    The animal drawn ridger-blade, an equipment to be used as an attachment to Wheeled Tool Carrier, has been developed from a ridger of adjustable wings. One of the wing of the ridger body needs to be replaced by a 5 mm metallic sheet of 30 cm x 110 cm. The operational field capacity of the implement varies from 0.16 a 0.47 ha/hr for implantation, reformation or weeding of a new type of in situ rainwater harvesting system. This new system consists of narrow and broad, ridges spaced alternatively on contour. The broad ridges of triangular shape serve for rainwater harvesting, the narrow ridges of conventional shape as planting zone and furrows in between these two as water storing zone. The ratio between water harvesting area to the planting zone area is maintained in the range of 2:1.A lâmina-enleitadora a tração animal é um equipamento para uso como conexo de chassis-porta-implementos, adaptado a um sulcador tipo asas extensas. Este sistema consiste de camalhões largos e estreitos alternados em curva de nível.     Electrospun nanofibrous meshes cultured with Wharton’s Jelly Stem Cell: an alternative for cartilage regeneration, without the need of growth factors

    Many efforts are being directed worldwide to the treatment of OA-focal lesions. The majority of those efforts comprise either the refinement of surgical techniques or combinations of biomaterials with various autologous cells. Herein, we tested electrospun polycaprolactone (PCL) nanofibrous meshes for cartilage tissue engineering. For that, articular chondrocytes (hACs) isolated from human osteoarthritic joints and Whartonâ s Jelly Stem Cells (hWJSCs) are cultured on electrospun nanofiber meshes, without adding external growth factors. We observed higher glycosaminoglycans production and higher overexpression of cartilage-related genes from hWJSCs cultured with basal medium, when compared to hACs isolated from osteoarthritic joints. Moreover, the presence of sulfated proteoglycans and collagen type II is observed on both types of cell cultures. We believe that this effect is due to either the electrospun nanofibers topography or the intrinsic chondrogenic differentiation potential of hWJSCs.     Fitomassa de adubos verdes e controle de plantas daninhas em diferentes densidades populacionias de leguminosas.

    O objetivo deste trabalho foi avaliar a fitomassa de calopogônio, mucuna-preta, mucunarajada,feijão-de-porco, guandu de porte alto, Crotalaria spectabilis e C. breviflora sob diferentes densidades de semeadura (10, 20, 40, 80 e 160 sementes viáveis m-2), e o crescimento de plantas daninhas nessas densidades, em área de tabuleiros costeiros. O experimento foi desenvolvido de maio a agosto de 1996, no Campo Experimental “Antônio Martins”(EMDAGRO/Embrapa-CPATC), em Lagarto, SE. O número de plantas vivas na floração (NPVF) e a matéria seca da parte aérea das leguminosas (MSPA) foram determinados quando, em cada espécie, cerca de 50% das plantas floresceram. Maiores incrementos de MSPA, em resposta ao adensamento populacional, foram observados em C. spectabilis e C. breviflora, seguidas pelo calopogônio, mucuna-preta e mucuna-rajada. Em relação ao feijão-de-porco, a resposta foi negativa, enquanto com o guandu não houve influência. Quanto ao NPVF, as respostas ao adensamento foram lineares e positivas em C. spectabilis, C. breviflora e calopogônio, e quadráticas com ponto de máxima em feijão-de-porco,guandu e mucuna-rajada. Embora nenhum modelo tenha sido ajustado para expressar a relação entre NPVF e adensamento na semeadura de mucuna-preta, a sobrevivência dessa espécie foi reduzida em todas as densidades.     Photosynthesis by six portuguese maize cultivars during drought stress and recovery

    Photosynthesis, chlorophyll fluorescence and leaf water parameters were measured in six Portuguese maize (Zea mays L.) cultivars during and following a period of drought stress. The leaf relative water content (RWC) responded differently among cultivars but, except for cultivar PB369, recovered close to initial values after watering was restored. Photosynthetic rate and stomatal conductance decreased with drought but more slowly in cultivars PB269 and PB260 than in cultivars AD3R, PB64, PB304 and PB369. Water use efficiency (WUE) decreased during the water stress treatment although with cultivar PB260 the decrease was marked only when the RWC fell below 40%. Recovery of WUE was seen with all cultivars except PB369. The maximum quantum efficiency of photosystem II, the photochemical quenching coefficient, the electron transport rate in PSII and the estimated functional plastoquinone pool tended to decrease with drought, while the non -photochemical quenching coefficient increased. The parameters estimated from chlorophyll fluorescence did not recover in PB369, during re - watering. The results show that PB260 and PB269 were the most tolerant and PB369 was the least tolerant cultivars to water stress. The variation found amongst the cultivars tested suggests the existence of valuable genetic resources for crop improvement in relation to drought tolerance

    An issue of concern: unique truncated ORF8 protein variants of SARS-CoV-2

    Open reading frame 8 (ORF8) shows one of the highest levels of variability among accessory proteins in Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), the causative agent of Coronavirus Disease 2019 (COVID-19). It was previously reported that the ORF8 protein inhibits the presentation of viral antigens by the major histocompatibility complex class I (MHC-I), which interacts with host factors involved in pulmonary inflammation. The ORF8 protein assists SARS-CoV-2 in evading immunity and plays a role in SARS-CoV-2 replication. Among many contributing mutations, Q27STOP, a mutation in the ORF8 protein, defines the B.1.1.7 lineage of SARS-CoV-2, engendering the second wave of COVID-19. In the present study, 47 unique truncated ORF8 proteins (T-ORF8) with the Q27STOP mutations were identified among 49,055 available B.1.1.7 SARS-CoV-2 sequences. The results show that only one of the 47 T-ORF8 variants spread to over 57 geo-locations in North America, and other continents, which include Africa, Asia, Europe and South America. Based on various quantitative features, such as amino acid homology, polar/non-polar sequence homology, Shannon entropy conservation, and other physicochemical properties of all specific 47 T-ORF8 protein variants, nine possible T-ORF8 unique variants were defined. The question as to whether T-ORF8 variants function similarly to the wild type ORF8 is yet to be investigated. A positive response to the question could exacerbate future COVID-19 waves, necessitating severe containment measures

    Agricultural Expansion in the Brazilian Cerrado: Increased Soil and Nutrient Losses and Decreased Agricultural Productivity

    While food and nutrition security are issues that national and international organizations are tackling, one of the central problems often overlooked is the essential role of soils in providing nutritious food. Soils are the base for food production and food security. However, the majority of soils are in fair and poor conditions, with the most significant threats being erosion and loss of nutrients. In this study, we estimate the potential of soil loss, agricultural productivity loss, and nutrient loss for Brazil’s most important agricultural region, the Brazilian Cerrado, for the years 2000 and 2012. For this, we applied the Revised Universal Soil Loss Equation (RUSLE) model integrated with a geographical information system (GIS) to estimate annual soil loss rate and agricultural productivity loss, and used total nitrogen and total phosphorus in soil to estimate the annual nutrient loss rate caused by soil loss. All model factors and data were obtained from the literature. The results show that agricultural expansion in the Brazilian Cerrado is increasing the area of severe erosion, occasioning agricultural productivity decrease and soil nutrient depletion. The annual soil loss rate increased from 10.4 (2000) to 12.0 Mg ha−1 yr−1 (2012). Agricultural productivity loss occurred in more than 3 million hectares of crops and silviculture in 2000 and in more than 5.5 million hectares in 2012. Severely eroded areas lost between 13.1 and 25.9 times more nutrients than areas with low and moderate soil loss rates. These findings show that government policy should be directed to ensure the sustainable use of soils, mainly in agriculturally consolidated regions of the Brazilian Cerrado

    Monoclonal antibody induced with inactived EV71-Hn2 virus protects mice against lethal EV71-Hn2 virus infection

    <p>Abstract</p> <p>Background</p> <p>Enterovirus 71 (EV71) is a viral pathogen that belongs to the <it>Picornaviridae </it>family, EV71-infected children can develop severe neurological complications leading to rapid clinical deterioration and death.</p> <p>Results</p> <p>In this study, several monoclonal antibodies (MAbs) were produced by immunizing mice with the inactived EV71 Henan (Hn2) virus strain. The isolated MAbs were characterised by <it>in vitro </it>neutralizing analysis and peptide ELISA. ELISA assay showed that the neutralizing monoclonal antibody 4E8 specifically reacted with synthetic peptides which contain amino acid 240-250 and 250-260 of EV71 VP1. The <it>in vivo </it>protection assay showed that 4E8 can protect two-day-old BALB/c mice against the lethal challenge of EV71 virus.</p> <p>Conclusion</p> <p>The MAb 4E8 could be a promising candidate to be humanized and used for treatment of EV71 infection.</p
