    A high resolution electrochemical impedance spectroscopy study on anode supported single cells (ASC) is presented. The cells were characterised over a broad range of operating conditions, including different temperatures, current densities and various cathode and anode gas compositions. The analysis of the distribution of relaxation times combined with the numeric accuracy of a CNLS fit enabled the identification of five different processes contributing to the total polarisation loss of an ASC

    A collagen based scaffold for the repair of annulus fibrosus defects

D.)--Massachusetts Institute of Technology, Dept. of Mechanical Engineering, 2007.Includes bibliographical references.The intervertebral disc (IVD) is the natural cartilaginous cushion found between the osseous vertebrae of the spinal column. It is an essential element for the flexibility of the spine but undergoes major degeneration with age, resulting in the loss of its functionality and in back and leg pain. Due to the large size of the IVD, its avascular nature and the difficulty for nutrients and waste particles to diffuse in or out of the IVD, damage and defects incurred by the IVD, be they natural or accidental, are very unlikely to heal and often trigger faster degeneration. The annulus fibrosus (AF) defines the outer boundaries of the disc and provides the structural integrity of the IVD. The purpose of this thesis was to evaluate the regenerative potential of a collagen type II scaffold on standardized defects of the AF. Specifically, this thesis evaluated in vitro the effects of the culture conditions, of a select group of growth factors, of the cross-linking method and of the biochemical composition of the scaffold, in the perspective of preparing a suitable autologous cell-seeded implant for a goat animal model. Cross-linking method and the presence of GAG dramatically affected cell proliferation and new tissue synthesis, with the most promising results obtained with dehydrothermal treatment (DHT) cross-linking and with scaffolds containing 5 to 10% of chrondroitin-6-sulfate.(cont.) Autologous cells seeded in this promising scaffold and cultured in a serum-free environment supplemented with a mix of Transforming Growth Factor-[beta]1 and Fibroblast Growth Factor-2, maintained annulus cell morphology, GAG and type II': collagen synthesis, while exhibiting controlled contraction and a significant amount of new synthesized tissue just after a week in culture. Although culture in a rotating bioreactor showed encouraging results, lack of reliability in the replication of the experiments as well as uniformity in the resulting scaffold geometry and poor cell distribution in the scaffold, did not enable this culture condition to be a good candidate for implantation. To investigate the regenerative potential of this implant, an autologous AF cell-seeded scaffold was cultured for 10 days in a DHT cross-linked scaffold in the serum-free culture medium mentioned above. At this time, the implant consisted in a loose matrix of connective tissue mostly composed of fibrochondrocytic cells and type I and II collagens. When surgically implanting this scaffold in a standardized defect of the annulus fibrosus at the L4-L5 lumbar disc level, results showed slight reduction in fibrous tissue and increase in the fibrocartilaginous tissues 2 months postsurgery compared to controls or controls where a scaffold without cells was used.(cont.) However, there was no significant improvement in the gross morphology of the disc. The results of this thesis recommend the use of an autologous AF cell-seeded type II collagen scaffold in the perspective of regeneration of defects of the annulus fibrosus. Future animal experiments should aim at locating the cells in the implant, evaluate the mechanical properties of the regenerated tissue and compare AF cells to stem cells that are easier to be harvested.by Leonide C. The Lazio region is drawing up a new strategy for sustainable energy that aims to define the necessary conditions for development of a regional energy system increasingly turned to the use of renewable sources and efficient energy use as a means for greater environmental protection, in particular for the purpose of reduction of greenhouse gases (GHG).     Macrofauna edáfica em sistemas de integração lavoura-pecuária num Latossolo Vermelho do Cerrado

    The objective of this work was to assess the effects of integrated crop-livestock systems, associated with two tillage and two fertilization regimes, on the abundance and diversity of the soil macrofauna. Four different management systems were studied: continuous pasture (mixed grass); continuous crop; two crop-livestock rotations (crop/pasture and pasture/crop); and native Cerrado as a control. Macrofauna was sampled using a modified Tropical Soil Biology and Fertility method, and all individuals were counted and identified at the morphospecies level for each plot. A total of 194 morphospecies were found, distributed among 30 groups, and the most representative in decreasing order of density were: Isoptera, Coleoptera larvae, Formicidae, Oligochaeta, Coleoptera adult, Diplopoda, Hemiptera, Diptera larvae, Arachnida, Chilopoda, Lepidoptera, Gasteropoda, Blattodea and Orthoptera. Soil management systems and tillage regimes affected the structure of soil macrofauna, and integrated crop-livestock systems, associated with no-tillage, especially with grass/legume species associations, had more favorable conditions for the development of "soil engineers" compared with continuous pasture or arable crops. Soil macrofauna density and diversity, assessed at morphospecies level, are effective data to measure the impact of land use in Cerrado soils.O objetivo deste trabalho foi avaliar os efeitos de sistemas de integração lavoura-pecuária, associados a dois tipos de preparo e de fertilização do solo, sobre a abundância e a diversidade da macrofauna edáfica. Quatro sistemas de manejo foram estudados: pastagem contínua de gramíneas; lavoura contínua de culturas anuais; dois sistemas integrados lavoura-pecuária (lavoura/pastagem e pastagem/lavoura); e Cerrado nativo (controle). A macrofauna foi avaliada pelo método "Tropical Soil Biology and Fertility" modificado e todos os indivíduos coletados nas parcelas foram contados e identificados ao nível de morfoespécies. O total de 194 morfoespécies foi encontrado, distribuído entre 30 grandes grupos, dos quais os mais representativos em ordem decrescente de densidade foram: Isoptera, larvas de Coleoptera, Formicidae, Oligochaeta, adultos de Coleoptera, Diplopoda, Hemiptera, larvas de Diptera, Arachnida, Chilopoda, Lepidoptera, Gasteropoda, Blattodea e Orthoptera. Os sistemas de uso e de preparo do solo afetaram a estrutura das comunidades da macrofauna edáfica. Os sistemas de integração lavoura-pecuária, associados ao plantio direto na fase lavoura, especialmente com a rotação gramíneas/leguminosas, tiveram as melhores condições para o desenvolvimento das espécies de "engenheiros do solo" em comparação às pastagens e às culturas contínuas. A densidade e a diversidade da macrofauna, avaliadas em nível de morfo-espécies, são dados eficientes para medir o impacto de diferentes sistemas de uso da terra, em solos do Cerrado

    Anabolizer vs Reprodução em Equinos

    The use of anabolic agents in equines aims to improve weight gain, energy levels and muscle mass, especially in horses recovering from disease, surgery, overexertion, among others. Adverse effects on equine reproduction as a consequence of inadequate handling of these drugs should be taken into account. In mares it can generate ovarian dysfunction, alterations in the estrous cycle and development of masculine characteristics. On the other hand, in stallions it results in decreased testicular mass, alteration in sperm production and behavioral changes, which can be reversible or irreversible depending on the age, sex, route, dose and frequency of administration of the anabolic agent. This work compiles information on the effects of anabolic agents on equine reproduction, with the purpose of generating awareness on the use of these substances and creating prevention policies on the indiscriminate application of these drugs.El uso de agentes anabólicos en equinos tiene como objetivo mejorar la ganancia de peso, los niveles de energía y la masa muscular especialmente en caballos que se están recuperando de alguna enfermedad, cirugía, sobreesfuerzo, entre otros. Deben tenerse en cuenta los efectos adversos en la reproducción equina como consecuencia del manejo inadecuado de estos fármacos. En yeguas puede generar disfunción ovárica, alteraciones en el ciclo estral y desarrollo de características masculinas. Por otra parte, en los sementales tiene como resultado la disminución de la masa testicular, la alteración en la producción espermática y cambios comportamentales, que pueden ser reversibles o irreversibles teniendo en cuenta la edad, el sexo, la vía, la dosis y la frecuencia de administración del anabolizante. Este trabajo recopila información sobre los efectos de los anabolizantes en la reproducción equina, con el propósito de generar conciencia en el uso de estas sustancias y crear políticas de prevención en la aplicación indiscriminada de estos fármacos.A utilização de anabolizantes em cavalos visa melhorar o ganho de peso, os níveis de energia e a massa muscular, principalmente em cavalos que estão em recuperação de doença, cirurgia, esforço excessivo, entre outros. Os efeitos adversos na reprodução equina decorrentes do manejo inadequado desses medicamentos devem ser levados em consideração. Em éguas pode causar disfunção ovariana, alterações no ciclo estral e desenvolvimento de características masculinas. Por outro lado, em garanhões resulta em diminuição da massa testicular, alteração na produção de espermatozóides e alterações comportamentais, que podem ser reversíveis ou irreversíveis tendo em conta a idade, sexo, via, dose e frequência de administração do anabolizante. Este trabalho compila informações sobre os efeitos dos anabolizantes na reprodução equina, com o objetivo de conscientizar sobre o uso dessas substâncias e criar políticas de prevenção à aplicação indiscriminada dessas drogas
