    Automation of an atomic force microscope via Arduino

This technique often overcomes the problems and capabilities of electron microscopy when characterizing few nanometers thin coatings over solid substrates. They are expensive, in the half million dollar range for standard units, and therefore it is often difficult to upgrade to new units with improved characteristics. One of these improvements, motorization and automation of the measurements is very interesting to sample different parts of a substrate in an unattended way. Here we report a low cost upgrade under 60 $ to a Dimension 3000 AFM based on a control unit using an Arduino Leonardo.     Fortification and bioaccessibility of saffron apocarotenoids in potato tubers

    Carotenoids are C40 isoprenoids with well-established roles in photosynthesis, pollination, photoprotection, and hormone biosynthesis. The enzymatic or ROS-induced cleavage of carotenoids generates a group of compounds named apocarotenoids, with an increasing interest by virtue of their metabolic, physiological, and ecological activities. Both classes are used industrially in a variety of fields as colorants, supplements, and bio-actives. Crocins and picrocrocin, two saffron apocarotenoids, are examples of high-value pigments utilized in the food, feed, and pharmaceutical industries. In this study, a unique construct was achieved, namely O6, which contains CsCCD2L, UGT74AD1, and UGT709G1 genes responsible for the biosynthesis of saffron apocarotenoids driven by a patatin promoter for the generation of potato tubers producing crocins and picrocrocin. Different tuber potatoes accumulated crocins and picrocrocin ranging from 19.41–360 to 105–800 μg/g DW, respectively, with crocetin, crocin 1 [(crocetin-(β-D-glucosyl)-ester)] and crocin 2 [(crocetin)-(β-D-glucosyl)-(β-D-glucosyl)-ester)] being the main compounds detected. The pattern of carotenoids and apocarotenoids were distinct between wild type and transgenic tubers and were related to changes in the expression of the pathway genes, especially from PSY2, CCD1, and CCD4. In addition, the engineered tubers showed higher antioxidant capacity, up to almost 4-fold more than the wild type, which is a promising sign for the potential health advantages of these lines. In order to better investigate these aspects, different cooking methods were applied, and each process displayed a significant impact on the retention of apocarotenoids. More in detail, the in vitro bioaccessibility of these metabolites was found to be higher in boiled potatoes (97.23%) compared to raw, baked, and fried ones (80.97, 78.96, and 76.18%, respectively). Overall, this work shows that potatoes can be engineered to accumulate saffron apocarotenoids that, when consumed, can potentially offer better health benefits. Moreover, the high bioaccessibility of these compounds revealed that potato is an excellent way to deliver crocins and picrocrocin, while also helping to improve its nutritional value

Methodological aspects, data issues, approaches, limitations, gaps and further steps for improvement are analysed for providing good practices and decision making guidance. The EU initiative on Mapping and Assessment of the state of Ecosystems and their Services (MAES), with the support of all Member States, contributes to improve the knowledge on ecosystem services.     Esta memoria refleja el trabajo de la red 5474 sobre el conocimiento y percepción de un canon de lecturas de cómic y álbum ilustrados que aborden imaginarios de Hispanoamérica entre el alumnado universitario. Se explica la elaboración y difusión de una encuesta en relación a diversas lecturas vinculadas a América Latina seleccionados y supervisados por el grupo Unicòmic de la Universidad de Alicante a partir de publicaciones especializadas, y con la cual se pretende conocer el grado de conocimiento del imaginario cultural, histórico y literario de América Latina entre alumnado universitario. Inicialmente, la pretensión era que esta encuesta se extendiera a facultades diversas de la UA y la UMH y de la Universidad de Almería. Sin embargo, el cuestionario se ha centrado en alumnado de la Facultad de Educación de la UA. En todo caso, este proyecto representa un paso adelante en un proceso de investigación que, después otras redes docentes en esta universidad y en las cuales se ha ido compilando un corpus bibliográfico alrededor del canon artístico del cómic, ahora nos permite avanzar en la elaboración de listados inclusivos con una mayor visibilización del mundo cultural y literario de América Latina

    Natural Assurance Schemes Canvas: A Framework to Develop Business Models for Nature-Based Solutions Aimed at Disaster Risk Reduction

    International audienceNature-based solutions (NBS) are increasingly being promoted because they can solve different pursued aims together with providing an additional array of multiple ecosystem services or co-benefits. Nevertheless, their implementation is still being curbed by several barriers, for example, a lack of examples, a lack of finance, and a lack of business cases. Therefore, there is an urgent need to facilitate the construction of business models and business cases that identify the elements required to capture value. These are necessary to catalyze investments for the implementation of NBS. This article presents a tool called a Natural Assurance Schemes (NAS) canvas and explains how it can be applied to identify business models for NBS strategies providing climate adaptation services, showing an eye-shot summary of critical information to attract funding. The framework is applied in three case studies covering different contexts, scales, and climaterelated risks (floods and droughts). Finally, a reflective analysis is done, comparing the tool with other similar approaches while highlighting the differential characteristics that define the usefulness, replicability, and flexibility of the tool for the target users, namely policymakers, developers, scientists, or entrepreneurs aiming to promote and implement NAS and NBS projects

    KIR2DL2/S2 and KIR2DS5 in alcoholic cirrhotic patients undergoing liver transplantation

    Introduction: The molecular mechanisms underlying alcoholic liver fibrosis and cirrhosis are not completely understood. Hepatic fibrosis involves the interplay of diverse cells and factors, including hepatic stellate cells (HSCs), Kupffer, NK cells, and T-lymphocyte subsets. Killer-cell immunoglobulin-like receptors (KIR) are membrane receptors involved in mediation between NK and activated HSCs, regulating NK cell function through their interaction with HLA-I molecules. The aim of this study was to analyse the genetic association between KIR genes and the susceptibility to or protection from alcoholic cirrhosis (AC) in a cohort of male AC patients undergoing liver transplantation (LT) with and without concomitant viral infections. Material and methods: KIR genotyping was performed in nuclear DNA extracted from 281 AC patients and compared with 319 male controls. Results: Significant differences between total AC patients and healthy controls were only found in the case of KIR2DL2 and KIR2DS5. KIR2DL2 was significantly underrepresented in non-viral AC patients (52.6% vs. 63.3%; p = 0.015), while patients heterozygous for KIR2DL2 were also underrepresented in the non-viral AC group compared with controls (p = 0.034). KIR2DS5 was overrepresented in this group compared with healthy controls (p = 0.002). All these observations were only evident in AC patients older than 54 years old. Conclusions: Our data suggest a contrary effect of KIR2DL2 and KIR2DS5 in AC patients older than 54 years, in whom the presence of KIR2DL2 appears to be protective against AC, whereas the presence of KIR2DS5 seems to promote the fibrotic process, particularly in patients with no associated viral infection
