    Maternal and infant infections stimulate a rapid leukocyte response in breastmilk

    Breastmilk protects infants against infections; however, specific responses of breastmilk immune factors to different infections of either the mother or the infant are not well understood. Here, we examined the baseline range of breastmilk leukocytes and immunomodulatory biomolecules in healthy mother/infant dyads and how they are influenced by infections of the dyad. Consistent with a greater immunological need in the early postpartum period, colostrum contained considerable numbers of leukocytes (13–70% out of total cells) and high levels of immunoglobulins and lactoferrin. Within the first 1–2 weeks postpartum, leukocyte numbers decreased significantly to a low baseline level in mature breastmilk (0–2%) (P\u3c0.001). This baseline level was maintained throughout lactation unless the mother and/or her infant became infected, when leukocyte numbers significantly increased up to 94% leukocytes out of total cells (P\u3c0.001). Upon recovery from the infection, baseline values were restored. The strong leukocyte response to infection was accompanied by a more variable humoral immune response. Exclusive breastfeeding was associated with a greater baseline level of leukocytes in mature breastmilk. Collectively, our results suggest a strong association between the health status of the mother/infant dyad and breastmilk leukocyte levels. This could be used as a diagnostic tool for assessment of the health status of the lactating breast as well as the breastfeeding mother and infant

    Zusammenfassung: Heterotope Ossifikationen werden als abnorme Bildung von Knochen in Weichteilgeweben definiert. Man unterscheidet die erworbene von den angeborenen Formen. Die erworbene Form, deren Pathogenese weitgehend ungeklärt ist, wird häufig bei Patienten mit Schädel-Hirn-Trauma, Rückenmarkverletzung, Trauma des Bewegungsapparats oder nach Verbrennungen beobachtet. Klinisch präsentieren sich die heterotopen Ossifikationen häufig symptomfrei, im Initialstadium mit unspezifischen Symptomen, ausgeprägte Formen können jedoch zu schwerwiegenden Funktionseinschränkungen führen. Zur Diagnosesicherung wird insbesondere die Knochenszintigraphie verwendet. Lokale Bestrahlung und nichtsteroidale Antiphlogistika bilden die klassischen Therapie- und Prophylaxeoptionen.     Rheumatoid arthritis synovium contains plasmacytoid dendritic cells

    We have previously described enrichment of antigen-presenting HLA-DR(+ )nuclear RelB(+ )dendritic cells (DCs) in rheumatoid arthritis (RA) synovium. CD123(+)HLA-DR(+ )plasmacytoid DCs (pDCs) and their precursors have been identified in human peripheral blood (PB), lymphoid tissue, and some inflamed tissues. We hypothesized recruitment of pDCs into the inflamed RA synovial environment and their contribution as antigen-presenting cells (APCs) and inflammatory cells in RA. CD11c(+ )myeloid DCs and CD123(+ )pDCs were compared in normal and RA PB, synovial fluid (SF), and synovial tissue by flow cytometry, immunohistochemistry, and electron microscopy and were sorted for functional studies. Nuclear RelB(-)CD123(+ )DCs were located in perivascular regions of RA, in a similar frequency to nuclear RelB(+)CD123(- )DCs, but not normal synovial tissue sublining. Apart from higher expression of HLA-DR, the numbers and phenotypes of SF pDCs were similar to those of normal PB pDCs. While the APC function of PB pDCs was less efficient than that of PB myeloid DCs, RA SF pDCs efficiently activated resting allogeneic PB T cells, and high levels of IFN-γ, IL-10, and tumor necrosis factor α were produced in response to incubation of allogeneic T cells with either type of SF DCs. Thus, pDCs are recruited to RA synovial tissue and comprise an APC population distinct from the previously described nuclear RelB(+ )synovial DCs. pDCs may contribute significantly to the local inflammatory environment

    Improvements in Gold Nanorod Biocompatibility with Sodium Dodecyl Sulfate Stabilization

    Due to their well-defined plasmonic properties, gold nanorods (GNRs) can be fabricated with optimal light absorption in the near-infrared region of the electromagnetic spectrum, which make them suitable for cancer-related theranostic applications. However, their controversial safety profile, as a result of surfactant stabilization during synthesis, limits their clinical translation. We report a facile method to improve GNR biocompatibility through the presence of sodium dodecyl sulfate (SDS). GNRs (120 Ă— 40 nm) were synthesized through a seed-mediated approach, using cetyltrimethylammonium bromide (CTAB) as a cationic surfactant to direct the growth of nanorods and stabilize the particles. Post-synthesis, SDS was used as an exchange ligand to modify the net surface charge of the particles from positive to negative while maintaining rod stability in an aqueous environment. GNR cytotoxic effects, as well as the mechanisms of their cellular uptake, were examined in two different cancer cell lines, Lewis lung carcinoma (LLC) and HeLa cells. We not only found a significant dose-dependent effect of GNR treatment on cell viability but also a time-dependent effect of GNR surfactant charge on cytotoxicity over the two cell lines. Our results promote a better understanding of how we can mediate the undesired consequences of GNR synthesis byproducts when exposed to a living organism, which so far has limited GNR use in cancer theranostics

    This paper aims to identify the possible institution of a Sociology of the Body in Brazil. The body is understood as a social construct, as a phenomenon that exists within, and through, the social sphere, producing sense and meaning through socially located and manifest forms of embodiment. From a survey of Brazilian Social Science (specifically Sociology and Anthropology) and Physical Education papers, we identified sufficient research output to suggest the incipient nature of the Sociology of the Body in Brazil.El objetivo de este artículo es identificar la posible existencía de una sociología del cuerpo en Brasil. El cuerpo es entendido como una construcción social – como fenómeno que existe dentro y a través del social – produciendo sentidos y significados ubicados socialmente y que se manifestan en y a través de la corporeidad. A partir de uma revisión de la literatura en periódicos brasileños de los campos de conocimiento de las ciências sociales (específicamente sociología y antropología) y de la educación física, fueron identificados resultados que presentan el caracter incipiente de la sociologia del cuerpo en Brasil.O objetivo deste artigo é identificar a possível constituição de uma sociologia do corpo no Brasil. O corpo é entendido como uma construção social – como um fenômeno que existe dentro e através da esfera social – produzindo sentidos e significados localizados socialmente e manifestados na e pela corporeidade.     Impact of extracellular vesicle isolation methods on downstream mirna analysis in semen: A comparative study

    Seminal plasma (SP) contains a unique concentration of miRNA, mostly contained in small extracellular vesicles (sEVs) such as exosomes, some of which could be clinically useful for diagnosis and/or prognosis of urogenital diseases such as prostate cancer (PCa). We optimized several exosome-EV isolation technologies for their use in semen, evaluating EV purifying effectiveness and impact on the downstream analysis of miRNAs against results from the standard ultracentrifugation (UC) method to implement the use of SP sEV_miRNAs as noninvasive biomarkers for PCa. Our results evidenced that commercial kits designed to isolate exosomes/EVs from blood or urine are mostly applicable to SP, but showed quantitative and qualitative variability between them. ExoGAG 3500x g and the miRCURY Cell/Urine/CSF 1500x g methods resulted as equivalent alternative procedures to UC for isolating exosomes/sEVs from semen for nanoparticle characteristics and quality of RNA contained in vesicles. Additionally, the expression profile of the altered semen sEV-miRNAs in PCa varies depending on the EV isolation method applied. This is possibly due to different extraction techniques yielding different proportions of sEV subtypes. This is evidence that the exosome-EV isolation method has a significant impact on the analysis of the miRNAs contained within, with important consequences for their use as clinical biomarkers. Therefore, miRNA analysis results for EVs cannot be directly extrapolated between different EV isolation methods until clear markers for delineation between microvesicles and exosomes are established. However, EV extraction methodology affects combined models (semen exosome miRNA signatures plus blood Prostate specific antigen (PSA) concentration for PCa diagnosis) less; specifically our previously described (miR-142-3p + miR-142-5p + miR-223-3p + PSA) model functions as molecular marker from EVs from any of the three isolation methods, potentially improving the efficiency of PSA PCa diagnosis
