    Sensitivity analysis of a branching process evolving on a network with application in epidemiology

    We perform an analytical sensitivity analysis for a model of a continuous-time branching process evolving on a fixed network. This allows us to determine the relative importance of the model parameters to the growth of the population on the network. We then apply our results to the early stages of an influenza-like epidemic spreading among a set of cities connected by air routes in the United States. We also consider vaccination and analyze the sensitivity of the total size of the epidemic with respect to the fraction of vaccinated people. Our analysis shows that the epidemic growth is more sensitive with respect to transmission rates within cities than travel rates between cities. More generally, we highlight the fact that branching processes offer a powerful stochastic modeling tool with analytical formulas for sensitivity which are easy to use in practice.Comment: 17 pages (30 with SI), Journal of Complex Networks, Feb 201

    The importance of quality family time on child development

    Minimal research has been done to date on how family time and children’s activities impact a child’s development. The amount of quality time that families spend together has decreased in recent years and children have fallen behind in different developmental areas. This idea prompts the question if there is a correlation between how children spend their time and the child’s developmental state.A longitudinal study uncovered that how a mother spends time impacts her child’s behavior and academics. This study found a positive link between maternal education levels and the child’s academic success as well as increased time spent with the mother, the child displayed less “delinquent behaviors” (Milkie et al., 2015). Another study aimed to discover how COVID-19 impacted a child’s behavior, speech and language. This study found that children with disadvantaged backgrounds fell behind on their social skills (Ofsted, 2020). The results displayed that about ÂŒ of the participants thought that these areas had improved with families spending more time talking and reading together. Hofferth & Sandberg (2001) reported on how family factors impacted the child’s behaviors and cognitive abilities. The results indicated that child’s activities as well as demographics played a role in family activities which influenced the child’s development. This correlational study aims to further investigate how family time may impact a child’s development. The online survey allowed participants to answer questions about demographics, how the child spent time, how parents spent time with their child, and the child’s behavior and academic abilities. Findings suggest that how a child spends their time, impacts their behavior and academic achievements. Race, parental education, time spent eating, reading for pleasure, and family time all had impacted the child’s behavior or their academic abilities. Some of these findings have consistencies with past research to suggest that family time is important for a child. For example, children who read for pleasure scored higher on achievement tests. Results also suggest that the amount of time spent engaged in family activities decreased behavioral issues in the child. With the limited number of participants in the current study, future research should aim to expand their number of participants. Interviewing participants may also result in consistent interpretation of all survey questions. In addition, future researchers may analyze the activities studied to determine if new activities should be addressed

    HDAC inhibition potentiates immunotherapy in triple negative breast cancer.

    Triple-negative breast cancer (TNBC) represents a more aggressive and difficult subtype of breast cancer where responses to chemotherapy occur, but toxicity is significant and resistance often follows. Immunotherapy has shown promising results in various types of cancer, including breast cancer. Here, we investigated a new combination strategy where histone deacetylase inhibitors (HDACi) are applied with immune checkpoint inhibitors to improve immunotherapy responses in TNBC. Testing different epigenetic modifiers, we focused on the mechanisms underlying HDACi as priming modulators of immunotherapy. Tumor cells were co-cultured with human peripheral blood mononuclear cells (PBMCs) and flow cytometric immunophenotyping was performed to define the role of epigenetic priming in promoting tumor antigen presentation and immune cell activation. We found that HDACi up-regulate PD-L1 mRNA and protein expression in a time-dependent manner in TNBC cells, but not in hormone responsive cells. Focusing on TNBC, HDACi up-regulated PD-L1 and HLA-DR on tumor cells when co-cultured with PBMCs and down-regulated CD4+ Foxp3+ Treg in vitro. HDACi significantly enhanced the in vivo response to PD-1/CTLA-4 blockade in the triple-negative 4T1 breast cancer mouse model, the only currently available experimental system with functional resemblance to human TNBC. This resulted in a significant decrease in tumor growth and increased survival, associated with increased T cell tumor infiltration and a reduction in CD4+ Foxp3+ T cells in the tumor microenvironment. Overall, our results suggest a novel role for HDAC inhibition in combination with immune checkpoint inhibitors and identify a promising therapeutic strategy, supporting its further clinical evaluation for TNBC treatment

    Gymnosperms from the Middle Triassic of Antarctica: The First Structurally Preserved Cycad Pollen Cone

    The first permineralized cycad pollen cone is described from the lower Middle Triassic of Antarctica. The cone is characterized by helically arranged, wedge‐shaped microsporophylls, each with five or more spinelike projections extending from the rhomboid distal face. The vascular cylinder is dissected and produces paired traces to each microsporophyll. Three vascular bundles enter the base of the microsporophyll and divide to produce at least five vascular strands in the sporophyll lamina. Pollen sacs occur in two radial clusters near the lateral margins on the abaxial surface of the microsporophyll. Each cluster bears up to eight elongate pollen sacs that are fused for approximately half their length and display longitudinal dehiscence. Pollen sacs are sessile and attached to a vascularized, receptacle‐like pad of tissue that is raised from the surface of the microsporophyll. Pollen is ovoid, psilate, and monosulcate. Although the affinities of this cone with the Cycadales are obvious, the complement of characters in the fossil is unique and thus does not permit assignment to an extant family.     Comparison of a Continuous and Discontinuous GXT on VO2 in Resistance-Trained and Endurance-Trained Males

Traditional graded exercise testing to assess maximal oxygen uptake (VO2max) may not well represent resistance-trained athletes due to their unfamiliarity with continuous exercise. For this reason, it is possible discontinuous exercise protocols may better represent the maximum capacity for aerobic metabolism in resistance-trained athletes, in order to provide a more valid assessment of VO2max and risk of developing cardiovascular disease. Purpose: The purpose of this experiment was to compare VO2peak during a continuous and discontinuous modified Bruce protocol in both highly resistance-trained and endurance-trained males. Methods: 19 college-aged males (age: 20.6 ± 1.9 yr, height: 176.5 ± 7.6 cm, weight: 85.0 ± 25.6 kg) of intermediate resistance- or endurance-trained status were recruited for this study. Participants completed a continuous and discontinuous modified Bruce protocol on two visits separated by seven days. Results: A 2x2 one-way ANOVA revealed a significant group main effect for VO2peak (p = 0.004) in which endurance athletes achieved significantly higher VO2peak values compared to resistance-trained athletes. A significant group main effect for RPE was found (p = 0.045) in which endurance-trained reported significantly higher RPE values than the resistance-trained. A significant main effect for protocol for heart rate (p = 0.033) was found in which individuals achieved higher heart rates during the continuous protocol compared to the discontinuous. Conclusion: Although a discontinuous protocol with rest periods between stages is comparable to the exercise mode familiar to resistance-trained athletes, it did not provide any additional benefit to VO2peak values

    Bursty egocentric network evolution in Skype

    In this study we analyze the dynamics of the contact list evolution of millions of users of the Skype communication network. We find that egocentric networks evolve heterogeneously in time as events of edge additions and deletions of individuals are grouped in long bursty clusters, which are separated by long inactive periods. We classify users by their link creation dynamics and show that bursty peaks of contact additions are likely to appear shortly after user account creation. We also study possible relations between bursty contact addition activity and other user-initiated actions like free and paid service adoption events. We show that bursts of contact additions are associated with increases in activity and adoption - an observation that can inform the design of targeted marketing tactics.Comment: 7 pages, 6 figures.
