LCDM simulations of 12 M_vir~10^14 Msun galaxy groups have been performed, invoking star formation, chemical evolution with non-instantaneous recycling, metallicity dependent radiative cooling, strong star-burst driven galactic super-winds and effects of a meta-galactic UV field. At z=0, intra-group light (IGL) fractions are found to be 12-45%. Low values refer to groups with only a small difference between the R-band magnitudes of the first and second ranked group galaxy, large are typical of "fossil" groups (FGs). The IG stars in the 4 FGs are 0.3-0.5 Gyr older than in the 8 nonFGs. For the IGL, B-R=~1.4, in good agreement with observations. For FGs/nonFGs the iron abundance of the IG stars is slightly sub-solar in the central parts (r~100 kpc) decreasing to about 40% solar at about 0.5 r_vir The IG stars are alpha-element enhanced with [O/Fe] increasing with r, and an overall [O/Fe]~0.45, indicating predominant SNII enrichment. The velocity distributions of the IG stars and group galaxies are, at r>~30 kpc, significantly more radially anisotropic for FGs than for nonFGs. So a characteristic of FG formation, apart from formation time (D'Onghia et al.), may be the "initial" velocity distribution of the group galaxies. For FGs one can dynamically infer the (dark matter dominated) mass distribution of the groups all the way to r_vir, from the kinematics of the IG stars or group galaxies. For the nonFGs this method overestimates the group mass at r>~200 kpc, by up to a factor of two at r_vir. This is interpreted as FGs being, in general, more relaxed than nonFGs. Finally, FGs of the above M_vir should host ~500 planetary nebulae at projected distances between 100 and 1000 kpc from the first ranked galaxy.     Study of ‘Fingerprints’ for Green Tea from Different Planting Areas in Eastern China

    Green tea is one of the main teas in China, which is unfermented and retains more natural substances of fresh tea leaves. This is the preliminary study of  application of ‘fingerprints’ based on differences in component composition of green tea. Five green teas from different areas in eastern China are  analyzed, which are processed by microwave-assisted solvent (ethanol) extraction method to obtain tea polyphenols, flavonoids, polysaccharides,  pigments (thearubigins, theaflavins, theabrownins). The results show that the component composition of five green teas are varied from each other;  based on these contents varieties, we have constructed a ‘fingerprint’ and applied linear discriminant analysis (LDA) and hierarchical cluster analysis  (HCA) to assist in the identification of these five green teas. This method does not require large, expensive instruments (such as high performance liquid  chromatograph, gas chromatograph, mass spectrometer, etc.), and is easy to use, which provides a new avenue for the identification of tea.&nbsp

    El presente trabajo está referido a la investigación, discusión y desarrollo de un Centro Cultural Especializado en Danzas Folclóricas Tradicionales en el distrito de Cajamarca, cuyo propósito es evidenciar y atender a las necesidades que demandan los usuarios principales, es decir, los danzantes informales. Se hizo estudiando estrategias de acondicionamiento, aislamiento y confort acústico para el diseño del proyecto. Se aplicó un diseño experimental, el cual consistió en la recolección de datos mediante encuestas realizadas a los danzantes formales e informales del distrito y ciudad de Cajamarca, los cuales nos arrojaron datos específicos sobre agrupaciones, tiempo y tipos de danzas. Estos datos fueron fundamentales para la factibilidad y veracidad del estudio. Los resultados evidenciaron con claridad la necesidad de un centro que pueda acoger a estas personas, para que puedan desarrollar su actividad artística con calidad. De acuerdo a todo lo descrito y expuesto líneas arriba concluimos en la idea de que diseñar ambientes óptimos para la realización de la actividad dancística, era un proyecto viable que permitirá aportar bienestar, confort y seguridad a quienes cultivan el arte de la danza y a todas las personas en general

    Simulating galaxy Clusters -II: global star formation histories and galaxy populations

    Cosmological (LambdaCDM) TreeSPH simulations of the formation and evolution of galaxy groups and clusters have been performed. The simulations invoke star formation, chemical evolution with non-instantaneous recycling, metal dependent radiative cooling, strong star burst and (optionally) AGN driven galactic super winds, effects of a meta-galactic UV field and thermal conduction. The properties of the galaxy populations in two clusters, one Virgo-like (T~3 keV) and one (sub) Coma-like (T~6 keV), are discussed. The global star formation rates of the cluster galaxies are found to decrease very significantly with time from redshift z=2 to 0, in agreement with observations. The total K-band luminosity of the cluster galaxies correlates tightly with total cluster mass, and for models without additional AGN feedback, the zero point of the relation matches the observed one fairly well. The match to observed galaxy luminosity functions is reasonable, except for a deficiency of bright galaxies (M_B < -20), which becomes increasingly significant with super-wind strength. Results of a high resolution test indicate that this deficiency is not due to ``over--merging''. The redshift evolution of the luminosity functions from z=1 to 0 is mainly driven by luminosity evolution, but also by merging of bright galaxies with the cD. The colour--magnitude relation of the cluster galaxies matches the observed "red sequence" very well and, on average, galaxy metallicity increases with luminosity. As the brighter galaxies are essentially coeval, the colour--magnitude relation results from metallicity rather than age effects, as observed.Comment: 15 pages, 10 figures.     Study of an unusually high level of N-glycolylneuraminic acid (NGNA) sialylation on a monoclonal antibody expressed in Chinese hamster ovary cells

    Sialic or neuraminic acids of recombinant therapeutic glycoproteins produced in mammalian cells, including monoclonal antibodies, have significant impact on the half-life, stability, and biological activity of these proteins (Hossler et al., 2009; Ghaderi et al., 2012). The predominant sialic acid N-acetylneuraminic acid (NANA or Neu5Ac) is added from precursor CMP-NANA to galactose residues of N-linked glycoproteins by sialytransferases. In most mammals CMP-NANA can also be modified to its hydroxylated derivative CMP-NGNA by CMP-N-acetylneuraminic acid hydroxylase (CMAH). NGNA can then be added from CMP-NGNA to galactose residues of the N-linked glycoproteins, also by sialytransferases. However, humans cannot make functional CMAH due to an inactivating exon deletion mutation in CMAH gene (Okerblom and Varki, 2017), and therefore cannot convert CMP-NANA to CMP-NGNA. Consequently, when injected into human patients, NGNA sialic acid containing mAbs or other recombinant glycoproteins may induce immune responses, which could negatively impact pharmacokinetics or efficacy. Therefore high levels of NGNA on therapeutic mAbs or other recombinant glycoproteins are an undesirable product quality attribute. The level of total sialic acids of recombinant glycoproteins produced in Chinese hamster ovary (CHO) cells is dictated largely by the selected cell lines, upstream process, and to a lesser degree, downstream process. NGNA sialylation is generally rare in CHO cells (Könitzer et al., 2015). Hence, therapeutic glycoproteins manufactured in these cells are considered safe for human use. However, during a first-in-human (FIH) upstream process development for a novel mAb, an initially selected desirable cell line (A) was found to produce the mAb with an unexpectedly high level of the NGNA sialic acid (\u3e30%). To the best of our knowledge such high level of NGNA sialylation on a mAb produced by CHO cells has not been reported. To mitigate potential risks associated with high NGNA in human patients, a new cell line (B) that produces the mAb with very low NGNA was selected as the manufacturing cell line for this project. In order to understand the molecular mechanism causing the high NGNA content in cell line A, we initiated comprehensive genetic gap analyses using next-generation sequencing technologies and determined the differences in genomic, transcriptomic, and miRnomic profiles of the two cell lines. The results indicate spontaneous upregulation of CMAH mRNA expression, at least 10 fold higher in cell line A compared to cell line B.
