Probiotics based on bacteria of the genus Bacillus with a multifactorial mechanism of action are considered as a possible alternative to antibiotics in the treatment of purulent wounds. The aim of the study was to determine the antimicrobial and therapeutic effect of the Arederma probiotic preparation containing probiotic strains of the genus Bacillus in an experimental model of a purulent wound in animals. The antimicrobial efficacy of the probiotic against test strains and clinical isolates of pathogenic and opportunistic microorganisms was studied using the method of delayed antagonism. Staphylococcus aureus ATCC 6538 and Streptococcus pyogenes K-7 were used to model a purulent wound. From the surface of the wounds, bacteria of Staphylococcus, Streptococcus, Enterobacteriaceae, Pseudomonas genera and Enterobacteriaceae family were sown on appropriate selective media for the cultivation and enumeration of different groups of microorganisms by generally accepted microbiological research methods. The formation of a purulent wound in rabbits caused by mechanical skin damage and subsequent double infection with Staphylococcus aureus ATCC 6538 and Streptococcus pyogenes K-7 strains was accompanied with a pronounced inflammatory process, necrosis, the formation of purulent exudate and general intoxication. Representatives of the genera Staphylococcus and Streptococcus, microscopic fungi and, to a lesser extent, members of the family Enterobacteriaceae and Pseudomonas were found on the surface of purulent wounds, which confirmed the development of the infectious-inflammatory process. Treatment of purulent wounds with a suspension of probiotic preparation once a day for 4 days led to their faster healing (gradual attenuation of the inflammatory process, reduction of edema and discharge, as well as their disappearance) compared with untreated purulent wounds (control). Representatives of the Staphylococcus and Streptococcus genera, as well as microscopic fungi, presented in purulent wounds treated with probiotic preparation in much smaller numbers than in the control, and bacteria of the Pseudomonas genus and the Enterobacteriaceae family were not detected at all. The effective antimicrobial effect of this probiotic preparation against opportunistic and pathogenic microorganisms was confirmed by in vitro studies. Therefore, the Arederma probiotic preparation showed an effective therapeutic and antimicrobial effect in the experimental model of a purulent wound in animals, so it can be recommended for further preclinical and clinical studies

BACKGROUND: Diabetic foot ulcer (DFU) is a dangerous complication of diabetes mellitus (DM), which can lead to the development of chronic wounds and amputations. Recombinant human epidermal growth factor (rhEGF) can be used as an adjuvant treatment for chronic wounds resistant to standard treatment. Studies have demonstrated its clinical efficacy, however, there is insufficient information on the long-term results of treatment, its safety and the effect on the progression of diabetes complications, adverse cardiovascular events and the development of cancer.AIM: To assess the long-term results of rhEGF therapy for trophic foot ulcers in individuals with multiple complications of diabetes.METHODS: The study included 20 patients with type 1 and type 2 diabetes and various forms of DFS without critical ischemia, who had previously been treated with DFS using rhEGF in order to assess the general condition, progression of microand macrovascular complications of diabetes, adverse cardiovascular events, the development of cancer and the quality of life.RESULTS: There was a statistically significant difference between the area of wound defects, the percentage of granulation tissue that filled the wound defect, before the start of rhEGF treatment and at the time of discharge from the hospital (p <0.05). During treatment with rhEGF, mild adverse events were observed in 35%. Complete epithelialization of wounds in most patients occurred in 3 [2; 4] months. In 11.76%, the wound was not completely epithelialized. Relapse occurred in 5.8% due to non-compliance with limb unloading. Minor amputation was performed in 1 patient. Progression of diabetic retinoand nephropathy was revealed in 23.5%. 11.76% suffered myocardial infarction of unknown age, 1 patient (5.88%) suffered acute cerebrovascular accident. Serious adverse events in the form of PE with a fatal outcome and critical ischemia of the lower limb were recorded in 5.8%.CONCLUSIONS: As a result of the study of long-term results of rhEGF treatment of chronic foot wounds, a low percentage of relapses and small amputations, the absence of high amputations and oncological diseases, the development of serious adverse events in 2 patients, the progression of diabetic retinoand nephropathy in 4 patients, the development of IM of unknown age in 2 patients was recorded. and stroke in 1 patient after rhEGF therapy

6^6Li-loaded liquid scintillators produced by direct dissolution of compounds in diisopropylnaphthalene (DIPN)

The paper describes preparation of 6^6Li-loaded liquid scintillators by methods involving direct dissolution of 6^6Li salts in the commercial diisopropylnaphthalene (DIPN) solvent, without the formation of water-in-oil emulsions. Methods include incorporation of 6^6Li that, unlike previously reported formulations, does not require additions of water or a strong acid such as hydrochloric acid (HCl). Results of the conducted experiments show that dissolution of aromatic and aliphatic 6^6Li salts in DIPN can be easily achieved at 0.1- 0.3% by weight of atomic 6^6Li, using small additions of waterless surfactants, or mild carboxylic acids. An alternative way suggests incorporation of 6^6Li as a part of a surfactant molecule that can be dissolved in DIPN without any solubilizing additions. Proposed methods enable preparation of efficient 6^6Li-loaded liquid scintillators that, at a large scale of 50 cm, exhibit good pulse shape discrimination (PSD) properties combined with up to 107% of light output and up to 115% of the attenuation length measured relative to standard undoped EJ-309 liquid scintillator.Comment: Submitted to Nuclear Instruments and Methods in Physics Research

NRF2-driven miR-125B1 and miR-29B1 transcriptional regulation controls a novel anti-apoptotic miRNA regulatory network for AML survival

    Get PDF
Transcription factor NRF2 is an important regulator of oxidative stress. It is involved in cancer progression, and has abnormal constitutive expression in acute myeloid leukaemia (AML). Posttranscriptional regulation by microRNAs (miRNAs) can affect the malignant phenotype of AML cells. In this study, we identified and characterised NRF2-regulated miRNAs in AML. An miRNA array identified miRNA expression level changes in response to NRF2 knockdown in AML cells. Further analysis of miRNAs concomitantly regulated by knockdown of the NRF2 inhibitor KEAP1 revealed the major candidate NRF2-mediated miRNAs in AML. We identified miR-125B to be upregulated and miR-29B to be downregulated by NRF2 in AML. Subsequent bioinformatic analysis identified putative NRF2 binding sites upstream of the miR-125B1 coding region and downstream of the mir-29B1 coding region. Chromatin immunoprecipitation analyses showed that NRF2 binds to these antioxidant response elements (AREs) located in the 5′ untranslated regions of miR-125B and miR-29B. Finally, primary AML samples transfected with anti-miR-125B antagomiR or miR-29B mimic showed increased cell death responsiveness either alone or co-treated with standard AML chemotherapy. In summary, we find that NRF2 regulation of miR-125B and miR-29B acts to promote leukaemic cell survival, and their manipulation enhances AML responsiveness towards cytotoxic chemotherapeutics

    Get PDF
Objective: to study the impact of pain intensity on the progression of knee osteoarthrosis (OA). Subjects and methods. One hundred and ten patients with knee OA were examined at a 5-year interval. All the patients underwent a questionnaire survey and knee joint pain assessment using a visual analog scale (VAS) and standard radiography. Results. After 5-year follow-up, radiographic OA progression was seen in 40 patients (Group 2); its stage remained the same in 70 patients (Group 1). In both groups, the patients were matched for age (59.2+9.5 and 59.0+8.1 years) and disease duration (11.1+10.6 and 13.7+9.9 years). During the first examination, pain on walking was more severe in Group 1 than in Group 2: 57.8+16.6 and 48.7+13.3 mm by VAS (р=0.002), as well as severe joint pain was predominant in these patients: 22.5 and 11.4%, respectively. Over the 5-year period, there was an increase in pain intensity. At the end of the follow-up, the patients with progressive OA rated their knee joint pain as severe in 35% of cases whereas in this index the non-progression group was only 12.9 (p = 0.012). Conclusion. In the OA progression group, pain intensity was initially statistically higher than that in the non-progression group. During 5-year follow-up, Group 1 showed an increase in knee joint pain intensity on walking, which can be considered as one of the predictors of gonarthrosis progression
