    Air-guided photonic-crystal-fiber pulse-compression delivery of multimegawatt femtosecond laser output for nonlinear-optical imaging and neurosurgery

    Cataloged from PDF version of article.Large-core hollow photonic- crystal fibers (PCFs) are shown to enable a fiber-format air-guided delivery of ultrashort infrared laser pulses for neurosurgery and nonlinear-optical imaging. With an appropriate dispersion precompensation, an anomalously dispersive 15-mu m-core hollow PCF compresses 510-fs, 1070-nm light pulses to a pulse width of about 110 fs, providing a peak power in excess of 5 MW. The compressed PCF output is employed to induce a local photodisruption of corpus callosum tissues in mouse brain and is used to generate the third harmonic in brain tissues, which is captured by the PCF and delivered to a detector through the PCF cladding.     Genomic analysis reveals hidden biodiversity within colugos, the sister group to primates

    Colugos are among the most poorly studied mammals despite their centrality to resolving supraordinal primate relationships. Two described species of these gliding mammals are the sole living members of the order Dermoptera, distributed throughout Southeast Asia. We generated a draft genome sequence for a Sunda colugo and a Philippine colugo reference alignment, and used these to identify colugo-specific genetic changes that were enriched in sensory and musculoskeletal-related genes that likely underlie their nocturnal and gliding adaptations. Phylogenomic analysis and catalogs of rare genomic changes overwhelmingly support the contested hypothesis that colugos are the sister group to primates (Primatomorpha), to the exclusion of treeshrews. We captured ~140 kb of orthologous sequence data from colugo museum specimens sampled across their range and identified large genetic differences between many geographically isolated populations that may result in a >300% increase in the number of recognized colugo species.     Implantable fiber-optic interface for parallel multisite long-term optical dynamic brain interrogation in freely moving mice

    Seeing the big picture of functional responses within large neural networks in a freely functioning brain is crucial for understanding the cellular mechanisms behind the higher nervous activity, including the most complex brain functions, such as cognition and memory. As a breakthrough toward meeting this challenge, implantable fiber-optic interfaces integrating advanced optogenetic technologies and cutting-edge fiber-optic solutions have been demonstrated, enabling a long-term optogenetic manipulation of neural circuits in freely moving mice. Here, we show that a specifically designed implantable fiber-optic interface provides a powerful tool for parallel long-term optical interrogation of distinctly separate, functionally different sites in the brain of freely moving mice. This interface allows the same groups of neurons lying deeply in the brain of a freely behaving mouse to be reproducibly accessed and optically interrogated over many weeks, providing a long-term dynamic detection of genome activity in response to a broad variety of pharmacological and physiological stimuli

    Electron spin manipulation and readout through an optical fiber

    The electron spin of nitrogen--vacancy (NV) centers in diamond offers a solid-state quantum bit and enables high-precision magnetic-field sensing on the nanoscale. Implementation of these approaches in a fiber format would offer unique opportunities for a broad range of technologies ranging from quantum information to neuroscience and bioimaging. Here, we demonstrate an ultracompact fiber-optic probe where a diamond microcrystal with a well-defined orientation of spin quantization NV axes is attached to the fiber tip, allowing the electron spins of NV centers to be manipulated, polarized, and read out through a fiber-optic waveguide integrated with a two-wire microwave transmission line. The microwave field transmitted through this line is used to manipulate the orientation of electron spins in NV centers through the electron-spin resonance tuned by an external magnetic field. The electron spin is then optically initialized and read out, with the initializing laser radiation and the photoluminescence spin-readout return from NV centers delivered by the same optical fiber

    CD133/prominin-1 is a potential therapeutic target for antibody-drug conjugates in hepatocellular and gastric cancers

    CD133/prominin-1 is a pentaspan transmembrane glycoprotein overexpressed in various solid tumours including colorectal and glioblastomas. CD133 was found here to be highly expressed in ⩾50% of pancreatic, gastric and intrahepatic cholangiocarcinomas. Quantitative flow cytometric analysis showed that a panel of established hepatocellular, pancreatic and gastric cancer cell lines expressed CD133 at levels higher than normal epithelial cells or bone marrow progenitor cells. A murine anti-human CD133 antibody (AC133) conjugated to a potent cytotoxic drug, monomethyl auristatin F (MMAF), effectively inhibited the growth of Hep3B hepatocellular and KATO III gastric cancer cells in vitro with IC50 values of 2–7 ng ml−1. MMAF induced apoptosis in the cancer cells as measured by caspase activation. The anti-CD133-drug conjugate (AC133-vcMMAF) was shown to internalise and colocalised with the lysosomal marker CD107a in the sensitive cell lines. In contrast, in the resistant cell line Su.86.86, the conjugate internalised and colocalised with the caveolae marker, Cav-1. Addition of ammonium chloride, an inhibitor of lysosomal trafficking and processing, suppressed the cytotoxic effect of AC133-vcMMAF in both Hep3B and KATO III. Anti-CD133-drug conjugate treatment resulted in significant delay of Hep3B tumour growth in SCID mice. Anti-CD133 antibody-drug conjugates warrant further evaluation as a therapeutic strategy to eradicate CD133+ tumours

There was developed several ways for surgical correction of foot deformation at DNOAP in recent years, however, there is no much known about their use long-term results. This article presents the successful complex treatment and 10-years outcomes results in patient with type 2 diabetes and both feet deformation due to the DNOAP.Диабетическая нейроостеоартропатия (ДНОАП) – тяжелое осложнение сахарного диабета (СД), которое может привести к потере конечности. В последние годы было разработано несколько вариантов хирургической коррекции деформаций стопы при ДНОАП, однако об отдаленных результатах их использования пока что известно мало.
