Extreme Red Sea: Life in the deep-sea anoxic brine lakes
Tectonic splitting of the Arabian and African plates originated the Red Sea together
with one of the most unique, remote, and extreme environments on Earth: deep-sea
anoxic brine lakes. They combine multiple extremes namely increased salinity (7-fold),
temperature (up to 70°C), concentration of heavy metals (1,000- to 10,000-fold), and
hydrostatic pressure [1].
Despite such harsh conditions, they harbor an unexpectedly high biodiversity and are
teeming with life. Increased interest in their microbiology led to multiple recent and
ongoing studies. Highlights of this research include: the isolation, physiological characterization
and genome sequencing of unusual new extremophilic microbes; the identification
of several novel phylogenetic lineages; and ongoing cultivation- and molecularbased
assessment of microbial community variation between and within different brines
[2-7].
The uniqueness of these environments offers a high potential for discovery of new microbes,
strategies and biomolecules to cope with extreme conditions, and biotechnological
applications
Carbon nanotubes-reinforced cell-derived matrix-silk fibroin hierarchical scaffolds for bone tissue engineering applications
Dissertação de mestrado em Biofísica e BionanossistemasAs lesões ósseas são o tipo lesões traumáticas mais comuns. O processo regenerativo do osso é um
processo biológico complexo que é influenciado por vários fatores. Atualmente, os autoenxertos representam
o padrão mais elevado da medicina regenerativa óssea, contudo apresentam várias desvantagens, tais como
disponibilidade limitada do tecido dador ou necessidade de intervenção cirúrgica adicional para colheita do
tecido dador. Assim, na área da engenharia de tecidos do osso têm-se procurado desenvolver novos sistemas
artificiais que simultaneamente enderecem esta problemática e consigam mimetizar com maior precisão o
tecido nativo. Neste trabalho, as excelentes propriedades regenerativas da fibroína de seda foram
combinadas com nanotubos de carbono e uma matriz descelularizada derivada de células para a obtenção
de novas matrizes porosas (scaffolds) e hierarquizadas para aplicação em abordagens da engenharia do
tecido ósseo. Estes scaffolds foram fabricados recorrendo ao método de reticulação enzimática, modelagem
através de congelamento e descelularização. A caracterização físico-química revelou que os poros do scaffold
tinham cerca de 112 ± 22 µm e este apresentava uma porosidade total de 75 ± 3%. Para além disso,
observou-se que os scaffolds possuem um E’ a rondar os 5 kPa, e que são bioativos, in vitro. Adicionalmente,
os estudos ex vivo de avaliação do possível comportamento hemolítico revelaram que os scaffolds não
possuem qualquer efeito hemolítico. Por sua vez, os estudos realizados in vitro envolvendo o uso de células
estaminais obtidas da gordura humana (hASCs) mostraram que os scaffolds são capazes de suportar a
proliferação celular. Para além disto, apesar do nível de atividade metabólica exibido pelas células nos
scaffolds desenvolvidos ser semelhante aos scaffolds só de fibroína de seda, foi observada uma atividade da
ALP superior. Por sua vez, a análise histológica mostrou que células foram capazes de migrar para o interior
dos scaffolds e a produzir colagénio. A expressão de vários marcadores osteogénicos, tais como a ALP, OPN,
Runx-2 e Col Iα, também foi observada confirmando assim o potencial osteogénico dos scaffolds
desenvolvidos. Em suma, os scaffolds hierárquicos desenvolvidos neste trabalho mostraram um alto
potencial para uso em abordagens da engenharia de tecido ósseo.Bone injuries are the most common traumatic injuries. The regenerative process of bone healing is
a complex biological process that is influenced by several factors. The current gold standard in bone
regenerative medicine are autografts which present several drawbacks, such as limited supply or donor-site
morbidity. Thus, in the field of bone tissue engineering, the development of new scaffold systems that
addresses the current limitations and more closely mimic the native tissue is being pursued. In this work, the
excellent regenerative properties of silk fibroin were combined with carbon nanotubes and decellularized cell derived extracellular matrix to obtain new scaffolds for bone tissue engineering applications. These easy to
produce scaffolds were fabricated using enzymatic cross-liking, freeze modeling and decellularization
methods. The physicochemical characterization revealed that the developed scaffolds presented pores with
≈ 112 ± 22 µm in size and a total porosity of ≈ 75 ± 3%. Furthermore, scaffolds presented an E’ of ≈ 5 kPa
and were bioactive in vitro. Additionally, ex vivo hemolytic assay evidenced that scaffolds expressed no
hemolytic effect. Regarding biological evaluation, the cellular in vitro studies performed on adipose-derived
stem cells (hASCs) showed that scaffolds supported cell proliferation. Besides, despite the hASCs seeded on
developed scaffolds evidenced similar metabolic activity to standard silk fibroin scaffolds, they presented an
increased ALP activity. Moreover, the histological stainings showed that cells were capable to migrate into the
scaffolds and produce de novo collagen. The expression of several osteogenic markers such as ALP, OPN,
Runx-2 and Col Iα was also verified, further supporting the osteogenic potential of the developed scaffolds.
Overall, the hierarchical scaffolds produced in the present work show great promise for finding applications

Optimizing multi-channel use in digital marketing campaigns
Digital marketing has been gaining importance in the last years. It has become an easier, faster and more accountable way to reach customers than traditional, offline marketing, leading to more effective results and lower campaign costs. In digital marketing, the need to constantly reach large audiences is met by integrating online communication channels, as this increases customer touch points and ad exposures. It becomes thus important to know which channels perform better, individually or in combination with each other and traditional media. This dissertation evaluates the performance of different online advertising channels, including email (old/new formats), display (customary and social media formats) and search engine marketing (SEM), within a single, multi-channel campaign conducted in Portugal in 2014 for a large FMCG brand. First, the effectiveness and efficiency of different channels were statistically compared, and related to international benchmarks. Then, linear regression models were ran to estimate the effect of single and multiple channel use on campaign performance metrics. Results showed that classic email formats performed the best in terms of conversion rates, profit and ROI, whereas social media display ads performed the worst. Meanwhile, the use of SEM and new email formats did not improve campaign performance. It was possible to conclude that pre-defining and aligning campaign goals between agencies and advertisers is the key to optimize multi-channel use in digital marketing, as well as past experience with different channels.mais fácil, mais rápida e mais responsável para atingir os clientes que o marketing offline tradicional, levando a resultados mais eficazes e os custos de campanha inferiores. Em marketing digital, a necessidade de alcançar grandes audiências é cumprida por meio da integração de canais de comunicação on-line, pois isso aumenta os pontos de contacto com o cliente. Torna-se assim importante saber quais os canais que geram melhor desempenho, individualmente ou em combinação. Esta dissertação avalia o desempenho dos diferentes canais de publicidade on-line, incluindo e-mail (velho / novo formatos), display (formatos de media habitual e social) e search engine marketing (SEM), dentro de uma campanha multicanal realizada em Portugal em 2014 para uma grande marca FMCG. Em primeiro lugar, a eficácia e eficiência de diferentes canais foram comparados estatisticamente, e relacionado com benchmarks internacionais. Então, modelos de regressão linear foram feitos para estimar o efeito do uso único e múltiplo de canais, em métricas de desempenho da campanha. Os resultados mostraram que os formatos de e-mail clássicos tiveram o melhor desempenho em termos de taxas de conversão, lucro e ROI, enquanto os anúncios de exibição de media social realizada pelo pior. Enquanto isso, o uso de SEM e novos formatos de e-mail não melhorou o desempenho da campanha. Foi possível concluir que a pré-definição e alinhamento das metas de campanha entre agências e anunciantes é a chave para otimizar o uso de multicanal em marketing digital, bem como a experiência passada com diferentes canais
Atividade física, nutrição, farmacoterapia; a obesidade e seus tratamentos: uma revisão da literatura
The main point of this review is to evaluate strategies and pharmacological methods or not for obesity or overweight treatment. Obesity is now one of the major diseases of modern life. Nutritional transition trends occurring during this century direct to a more westernized diet, which, combined with a decrease in physical activity, converge for a high number of obesity all around the world. This represents an increase in morbidity and mortality associated with obesity, once it is a risk factor for several diseases. As a CNCD (chronic non-communicable disease), the IHME (Institute for Health Metrics and Evaluation) attributes 3.4 million deaths and a prevalence of 37% of the adult population with obesity and overweight from 1980 - 2013. Obesity presents itself not only as a scientific and public health problem, but also as a challenge to the industry involving the development of drugs, modified foods, among others, and even government strategies encouraging regular physical activity and dietary orientation to promote improvement of healthy habits. Thus, the evaluation of resources and strategies to prevent obesity are objects of study of many researchers all around the world
The critique of essentialist critique of cyberculture
O objetivo deste artigo é analisar a perspectiva crítica da cibercultura a partir da discussão sobre a essência da tecnologia. Para tanto, o artigo retoma a discussão clássica sobre a essência da técnica e a atualiza a partir das visões dos novos críticos da cultura digital. O argumento central é que a perspectiva crítica tradicional (fundamentalista ou pessimista) falha ao abordar os fenômenos da cultura digital pelo viés essencialista. É proposta uma análise da cibercultura pela Teoria Ator-Rede (TAR) já que uma visão focada no oligóptico, presa às redes constituintes do fenômeno técnico, fiel às associações que formam o social, pode oferecer uma solução ao fracasso empírico da crítica.The objective of this paper is to analyze the critical perspective of cyberculture from the discussion on the essence of technology. The article revisits the classic discussion about the essence of technology and updates it from the visions of the new critics of digital culture. The central argument is that traditional critical perspective (fundamentalist or pessimistic) fails to address the phenomena of digital culture by essentialist bias. It proposes an analysis of cyberculture by Actor-Network Theory (ART) since a focused view, stucked to the constituent networks of technical phenomenon, and attached to social associations may offer a solution to the empirical failure of criticism
