A FABP4-PPARγ signaling axis regulates human monocyte responses to electrophilic fatty acid nitroalkenes
Nitro-fatty acids (NO2-FA) are electrophilic lipid mediators derived from unsaturated fatty acid nitration. These species are produced endogenously by metabolic and inflammatory reactions and mediate anti-oxidative and anti-inflammatory responses. NO2-FA have been postulated as partial agonists of the Peroxisome Proliferator-Activated Receptor gamma (PPARγ), which is predominantly expressed in adipocytes and myeloid cells. Herein, we explored molecular and cellular events associated with PPARγ activation by NO2-FA in monocytes and macrophages. NO2-FA induced the expression of two PPARγ reporter genes, Fatty Acid Binding Protein 4 (FABP4) and the scavenger receptor CD36, at early stages of monocyte differentiation into macrophages. These responses were inhibited by the specific PPARγ inhibitor GW9662. Attenuated NO2-FA effects on PPARγ signaling were observed once cells were differentiated into macrophages, with a significant but lower FABP4 upregulation, and no induction of CD36. Using in vitro and in silico approaches, we demonstrated that NO2-FA bind to FABP4. Furthermore, the inhibition of monocyte FA binding by FABP4 diminished NO2-FA-induced upregulation of reporter genes that are transcriptionally regulated by PPARγ, Keap1/Nrf2 and HSF1, indicating that FABP4 inhibition mitigates NO2-FA signaling actions. Overall, our results affirm that NO2-FA activate PPARγ in monocytes and upregulate FABP4 expression, thus promoting a positive amplification loop for the downstream signaling actions of this mediator.Fil: Lamas Bervejillo, M.. Universal Quantum Cloning in Cavity QED
We propose an implementation of an universal quantum cloning machine [UQCM,
Hillery and Buzek, Phys. Rev. A {\bf 56}, 3446 (1997)] in a Cavity Quantum
Electrodynamics (CQED) experiment. This UQCM acts on the electronic states of
atoms that interact with the electromagnetic field of a high cavity. We
discuss here the specific case of the cloning process using either a
one- or a two-cavity configuration
Toxicidade de sementes de fedegoso (Cassia occidentalis L.) para frangos de corte
Three experiments were carried out in order to determine toxic levels of Cassia occidentalis seeds added to broiler feed. On the first two experiments 640 one day-old sexed broiler chicks were used. The level of inclusion in starter feed of the first trial were 0; 2; 4 and 6% and the mortality rates obtained were 5.77; 84.62; 100 and 100%, respectively. In the second trial, levels utilized were 0; 0.5; 1 and 2% and the mortality rates were 0; 3.29; 15.73 and 89.47%, respectively. 960 3-day-old sexed chicks were used in the third experiment. In the starter (4 to 31 days of age) and finisher (32 to 52 days) experimental rations the seeds were added at 0; 0.1; 0.2; 0.3; 0.4 and 0.5%. The final body weights were 2.01; 1.95; 1.95; 1.90; 1.77 and 1.58 kg, respectively, being the three highest level groups different from the control. Feed consumption (4.33; 4.32; 4.32; 4.28; 4.08 and 3.80 kg, respectively) and feed conversion (2.15; 2.21; 2.22; 2.25; 2.31 and 2.41, respectively) were significantly different at 0.4 and 0.5% of seed inclusion comparing to the control group. Histologic aspects of birds that were fed with toxic seeds were characterized by a degenerative process found in the heart, liver, pancreas, kidneys, skeletic muscle and intestines. Mortality rates were 2.77; 2.08; 2.08; 0.69; 0.69 and 0%, respectively.Foram realizados três ensaios com o objetivo de se determinar o nível de toxicidade da contaminação de sementes de Cassia occidentalis na alimentação de frangos de corte. Nos dois primeiros ensaios foram utilizadas 640 aves de 1 dia de idade. Os níveis de adição da semente no primeiro ensaio foram, 0; 2; 4 e 6%, obtendo-se 5,77; 84,62; 100 e 100% de mortalidade, respectivamente. No segundo ensaio, os níveis utilizados foram 0; 0,5; 1 e 2%, obtendo-se 0; 3,29; 15,73 e 89,47% de mortalidade, respectivamente. No terceiro ensaio, utilizou-se 960 aves de 3 dias de idade. Adicionou-se a ração inicial (4-31 dias de idade) e final (32-52 dias) sementes moídas de fedegoso aos níveis de 0; 0,1; 0,2; 0,3; 0,4 e 0,5%. O peso médio final das aves foi 2,01; 1,95; 1,95; 1,90; 1,77 e 1,58 kg, respectivamente, observando-se diferença significativa (P < 0,05%) em relação ao controle para 0,3; 0,4 e 0,5% de adição. Os valores médios de consumo (4,33; 4,32; 4,32; 4,28; 4,08 e 3,80 kg, respectivamente) e da conversão alimentar (2,15; 2,21; 2,22; 2,25; 2,31 e 2,41, respectivamente) foram significativamente diferentes (P < 0,05%) para 0,4 e 0,5% de adição das sementes em relação ao grupo controle. O aspecto histológico de órgãos e tecidos das aves que receberam C.occidentalis, sacrificadas no término das fases inicial e final, foi característico de um processo degenerativo observado no coração, fígado, pâncreas, rins, músculo estriado esquelético e intestinos. Models of peer support to remediate post-intensive care syndrome: A report developed by the SCCM Thrive International Peer Support Collaborative
Objective: Patients and caregivers can experience a range of physical, psychological, and
cognitive problems following critical care discharge. The use of peer support has been
proposed as an innovative support mechanism.
Design: We sought to identify technical, safety and procedural aspects of existing
operational models of peer support, among the Society of Critical Care Medicine Thrive Peer
Support Collaborative. We also sought to categorize key distinctions between these models
and elucidate barriers and facilitators to implementation.
Subjects: 17 Thrive sites from the USA, UK, and Australia were represented by a range of
healthcare professionals.
Interventions: Via an iterative process of in-person and email/conference calls, members
of the Collaborative, defined the key areas on which peer support models could be defined
and compared; collected detailed self-reports from all sites; reviewed the information and
identified clusters of models. Barriers and challenges to implementation of peer support
models were also documented.
Results: Within the Thrive Collaborative, six general models of peer support were identified:
Community based, Psychologist-led outpatient, Models based within ICU follow-up clinics,
Online, Groups based within ICU and Peer mentor models. The most common barriers to
implementation were: recruitment to groups, personnel input and training: sustainability
and funding, risk management and measuring success.
Conclusion: A number of different models of peer support are currently being developed
to help patients and families recover and grow in the post-critical care setting
Mid-rapidity anti-proton to proton ratio from Au+Au collisions at GeV
We report results on the ratio of mid-rapidity anti-proton to proton yields
in Au+Au collisions at \rts = 130 GeV per nucleon pair as measured by the
STAR experiment at RHIC. Within the rapidity and transverse momentum range of
and 0.4 1.0 GeV/, the ratio is essentially independent of
either transverse momentum or rapidity, with an average of for minimum bias collisions. Within errors, no
strong centrality dependence is observed. The results indicate that at this
RHIC energy, although the -\pb pair production becomes important at
mid-rapidity, a significant excess of baryons over anti-baryons is still

An experimental DUAL model of advanced liver damage
Individuals exhibiting an intermediate alcohol drinking pattern in conjunction with signs of metabolic risk present clinical features of both alcohol-associated and metabolic-associated fatty liver diseases. However, such combination remains an unexplored area of great interest, given the increasing number of patients affected. In the present study, we aimed to develop a preclinical DUAL (alcohol-associated liver disease plus metabolic-associated fatty liver disease) model in mice. C57BL/6 mice received 10% vol/vol alcohol in sweetened drinking water in combination with a Western diet for 10, 23, and 52 weeks (DUAL model). Animals fed with DUAL diet elicited a significant increase in body mass index accompanied by a pronounced hypertrophy of adipocytes, hypercholesterolemia, and hyperglycemia. Significant liver damage was characterized by elevated plasma alanine aminotransferase and lactate dehydrogenase levels, extensive hepatomegaly, hepatocyte enlargement, ballooning, steatosis, hepatic cell death, and compensatory proliferation. Notably, DUAL animals developed lobular inflammation and advanced hepatic fibrosis. Sequentially, bridging cirrhotic changes were frequently observed after 12 months. Bulk RNA-sequencing analysis indicated that dysregulated molecular pathways in DUAL mice were similar to those of patients with steatohepatitis. Conclusion: Our DUAL model is characterized by obesity, glucose intolerance, liver damage, prominent steatohepatitis and fibrosis, as well as inflammation and fibrosis in white adipose tissue. Altogether, the DUAL model mimics all histological, metabolic, and transcriptomic gene signatures of human advanced steatohepatitis, and therefore serves as a preclinical tool for the development of therapeutic targets
