    Genetic variation in the emblematic Puya raimondii (Bromeliaceae) from HuascarĂĄn National Park, Peru

    Puya raimondii, the giant Peruvian and Bolivian terrestrial bromeliad, is an emblematic endemic Andean species well represented in HuascarĂĄn National Park in Peru. This park is the largest reserve of puna (high altitude plateau) vegetation. The objective of this study is to report on genetic variation in populations of P. raimondii from HuascarĂĄn and neighboring areas. AFLP profiles with four selective primer combinations were retrieved for 60 individuals from different zones. Genetic variability was estimated and a total of 172 bands were detected, of which 79.1% were polymorphic loci. The results showed genetic differentiation among populations, and gene flow. A cluster analysis showed that individuals of P. raimondii populations located in different mountain systems could be grouped together, suggesting long distance dispersal. Thus, conservation strategies for P. raimondii have to take into account exchange between populations located far apart in distance in order to preserve the genetic diversity of this showy species

    Il paper s’interroga su come rimettere in moto alcuni processi di trasformazione urbana che si sono fermati a causa della scarsità di risorse, il cui precipitato si è materializzato in vuoti urbani, servizi non offerti, processi di coesione interrotti. Tale approccio presuppone un duplice spostamento del punto di vista, nella distanza del punto d’osservazione, perché i quartieri pubblici presi in considerazione si inseriscono nel funzionamento di una porzione di territorio più ampia; e nell’oggetto osservato, rivolgendo l’attenzione non solo allo spazio “fisico” ma anche a quello abitato e delle politiche. Il caso studio del PEEP Circus di Venezia s’inserisce nel contesto tipico dei territori dispersi del Nord-Est, nel quale l'introduzione di edilizia residenziale pubblica ha seguito le stesse regole di quella privata. Nonostante siano state portate avanti alcune operazioni di miglioramento sia fisico che sociale da parte dell’amministrazione locale, è mancato un coordinamento dei diversi tipi di azione. Ne è risultato che lo spazio aperto collettivo del Circus continua a rimanere vuoto, presa di distanza, sociale oltre che fisica, da parte dei suoi abitanti

    Discovery of the cancer stem cell related determinants of radioresistance

    AbstractTumors are known to be heterogeneous containing a dynamic mixture of phenotypically and functionally different tumor cells. The two concepts attempting to explain the origin of intratumor heterogeneity are the cancer stem cell hypothesis and the clonal evolution model. The stochastic model argues that tumors are biologically homogenous and all cancer cells within the tumor have equal ability to propagate the tumor growth depending on continuing mutations and selective pressure. By contrast, the stem cells model suggests that cancer heterogeneity is due to the hierarchy that originates from a small population of cancer stem cells (CSCs) which are biologically distinct from the bulk tumor and possesses self-renewal, tumorigenic and multilineage potential. Although these two hypotheses have been discussed for a long time as mutually exclusive explanations of tumor heterogeneity, they are easily reconciled serving as a driving force of cancer evolution and diversity. Recent discovery of the cancer cell plasticity and heterogeneity makes the CSC population a moving target that could be hard to track and eradicate. Understanding the signaling mechanisms regulating CSCs during the course of cancer treatment can be indispensable for the optimization of current treatment strategies

    Identification of an Amylomaltase from the Halophilic Archaeon Haloquadratum walsbyi by Functional Metagenomics: Structural and Functional Insights

    Amylomaltases are prokaryotic 4-α-glucanotransferases of the GH77 family. Thanks to the ability to modify starch, they constitute a group of enzymes of great interest for biotechnological applications. In this work we report the identification, by means of a functional metagenomics screening of the crystallization waters of the saltern of Margherita di Savoia (Italy), of an amylomaltase gene from the halophilic archaeon Haloquadratum walsbyi, and its expression in Escherichia coli cells. Sequence analysis indicated that the gene has specific insertions yet unknown in homologous genes in prokaryotes, and present only in amylomaltase genes identified in the genomes of other H. walsbyi strains. The gene is not part of any operon involved in the metabolism of maltooligosaccharides or glycogen, as it has been found in bacteria, making it impossible currently to assign a precise role to the encoded enzyme. Sequence analysis of the H. walsbyi amylomaltase and 3D modelling showed a common structure with homologous enzymes characterized in mesophilic and thermophilic bacteria. The recombinant H. walsbyi enzyme showed starch transglycosylation activity over a wide range of NaCl concentrations, with maltotriose as the best acceptor substrate compared to other maltooligosaccharides.     The artificial introduction in the soil of antagonistic microorganisms can be a successful strategy, alternative to agrochemicals, for the control of the root-knot nematodes (Meloidogyne spp.) and for preserving plant health. On the other hand, plant roots and the associated rhizosphere constitute a complex system in which the contribution of microbial community is fundamental to plant health and development, since microbes may convert organic and inorganic substances into available plant nutrients. In the present study, the potential nematicidal activity of the biopesticide Aphanocladium album (A. album strain MX-95) against the root-knot nematode Meloidogyne javanica in infected tomato plants was investigated. Specifically, the effect of the A. album treatment on plant fitness was evaluated observing the plant morphological traits and also considering the nematode propagation parameters, the A. album MX-95 vitality and population density. In addition, the treatment effects on the rhizosphere microbiome were analysed by a metabarcoding procedure. Treatments with A. album isolate MX-95 significantly decreased root gall severity index and soil nematode population. The treatment also resulted in increased rhizosphere microbial populations. A. album MX-95 can be favourably considered as a new bionematicide to control M. javanica infestation

    Identification and characterization of the sucrose synthase 2 gene (Sus2) in durum wheat

    Sucrose transport is the central system for the allocation of carbon resources in vascular plants. Sucrose synthase (SUS), which reversibly catalyzes sucrose synthesis and cleavage, represents a key enzyme in the control of the flow of carbon into starch biosynthesis. In the present study the genomic identification and characterization of the Sus2-2A and Sus2-2B genes coding for SUS in durum wheat (cultivars Ciccio and Svevo) is reported. The genes were analyzed for their expression in different tissues and at different seed maturation stages, in four tetraploid wheat genotypes (Svevo, Ciccio, Primadur, and 5-BIL42). The activity of the encoded proteins was evaluated by specific activity assays on endosperm extracts and their structure established by modeling approaches. The combined results of sucrose synthase 2 expression and activity levels were then considered in the light of their possible involvement in starch yield

    Impedimetric Biosensor Based on a Hechtia argentea Lectin for the Detection of Salmonella spp.

    A sensitive electrochemical detection method for Salmonella spp. was described, based on the use of Hechtia argentea lectin immobilised on a screen-printed gold electrode. The lectin was extracted from Hechtia argentea, a plant belonging to the Bromeliaceae family. The lectin with molecular weight near 27.4 kDa showed selectivity towards D-mannose, contained on the lipopolysaccharide cell wall of Salmonella spp. Carbohydrate selectivity of the lectin was measured as a change in impedance with respect to concentration. The binding of the bacteria to the biosensor surface increased impedance with increasing concentrations of Salmonella spp., achieving a linear range of detection of 15–2.57 × 107 CFU mL−1, with a limit of detection of 5 CFU mL−1. Increases in impedance were measured using electrochemical impedance spectroscopy and analysed using Nyquist plots. The biosensor was applied in analysis of hen egg samples, and the results were consistent with those obtained using the official analysis methodologyS

    The Reserva de la Biosfera Barranca de MetztitlĂĄn (Hidalgo): An illustrated checklist of bromeliads and orchids and their high levels of Mexican endemisms

    This study presents a list of species of the two most important families with epiphytic elements, Bromeliaceae and Orchidaceae, from the Reserva de la Biosfera Barranca de MetztitlĂĄn (RBBM), the largest Reserve in Hidalgo, Mexico. Thirty-four species are included, 26 corresponding to species in three genera of bromeliads, and eight species in six genera of orchids. The new records represent 26.5% of the total listed in the area; nine of them are new records for the Reserve (RBBM) and one is new for Hidalgo State. This study reveals that endemism for both families is very important in the Reserve (55.88%), since it includes 13 Mexican bromeliads, of which two are endemic to Hidalgo and one to the Reserve, and three orchids, two endemic to Mexico and one to the Reserve. We found species with different types of relative abundance: rare (16) and occasional (7). Additionally, we include information about the category (IUCN, CITES, NOM-059-SEMARNAT) as well as uses reported in the literature for the species in the RBBM. The checklist is strictly based on information obtained from deposited herbarium specimens as well as from those collected during fieldwork. We suggest that a conservation plan (in situ and ex situ) for the RBBM is important and necessary. The predominant habit for both families is epiphytic (17 species); even though there are terrestrial (7) and saxicolous (2), and the remaining are facultative species (8). Nine species are included in some risk category. The present work is the most complete and updated list of Bromeliaceae and Orchidaceae for this important natural area in the Mexican State of Hidalgo. However, more fieldwork is needed to document the biodiversity of the area in general and its flora in particular, as a way to highlight the importance of protected areas in preserving biodiversity
