Potato R1 resistance gene confers resistance against Phytophthora infestans in transgenic tomato plants
Tomato is challenged by several pathogens which cause loss of production. One such pathogen is the oomycete Phytophthora infestans which is able to attack all the aerial parts of the plant. Although a wide range of resistance sources are available, genetic control of this disease is not yet successful. Pyramiding R-genes through genetic transformation could be a straightforward way to produce tomato and potato lines carrying durable resistance to P. infestans. In this work the R1 potato gene was transferred into tomato lines. The tomato transgenic lines were analyzed by using q-RT-PCR and progeny segregation to determine the gene copy number. To test the hypothesis that R1 represents a specifically regulated R-gene, transgenic tomato plants were inoculated with P. infestans isolate 88133 and IPO. All the plants containing the R1 gene were resistant to the late blight isolate IPO-0 and susceptible to isolate 88133. These results provide evidence for specific activation of the R1 gene during pathogen challenge. Furthermore, evidence for enhancement of PR-1 gene expression during P. infestans resistance response was obtained
Resistance to frost and tuber soft rot in near-pentaploid Solanum tuberosum - S. commersonii hybrids
The objectives of the present study were to evaluate the tolerance to low temperatures and tuber soft rot in sexual near-pentaploid hybrids between incongruent 2x (1EBN) Solanum commersonii (CMM) and 4x (4EBN) S. tuberosum (TBR). For freezing resistance, killing temperatures both under non-acclimated and un- der acclimated conditions were determined using the ion leakage procedure. Values for the hybrids were dis- tributed between the wild and cultivated parental values. Some hybrids displayed an acclimation capacity close to 2.5°C, typical of hardy species. Artificial inoculation of tubers with Pectobacterium carotovorum ssp. carotovorum (formerly Erwinia carotovora ssp. carotovora) provided evidence of variability in disease response. Highly resistant hybrids were identified. After conventional phenotypic selection, wild genome content was estimated based on the presence of CMM-specific AFLP fragments. Seven primer combinations were used (Eco-AGG/Mse-CAA; Eco-ACC/Mse-CAT; Eco-ACT/Mse-CAC; Eco-ACT/Mse-CAG; Eco- ACT/Mse-CAA; Eco-ACT/Mse-CAT; Eco-AGG/Mse-CAG). The percentages of CMM-specific AFLPs ranged from 4.3% to 56.7%, with an average value of 28.1%. AFLP analysis was employed for the selection of the hybrids to be used for further breeding objectives
Resistance to Ralstonia Solanacearum of sexual hybrids between Solanum commersonii and S. tuberosum
This research was carried out to study the levels of bacterial wilt resistance and genetic diversity of (near) pentaploid sexual hybrids between S. commersonii (2n = 2x = 24, 1EBN) and cultivated S. tuberosum. Following artificial inoculations with Ralstonia solanacearum, wilting degree was estimated on a scale from 0 to 4, and seven genotypes of 26 (27%) displaying a S. commersonii like behavior were identified. Latent bacterial colonizations were detected in roots of symptomless S. commersonii and hybrids, whereas no bacterial populations were detected within stems. This suggests that the movement and/or growth of the bacterium in the aerial part were strongly inhibited. A molecular study with AFLP markers clustered hybrids into nine groups and provided evidence that resistant hybrids were slightly more similar to cultivated S. tuberosum than to the wild parent. This is important in view of the re-establishment of the cultivated genetic background through backcrosses. Hybrids displayed good fertility and are being used for further breeding efforts
High-Throughput Genomics Enhances Tomato Breeding Efficiency
Tomato (Solanum lycopersicum) is considered a model plant species for a group of economically important crops, such as potato, pepper, eggplant, since it exhibits a reduced genomic size (950 Mb), a short generation time, and routine transformation technologies. Moreover, it shares with the other Solanaceous plants the same haploid chromosome number and a high level of conserved genomic organization. Finally, many genomic and genetic resources are actually available for tomato, and the sequencing of its genome is in progress. These features make tomato an ideal species for theoretical studies and practical applications in the genomics field. The present review describes how structural genomics assist the selection of new varieties resistant to pathogens that cause damage to this crop. Many molecular markers highly linked to resistance genes and cloned resistance genes are available and could be used for a high-throughput screening of multiresistant varieties. Moreover, a new genomics-assisted breeding approach for improving fruit quality is presented and discussed. It relies on the identification of genetic mechanisms controlling the trait of interest through functional genomics tools. Following this approach, polymorphisms in major gene sequences responsible for variability in the expression of the trait under study are then exploited for tracking simultaneously favourable allele combinations in breeding programs using high-throughput genomic technologies. This aims at pyramiding in the genetic background of commercial cultivars alleles that increase their performances. In conclusion, tomato breeding strategies supported by advanced technologies are expected to target increased productivity and lower costs of improved genotypes even for complex traits
Color Mutations Alter the Biochemical Composition in the San Marzano Tomato Fruit
San Marzano (SM) is a traditional Italian landrace characterized by red elongated fruits,
originating in the province of Naples (Italy) and cultivated worldwide. Three mutations, yellow
flesh (r), green flesh (gf) and colorless fruit epidermis (y) were introduced into SM by backcross and
the resulting introgression lines (ILs) produced the expected yellow, brown and pink fruit variants.
In addition, ILs carrying double combinations of those mutations were obtained. The six ILs plus the
SM reference were analyzed for volatile (VOC), non-polar (NP) and polar (P) metabolites. Sixty-eight
VOCs were identified, and several differences evidenced in the ILs; overall gf showed epistasis
over r and y and r over y. Analysis of the NP component identified 54 metabolites; variation in
early carotenoids (up to lycopene) and chlorophylls characterized respectively the ILs containing r
and gf. In addition, compounds belonging to the quinone and xanthophyll classes were present in
genotypes carrying the r mutation at levels higher than SM. Finally, the analysis of 129 P metabolites
evidenced different levels of vitamins, amino acids, lipids and phenylpropanoids in the ILs. A
correlation network approach was used to investigate metabolite–metabolite relationships in the
mutant lines. Altogether these differences potentially modified the hedonistic and nutritional value
of the berry. In summary, single and combined mutations in gf, r and y generated interesting visual
and compositional diversity in the SM landrace, while maintaining its original typolog
Accelerating tomato breeding by exploiting genomic selection approaches
Genomic selection (GS) is a predictive approach that was built up to increase the rate of genetic gain per unit of time and reduce the generation interval by utilizing genome-wide markers in breeding programs. It has emerged as a valuable method for improving complex traits that are controlled by many genes with small effects. GS enables the prediction of the breeding value of candidate genotypes for selection. In this work, we address important issues related to GS and its implementation in the plant context with special emphasis on tomato breeding. Genomic constraints and critical parameters affecting the accuracy of prediction such as the number of markers, statistical model, phenotyping and complexity of trait, training population size and composition should be carefully evaluated. The comparison of GS approaches for facilitating the selection of tomato superior genotypes during breeding programs is also discussed. GS applied to tomato breeding has already been shown to be feasible. We illustrated how GS can improve the rate of gain in elite line selection, and descendent and backcross schemes. The GS schemes have begun to be delineated and computer science can provide support for future selection strategies. A new promising breeding framework is beginning to emerge for optimizing tomato improvement procedures
Biochemical features of native red wines and genetic diversity of the corresponding grape varieties from Campania Region
Campania region has always been considered one of the most appreciated Italian districts for wine production. Wine distinctiveness arises from their native grapevines. To better define the chemical profile of Campania autochthonous red grape varieties, we analysed the phenolic composition of Aglianico di Taurasi, Aglianico del Vulture, Aglianico del Taburno, Piedirosso wines, and a minor native variety, Lingua di Femmina in comparison with Merlot and Cabernet Sauvignon, as reference cultivars. A genetic profiling was also carried out using microsatellite molecular markers with high polymorphic and unambiguous profiles. Principal component analysis applied to 72 wines based on the 18 biochemical parameters, explained 77.6% of the total variance and highlighted important biological entities providing insightful patterns. Moreover, comparison of SSR-based data with phenylpropanoid molecules exhibited a statistically significant correlation. Our approach might be reasonably adopted for future characterisations and traceability of grapevines and corresponding wines
Secondary metabolite profile in induced tetraploids of wild Solanum commersonii Dun.
The main aim of this work was to study the leaf secondary metabolite profiles of artificially induced tetraploids (2n1⁄44x1⁄448) of Solanum commersonii, a diploid (2n1⁄42x1⁄424) wild potato species. The tetraploid genotypes of S. commersonii were produced by oryzalin treatment. Both HPLC-UV and LC/ MS analyses revealed that there were no qualitative differences in the metabolite profiles between the diploid S. commersonii and its tetraploids. By contrast, the results showed that the phenylpropanoid content was generally significantly higher in the tetraploids than in the diploid S. commersonii. Concerning the glycoalkaloids (GAs), the results provided evidence that the content of minor GAs (solanidenediol triose, solanidadienol lycotetraose, and solanidenol lycotetraose) was higher in tetraploids than in the diploid progenitor, while the content of major GAs (dehydrodemissine and dehydrocommersonine) was significantly higher in diploid S. commersonii than in its tetraploid genotypes. The results are discussed from the practical perspective of potato biodiversity enhancement
Nutritional controlled preparation and administration of different tomato purées indicate increase of β-carotene and lycopene isoforms, and of antioxidant potential in human blood bioavailability: A pilot study
The isoforms of lycopene, carotenoids, and their derivatives including precursors of vitamin A are compounds relevant for preventing chronic degenerative diseases such as cardiovascular diseases and cancer. Tomatoes are a major source of these compounds. However, cooking and successive metabolic processes determine the bioavailability of tomatoes in human nutrition. To evaluate the effect of acute/chronic cooking procedures on the bioavailability of lycopene and carotene isoforms in human plasma, we measured the blood levels of these compounds and of the serum antioxidant potential in volunteers after a meal containing two different types of tomato sauce (rustic or strained). Using a randomized cross-over administration design, healthy volunteers were studied, and the above indicated compounds were determined by HPLC. The results indicate an increased bioavailability of the estimated compounds and of the serum antioxidant potential with both types of tomato purée and the subsequently derived sauces (the increase was greater with strained purée). This study sheds light on the content of nutrient precursors of vitamin A and other antioxidant compounds derived from tomatoes cooked with different strategies. Lastly, our study indicates that strained purée should be preferred over rustic purée
A novel synthetic peptide from a tomato defensin exhibits antibacterial activities against Helicobacter pylori
Defensins are a class of cysteine-rich proteins, which exert broad spectrum antimicrobial activity. In this work, we used a bioinformatic approach to identify putative defensins in the tomato genome. Fifteen proteins had a mature peptide that includes the well-conserved tetradisulfide array. We selected a representative member of the tomato defensin family; we chemically synthesized its gamma-motif and tested its antimicrobial activity. Here, we demonstrate that the synthetic peptide exhibits potent antibacterial activity against Gram-positive bacteria, such as Staphylococcus aureus A170, Staphylococcus epidermidis, and Listeria monocytogenes, and Gram-negative bacteria, including Salmonella enterica serovar Paratyphi, Escherichia coli, and Helicobacter pylori. In addition, the synthetic peptide shows minimal (<5%) hemolytic activity and absence of cytotoxic effects against THP-1 cells. Finally, SolyC exerts an anti-inflammatory activity in vitro, as it downregulates the level of the proinflammatory cytokines TNF-alpha and IFN-gamma
