    Exocytosis of catecholamine (CA)-containing and CA-free granules in chromaffin cells.

    Recent evidence suggests that endocytosis in neuroendocrine cells and neurons can be tightly coupled to exocytosis, allowing rapid retrieval from the plasma membrane of fused vesicles for future use. This can be a much faster mechanism for membrane recycling than classical clathrin-mediated endocytosis. During a fast exo-endocytotic cycle, the vesicle membrane does not fully collapse into the plasma membrane; nevertheless, it releases the vesicular contents through the fusion pore. Once the vesicle is depleted of transmitter, its membrane is recovered without renouncing its identity. In this report, we show that chromaffin cells contain catecholamine-free granules that retain their ability to fuse with the plasma membrane. These catecholamine-free granules represent 7% of the total population of fused vesicles, but they contributed to 47% of the fusion events when the cells were treated with reserpine for several hours. We propose that rat chromaffin granules that transiently fuse with the plasma membrane preserve their exocytotic machinery, allowing another round of exocytosis

    Construction of a Cantilever-Andreev-Tunneling rig and its applications to superconductors

    A technique for point-contact spectroscopy, based on an electro-mechanical mechanism for the contact formation, has been developed. It is designed to be used in both 4^4He and 3^3He cryostats. The performance has been demonstrated by conductance measurements on various kinds of superconductors, including the conventional superconductor Nb, the two-band superconductor MgB2_2, and the heavy-fermion superconductor CeCoIn5_5. Characteristic conductance spectra obtained prove this technique is useful for the investigation of the superconducting order parameter.     Phosphatidylinositol phosphate kinase type Iγ regulates dynamics of large dense-core vesicle fusion.

    Phosphatidylinositol-4,5-bisphosphate was proposed to be an important regulator of large dense-core vesicle exocytosis from neuroendocrine tissues. Here, we have examined the kinetics of secretion in chromaffin cells from mice lacking phosphatidylinositol phosphate kinase type Iγ, the major neuronal phosphatidylinositol-4-phosphate 5-kinase. Absence of this enzyme caused a reduction of the readily releasable vesicle pool and its refilling rate, with a small increase in morphologically docked vesicles, indicating a defect in vesicle priming. Furthermore, amperometry revealed a delay in fusion pore expansion. These results provide direct genetic evidence for a key role of phosphatidylinositol-4,5-bisphosphate synthesis in the regulation of large dense-core vesicle fusion dynamics

    Health literacy, health status, and healthcare utilization of Taiwanese adults: results from a national survey

    Abstract Background Low health literacy is considered a worldwide health threat. The purpose of this study is to assess the prevalence and socio-demographic covariates of low health literacy in Taiwanese adults and to investigate the relationships between health literacy and health status and health care utilization. Methods A national survey of 1493 adults was conducted in 2008. Health literacy was measured using the Mandarin Health Literacy Scale. Health status was measured based on self-rated physical and mental health. Health care utilization was measured based on self-reported outpatient clinic visits, emergency room visits, and hospitalizations. Results Approximately thirty percent of adults were found to have low (inadequate or marginal) health literacy. They tended to be older, have fewer years of schooling, lower household income, and reside in less populated areas. Inadequate health literacy was associated with poorer mental health (OR, 0.57; 95% CI, 0.35-0.91). No association was found between health literacy and health care utilization even after adjusting for other covariates. Conclusions Low (inadequate and marginal) health literacy is prevalent in Taiwan. High prevalence of low health literacy is not necessarily indicative of the need for interventions.     Microstructure characterization of ODS-RAFM steels

    Results of the microstructural characterization of four different RAFM ODS Eurofer 97 batches are presented and discussed. Analyses and observations were performed by nuclear microprobe and scanning and transmission electron microscopy. X-ray elemental distribution maps obtained with proton beam scans showed homogeneous composition within the proton beam spatial resolution and, in particular, pointed to a uniform distribution of ODS (yttria) nanoparticles in the Eurofer 97 matrix. This was confirmed by transmission electron microscopy. Scanning electron microscopy coupled with energy dispersive spectroscopy made evident the presence of chromium carbide precipitation. Precipitates occurred preferentially along grain boundaries (GB) in three of the batches and presented a discrete distribution in the other, as a result of different thermo-mechanical routes. Additional electron backscattered diffraction experiments revealed the crystalline textures in the ferritic polycrystalline structure of the ODS steel samples
