Effect of D222G Mutation in the Hemagglutinin Protein on Receptor Binding, Pathogenesis and Transmissibility of the 2009 Pandemic H1N1 Influenza Virus
Influenza viruses isolated during the 2009 H1N1 pandemic generally lack known molecular determinants of virulence associated with previous pandemic and highly pathogenic avian influenza viruses. The frequency of the amino acid substitution D222G in the hemagglutinin (HA) of 2009 H1N1 viruses isolated from severe but not mild human cases represents the first molecular marker associated with enhanced disease. To assess the relative contribution of this substitution in virus pathogenesis, transmission, and tropism, we introduced D222G by reverse genetics in the wild-type HA of the 2009 H1N1 virus, A/California/04/09 (CA/04). A dose-dependent glycan array analysis with the D222G virus showed a modest reduction in the binding avidity to human-like (α2-6 sialylated glycan) receptors and an increase in the binding to avian-like (α2-3 sialylated glycan) receptors in comparison with wild-type virus. In the ferret pathogenesis model, the D222G mutant virus was found to be similar to wild-type CA/04 virus with respect to lethargy, weight loss and replication efficiency in the upper and lower respiratory tract. Moreover, based on viral detection, the respiratory droplet transmission properties of these two viruses were found to be similar. The D222G virus failed to productively infect mice inoculated by the ocular route, but exhibited greater viral replication and weight loss than wild-type CA/04 virus in mice inoculated by the intranasal route. In a more relevant human cell model, D222G virus replicated with delayed kinetics compared with wild-type virus but to higher titer in human bronchial epithelial cells. These findings suggest that although the D222G mutation does not influence virus transmission, it may be considered a molecular marker for enhanced replication in certain cell types.Centers for Disease Control and Prevention (U.S.)United States. Faking like a woman? Towards an interpretative theorization of sexual pleasure.
This article explores the possibility of developing a feminist approach to gendered and sexual embodiment which is rooted in the pragmatist/interactionist tradition derived from G.H. Mead, but which in turn develops this perspective by inflecting it through more recent feminist thinking. In so doing we seek to rebalance some of the rather abstract work on gender and embodiment by focusing on an instance of 'heterosexual' everyday/night life - the production of the female orgasm. Through engaging with feminist and interactionist work, we develop an approach to embodied sexual pleasure that emphasizes the sociality of sexual practices and of reflexive sexual selves. We argue that sexual practices and experiences must be understood in social context, taking account of the situatedness of sex as well as wider socio-cultural processes the production of sexual desire and sexual pleasure (or their non-production) always entails interpretive, interactional processes
Determinants of Glycan Receptor Specificity of H2N2 Influenza A Virus Hemagglutinin
The H2N2 subtype of influenza A virus was responsible for the Asian pandemic of 1957-58. However, unlike other subtypes that have caused pandemics such as H1N1 and H3N2, which continue to circulate among humans, H2N2 stopped circulating in the human population in 1968. Strains of H2 subtype still continue to circulate in birds and occasionally pigs and could be reintroduced into the human population through antigenic drift or shift. Such an event is a potential global health concern because of the waning population immunity to H2 hemagglutinin (HA). The first step in such a cross-species transmission and human adaptation of influenza A virus is the ability for its surface glycoprotein HA to bind to glycan receptors expressed in the human upper respiratory epithelia. Recent structural and biochemical studies have focused on understanding the glycan receptor binding specificity of the 1957-58 pandemic H2N2 HA. However, there has been considerable HA sequence divergence in the recent avian-adapted H2 strains from the pandemic H2N2 strain. Using a combination of structural modeling, quantitative glycan binding and human respiratory tissue binding methods, we systematically identify mutations in the HA from a recent avian-adapted H2N2 strain (A/Chicken/PA/2004) that make its quantitative glycan receptor binding affinity (defined using an apparent binding constant) comparable to that of a prototypic pandemic H2N2 (A/Albany/6/58) HA.National Institute of General Medical Sciences (U.S.) (GM57073)National Institute of General Medical Sciences (U.S.) (U54 GM62116)Singapore. Exploring the cultural dimensions of environmental victimization
It has become increasingly clear in recent years that our understanding of ‘victimisation’ is informed by a whole range of societal and political factors which extend well beyond whatever particular form of words appears in any given directive, code or legislative instrument concerning crime, crime victims or criminal justice systems. In this paper, I will seek for the first time to apply recent developments in our understanding of so-called 'cultural victimology' to the issue of environmental harm and its impact on human and non-human animals. McCGarry and Waklate (2015) characterise cultural victimology as broadly comprising of two key aspects. These are the wider sharing and reflection of individual and collective victimisation experiences on the one hand and, on the other, the mapping of those experiences through the criminal justice process. In this discussion I will examine how environmental victimisation is viewed by and presented to society at large and will argue that such representations often fail, as a form of testimony, to adequately convey the traumas involved. Nor is this achieved through the application of present models of criminal, civil or administrative justice regimes in many jurisdictions. This lack of cultural acknowledgement of the harms vested on environmental victims, it is argued, afford us a clearer understand of the continued reticence amongst lawmakers, politicians and legal practitioners to adequately address the impacts of such victimisation through effective justice or regulatory mechanisms. This is unfortunate given that the often collective nature of environmental victimisation makes this particularly suited to a more cultural analysis and understanding. It is argued that various forms of environmental mediation processes might hold the key to this cultural reticence to accept environmental harm as a 'real' and pressing problem as compared to other criminal and civil justice concerns
Influence of heat input and powder density on energetic efficiency of high power diode laser cladding of carbon steel with AISI316L powder
High power diode laser with coaxial powder injection was used to deposit single tracks of austenitic stainlesssteel on to a carbon steel plate, and the influence of heat input and powder density on energetic efficiencyof the process, as well as on some geometrical features of the clad and on dilution was investigated. Theenergetic efficiency, calculated as the energy used to form the clad and the Heat Affected Zone, tends toincrease with the powder density, while it decreases on increasing heat input. The combination of a highpowder density and a low heat input is expected to optimize the energetic efficiency. In these conditions, thechemical dilution of the clad is minimized whilst aspect ratio is rather low but still acceptable. Moreover, theenergy spent for the powder and the substrate can be correlated to the iron contamination of the clad
EFFETTO DI INCLUSIONI METALLICHE NEI PROCESSI DI ANODIZZAZIONE DI LEGHE DI ALLUMINIO
La necessità di lavorare mediante asportazione di truciolo leghe di alluminio ha da tempo portato allo sviluppo di leghe specifiche, far cui le più diffuse contengono piombo. Tali leghe devono ora essere superate a causa delle restrizioni legate alla presenza del piombo, motivate principalmente da ragioni ambientali. Diverse leghe sono state sviluppate cercando di sostituire il piombo con il manganese, il ferro, il nichel o l’indio. Un altro promettente sostituto del piombo per queste applicazioni è il berillio, oggetto di studio negli ultimi anni come alligante dell’alluminio. Molto spesso per applicazioni che spaziano dalla motoristica alla attrezzatura sportiva alla componentistica per l’elettronica, i componenti in lega di alluminio devono essere modificati superficialmente per migliorarne sia la resistenza alla corrosione che le proprietà tribologiche. Uno dei trattamenti più diffusi è l’anodizzazione. La presenza però di una particolare microstruttura, contenente isole di metallo elettrochimicamente più nobile (Pb o Bi) può notevolmente influenzare la qualità dell’ossido anodico e di conseguenza le proprietà di resistenza alla corrosione del componente anodizzato. Tale aspetto non è stato ancora sufficientemente analizzato scientificamente. In questo studio sono stati considerati campioni di diverse leghe di alluminio contenenti sia Pb che Bi, sottoposti a trattamenti di anodizzazione. Sono state studiate le difettosità riscontrate nell’ossido individuate mediante microscopia elettronica ed ottica, collegandole al comportamento corrosivo e alla microstruttura del materiale (dimensioni e distribuzione delle isole di Pb o di Bi). Sono state eseguite prove elettrochimiche di vario tipo al fine di comprendere il meccanismo di degrado. Lo scopo finale del lavoro sperimentale è quello di fornire informazioni utili all’ottimizzazione di nuove leghe di alluminio, lavorabili per asportazione di truciolo esenti piombo, che mantengano buone proprietà protettive dell’ossido anodico
The interactions of disability and impairment
Theoretical work on disability is going through an expansive period, built on the growing recognition of disability studies as a discipline and out of the political and analytical push to bring disability into a prominent position within accounts of the intersecting social categories that shape people's lives. A current debate within critical disability studies is whether that study should include impairment and embodiment within its focus. This article argues it should and does so by drawing from symbolic interactionism and embodiment literatures in order to explore how differences in what bodies can do-defined as impairments-come to play a role in how people make sense of themselves through social interaction. We argue that these everyday interactions and the stories we tell within them and about them are important spaces and narratives through which impairment and disability are produced. Interactions and stories are significant both in how they are shaped by wider social norms, collective stories and institutional processes, and also how they at times can provide points of resistance and challenges to such norms, stories and institutions. Therefore, the significance of impairment and interaction is the role they play in both informing self-identity and also broader dynamics of power and inequality
Emerging role of tumor-associated macrophages as therapeutic targets in patients with metastatic renal cell carcinoma.
Tumor-associated macrophages (TAMs) derived from peripheral blood monocytes recruited into the renal cell carcinoma (RCC) microenvironment. In response to inflammatory stimuli, macrophages undergo M1 (classical) or M2 (alternative) activation. M1 cells produce high levels of inflammatory cytokines, such as tumor necrosis factor-α, interleukin (IL)-12, IL-23 and IL-6, while M2 cells produce anti-inflammatory cytokines, such as IL-10, thus contributing to RCC-related immune dysfunction. The presence of extensive TAM infiltration in RCC microenvironment contributes to cancer progression and metastasis by stimulating angiogenesis, tumor growth, and cellular migration and invasion. Moreover, TAMs are involved in epithelial-mesenchymal transition of RCC cancer cells and in the development of tumor resistance to targeted agents. Interestingly, macrophage autophagy seems to play an important role in RCC. Based on this scenario, TAMs represent a promising and effective target for cancer therapy in RCC. Several strategies have been proposed to suppress TAM recruitment, to deplete their number, to switch M2 TAMs into antitumor M1 phenotype and to inhibit TAM-associated molecules. In this review, we summarize current data on the essential role of TAMs in RCC angiogenesis, invasion, impaired anti-tumor immune response and development of drug resistance, thus describing the emerging TAM-centered therapies for RCC patients
Intermittent docetaxel chemotherapy as first-line treatment for metastatic castration-resistant prostate cancer patients
Aims: The intermittent administration of chemotherapy is a means of preserving patients' quality of life (QL). The aim of this study was to verify whether the intermittent administration of docetaxel (DOC) improves the patients' QL. Patients & methods: All patients received DOC 70 mg/m every 3 weeks for eight cycles. The patients were randomized to receive DOC continuously or with a fixed 3-month interval after the first four DOC courses. Results: The study involved 148 patients. There was no difference in QL between the groups receiving intermittent or continuous treatment. Intermittence had no detrimental effects on disease control. Conclusion: Although feasible and not detrimental, our results showed that true intermittent chemotherapy in metastatic castration-resistant prostate cancer patients failed to improve the patients' QL

The burials at the Neolithic cemetery Kadruka 23 in Sudan have yielded adornments and bone and lithic artefacts that occur as distinct stages of the chaîne opératoire. This article reports on a hitherto unrecognised funerary practice that highlights the importance of craftsmanship for Neolithic communities in life and beyond. Copyright © The Author(s), 2021.
