    A comparison of baleen whale density estimates derived from overlapping satellite imagery and a shipborne survey

    As whales recover from commercial exploitation, they are increasing in abundance in habitats that they have been absent from for decades. However, studying the recovery and habitat use patterns of whales, particularly in remote and inaccessible regions, frequently poses logistical and economic challenges. Here we trial a new approach for measuring whale density in a remote area, using Very-High-Resolution WorldView-3 satellite imagery. This approach has capacity to provide sightings data to complement and assist traditional sightings surveys. We compare at-sea whale density estimates to estimates derived from satellite imagery collected at a similar time, and use suction-cup archival logger data to make an adjustment for surface availability. We demonstrate that satellite imagery can provide useful data on whale occurrence and density. Densities, when unadjusted for surface availability are shown to be considerably lower than those estimated by the ship survey. However, adjusted for surface availability and weather conditions (0.13 whales per km2, CV = 0.38), they fall within an order of magnitude of those derived by traditional line-transect estimates (0.33 whales per km2, CV = 0.09). Satellite surveys represent an exciting development for high-resolution image-based cetacean observation at sea, particularly in inaccessible regions, presenting opportunities for ongoing and future research

    Lack of mitochondrial topoisomerase I (TOP1mt) impairs liver regeneration

    The liver has an exceptional replicative capacity following partial hepatectomy or chemical injuries. Cellular proliferation requires increased production of energy and essential metabolites, which critically depend on the mitochondria. To determine whether Top1mt, the vertebrate mitochondrial topoisomerase, is involved in this process, we studied liver regeneration after carbon tetrachloride (CCl4) administration. TOP1mt knockout (KO) mice showed a marked reduction in regeneration and hepatocyte proliferation. The hepatic mitochondrial DNA (mtDNA) failed to increase during recovery from CCl4 exposure. Reduced glutathione was also depleted, indicating increased reactive oxygen species (ROS). Steady-state levels of ATP, O2 consumption, mtDNA, and mitochondrial mass were also reduced in primary hepatocytes from CCl4-treated KO mice. To further test whether Top1mt acted by enabling mtDNA regeneration, we tested TOP1mt KO fibroblasts and human colon carcinoma HCT116 cells and measured mtDNA after 3-d treatment with ethidium bromide. Both types of TOP1mt knockout cells showed defective mtDNA regeneration following mtDNA depletion. Our study demonstrates that Top1mt is required for normal mtDNA homeostasis and for linking mtDNA expansion with hepatocyte proliferation

    Using a risk assessment framework to spatially and temporally spread the fishery catch limit for Antarctic krill in the west Antarctic Peninsula: A template for krill fisheries elsewhere

    The west Antarctic Peninsula is an important breeding and foraging location for marine predators that consume Antarctic Krill (Euphasia superba). It is also an important focus for the commercial fishery for Antarctic krill, managed by the Commission for the Conservation of Antarctic Marine Living Resources (CCAMLR). Aiming to minimise ecosystem risks from fishing, whilst enabling a sustainable fishery, CCAMLR has recently endorsed a new management framework that incorporates information about krill biomass estimates, sustainable harvest rates and a risk assessment to spatially and temporally distribute catch limits. We have applied a risk assessment framework to the west Antarctic Peninsula region (Subarea 48.1), with the aim of identifying the most appropriate management units by which to spatially and temporally distribute the local catch limit. We use the best data currently available for implementing the approach, recognising the framework is flexible and can accommodate new data, when available, to improve future estimates of risk. We evaluated 36 catch distribution scenarios for managing the fishery and provide advice about the scale at which the krill fishery can be managed. We show that the spatial distribution with which the fishery currently operates presents some of the highest risks of all scenarios evaluated. We highlight important issues that should be resolved. We emphasize that for the risk assessment to provide robust estimates of risk, it is important that the management units are at a similar scale to ecosystem function. Managing the fishery at small scales has the lowest risk but may necessitate a high level of management interaction. Our results offer advice to CCAMLR about near-term management and could provide a template for the rest of the southwest Atlantic (Area 48), or fisheries elsewhere. As each data layer influences the outcome of the risk assessment, we recommend that updated estimates of the distribution, abundance and consumption of krill, and estimates of available krill biomass will be key as CCAMLR moves forward to develop a longer-term management strategy. Finally, we highlight that the endeavours reported here are the result of a community effort, demonstrating the value of common enterprise

    Education for innovation and entrepreneurship in the food system: the Erasmus+ BoostEdu approach and results

    Innovation and entrepreneurship are key factors to provide added value for food systems. Based on the findings of the Erasmus+ Strategic Partnership BoostEdu, the objective of this paper is to provide answers to three knowledge gaps: 1) identify the needs for innovation and entrepreneurship (I&E) in the food sector; 2) understand the best way to organize learning; 3) provide flexibility in turbulent times. BoostEdu aimed to provide a platform for continuing education within I&E for food professionals and was carried out through co-creation workshops and the development of an e-learning course. The results of the project in particular during the Covid-19 pandemics, highlighted the need for flexible access to modules that are complementary to other sources and based on a mix of theoretical concepts and practical experiences. The main lessons learned concern the need of co-creation and co-learning processes to identify suitable practices for the use of innovative digital technologies

    Effect of plasma activated water (PAW) on rocket leaves decontamination and nutritional value

    Plasma Activated Water (PAW) obtained by exposing water to cold atmospheric pressure plasma, has recently emerged as a promising alternative for food decontamination, compared to the use of traditional chemical sanitizers. The aim of the study was to evaluate the efficacy of PAW treatments for rocket salad decontamination. Washing with PAW for 2, 5, 10 and 20 min was assessed against different endogenous spoilage microorganisms and compared to untreated water and hypochlorite solution. The chemical composition of PAW as a function of treatment and delay time was characterized and the effect on product quality and nutritional parameters was evaluated. Results showed that PAW allowed an average reduction of 1.7–3 Log CFU/g for total mesophilic and psychrotrophic bacteria and Enterobacteriaceae following 2–5 min washing with minimal variation of qualitative and nutritional parameters. Overall, experimental results highlighted the potentiality of PAW treatments as a promising alternative to chlorine having the advantage of a minor adverse impact on environment and consumers' health. Industrial relevance: To meet consumers demand, the minimally processed fruit and vegetable industry needs to find sustainable solutions as alternative to the use of traditional chemical sanitizers that allow to increase product shelf-life and preserve safety, qualitative and nutritional characteristics. Plasma activated water represents a promising strategy for food decontamination, but its effects on foods have been only limitedly investigated. The present research is the first study on the use of plasma activated water on fresh rocket leaves, providing new and important information on microbial inactivation and quality of the fresh cut product

    Identification of a novel heterozygous guanosine monophosphate reductase (GMPR) variant in a patient with a late-onset disorder of mitochondrial DNA maintenance

    Autosomal dominant progressive external ophthalmoplegia (adPEO) is a late-onset, Mendelian mitochondrial disorder characterised by paresis of the extraocular muscles, ptosis and skeletal-muscle restricted multiple mitochondrial DNA (mtDNA) deletions. While dominantly-inherited, pathogenic variants in POLG, TWNK and RRM2B are among the most common genetic defects of adPEO, identification of novel candidate genes and the underlying pathomechanisms remain challenging. We report the clinical, genetic and molecular investigations of a patient who presented in the seventh decade of life with PEO. Oxidative histochemistry revealed cytochrome c oxidase deficient fibres and occasional ragged red fibres showing subsarcolemmal mitochondrial accumulation in skeletal muscle, while molecular studies identified the presence of multiple mtDNA deletions. Negative candidate screening of known nuclear genes associated with PEO prompted diagnostic exome sequencing, leading to the prioritisation of a novel heterozygous c.547G > C variant in GMPR (NM_006877.3) encoding guanosine monophosphate reductase, a cytosolic enzyme required for maintaining the cellular balance of adenine and guanine nucleotides. We show that the novel c.547G > C variant causes aberrant splicing, decreased GMPR protein levels in patient skeletal muscle, proliferating and quiescent cells and is associated with subtle changes in nucleotide homeostasis protein levels and evidence of disturbed mtDNA maintenance in skeletal muscle. Despite confirmation of GMPR deficiency, demonstrating marked defects of mtDNA replication or nucleotide homeostasis in patient cells proved challenging. Our study proposes that GMPR is the nineteenth (19th) locus for PEO and highlights the complexities of uncovering disease mechanisms in late-onset PEO phenotypes

    Following their near extirpation by industrial whaling of the 20th century, the population status of Southern Hemisphere fin whales (SHFW) remains unknown. Systematic surveys estimating fin whale abundance in the Southern Ocean are not yet available. Records of fin whale sightings have been collected by a variety of organisations over the past few decades, incorporating both opportunistic data and dedicated survey data. Together, these isolated data sets represent a potentially valuable source of information on the seasonality, distribution and abundance of SHFW. We compiled records across 40 years from the Antarctic Peninsula and Scotia Sea from multiple sources and used a novel approach combining ensemble learning and a maximum entropy model to estimate abundance and distribution of SHFW in this region. Our results show a seasonal distribution pattern with pronounced centres of distribution from January-March along the West Antarctic Peninsula. Our new approach allowed us to estimate abundance of SHFW for discrete areas from a mixed data set of mainly opportunistic presence only data

    LETM1 couples mitochondrial DNA metabolism and nutrient preference

    The diverse clinical phenotypes of Wolf\u2013Hirschhorn syndrome (WHS) are the result of haploinsufficiency of several genes, one of which, LETM1, encodes a protein of the mitochondrial inner membrane of uncertain function. Here, we show that LETM1 is associated with mitochondrial ribosomes, is required for mitochondrial DNA distribution and expression, and regulates the activity of an ancillary metabolic enzyme, pyruvate dehydrogenase. LETM1 deficiency in WHS alters mitochondrial morphology and DNA organization, as does substituting ketone bodies for glucose in control cells. While this change in nutrient availability leads to the death of fibroblasts with normal amounts of LETM1, WHS-derived fibroblasts survive on ketone bodies, which can be attributed to their reduced dependence on glucose oxidation. Thus, remodeling of mitochondrial nucleoprotein complexes results from the inability of mitochondria to use specific substrates for energy production and is indicative of mitochondrial dysfunction. However, the dysfunction could be mitigated by a modified diet\u2014for WHS, one high in lipids and low in carbohydrates
