    This paper reports on the status of a project whose aim is to implement and demonstrate in a real-life environment an integrated eAuthentication and eAuthorisation framework to enable trusted collaborations and delivery of services across different organisational/governmental jurisdictions. This aim will be achieved by designing a framework with assurance of claims, trust indicators, policy enforcement mechanisms and processing under encryption to address the security and confidentiality requirements of large distributed infrastructures. The framework supports collaborative secure distributed storage, secure data processing and management in both the cloud and offline scenarios and is intended to be deployed and tested in two pilot studies in two different domains, viz, Bio-security incident management and Ambient Assisted Living (eHealth). Interim results in terms of security requirements, privacy preserving authentication, and authorisation are reported

    Impact of post-fire management on soil respiration, carbon and nitrogen content in a managed hemiboreal forest

    Boreal forests are an important carbon (C) sink and fire is the main natural disturbance, directly affecting the C-cycle via emissions from combustion of biomass and organic matter and indirectly through long-term changes in C-dynamics including soil respiration. Carbon dioxide (CO2) emission from soil (soil respiration) is one of the largest fluxes in the global C-cycle. Recovery of vegetation, organic matter and soil respiration may be influenced by the intensity of post-fire management such as salvage logging. To study the impact of forest fire, fire and salvage, and recovery time on soil respiration and soil C and N content, we sampled two permanent research areas in north-western Estonia that were damaged by fire: Vihterpalu (59 degrees 13' N 23 degrees 49' E) in 1992 and Nova (59 degrees 10' N 23 degrees 45' E) in 2008. Three types of sample plots were established: 1) unburned control with no harvesting (CO); 2) burned and uncleared (BU); and 3) burned and cleared (BC). Measurements were made in 2013, 21 years after wildfire in Vihterpalu and 5 years after wildfire in NOva. Soil respiration ranged from 0.00 to 1.38 g CO2 m(-2) h(-1). Soil respiration in the burned and cleared areas (BC) was not reduced compared to burned and uncleared (BU) areas but the average soil respiration in unburned control areas was more than twice the value in burned areas (average soil respiration in CO areas was 0.34 CO2 m(-2) h(-1), versus 0.16 CO2 m(-2) h(-1), the average soil respiration of BC and BU combined). Recovery over 20 years was mixed; respiration was insignificantly lower on younger than older burned sites (when BC and BU values were combined, the average values were 0.15 vs. 0.17 g CO2 m(-2) h(-1), respectively); soil-C was greater in the older burned plots than the younger (when BC and BU values were combined, the average values were 9.71 vs. 5.99 kg m(-2), respectively); but root biomass in older and recently burned areas was essentially the same (average 2.23 and 2.11 kg m(-2), respectively); soil-N was highest on burned areas 20 years after fire. Twenty years post-fire may be insufficient time for carbon dynamics to fully recover on these low productivity sandy sites.Peer reviewe

    Development and Application of 3-Dimensional Transmission Electron Microscopy (3D-TEM) for the Characterization of Metal-Zeolite Catalyst Systems

    With electron tomography (3D-TEM) a 3D-reconstruction is calculated from a series of TEM images taken at a tilt angle range (tilting range) of +70° to -70°. The reconstruction can be visualized with contour surfaces that give information about the surface of the sample as well as with slices through the reconstruction that give detailed information on the interior of the sample. Electron tomography gives much more information than Scanning Electron Microscopy (SEM), since SEM gives only information about the surface of a sample. As a case study, the imaging of silver clusters on zeolite NaY is given. The reconstruction shows silver particles at the external surface as well as a silver particle in a mesopore of the zeolite crystallite. It is concluded that 3D-TEM comprises a breakthrough in the characterization of nano-structured solid catalysts

    Hantavirus Pulmonary Syndrome: CD8+and CD4+Cytotoxic T Lymphocytes to Epitopes on Sin Nombre Virus Nucleocapsid Protein Isolated during Acute Illness

    AbstractIn 1993 a number of cases of unexplained adult respiratory syndrome occurred in the southwestern United States. The illness was characterized by a prodrome of fever, myalgia, and other symptoms followed by the rapid onset of a capillary leak syndrome with hemoconcentration, thrombocytopenia, and pulmonary edema. Viral RNA sequences in the lungs identified a new member of the hantavirus genus, Sin Nombre virus (SNV), unique to North America. Pulmonary endothelial cells were heavily infected but were not necrotic. We speculated that this capillary leak syndrome was initiated by immune responses to the SNV-infected pulmonary endothelial cells. We isolated a CD8+cytotoxic T lymphocyte (CTL) clone directly from the blood of a patient with the acute hantavirus pulmonary syndrome (HPS) which recognizes a SNV specific epitope on the virus nucleocapsid protein (aa 234–242) that is restricted by HLA C7 and produces IFNγ but not IL-4. We identified a second CD8+CTL epitope located within another site aa 131–139 on the nucleocapsid protein, which is HLA B35 restricted, and a CD4+CTL epitope located on a third site on nucleocapsid protein aa 372–380 using lymphocytes obtained during HPS from another patient that were stimulatedin vitro.Hantavirus specific CD8+and CD4+CTL may contribute to the immunopathology and capillary leak syndrome observed in the HPS

    Drought Prediction for Socio-Cultural Stability Project

    The primary objective of this project is to answer the question: "Can existing, linked infrastructures be used to predict the onset of drought months in advance?" Based on our work, the answer to this question is "yes" with the qualifiers that skill depends on both lead-time and location, and especially with the associated teleconnections (e.g., ENSO, Indian Ocean Dipole) active in a given region season. As part of this work, we successfully developed a prototype drought early warning system based on existing/mature NASA Earth science components including the Goddard Earth Observing System Data Assimilation System Version 5 (GEOS-5) forecasting model, the Land Information System (LIS) land data assimilation software framework, the Catchment Land Surface Model (CLSM), remotely sensed terrestrial water storage from the Gravity Recovery and Climate Experiment (GRACE) and remotely sensed soil moisture products from the Aqua/Advanced Microwave Scanning Radiometer - EOS (AMSR-E). We focused on a single drought year - 2011 - during which major agricultural droughts occurred with devastating impacts in the Texas-Mexico region of North America (TEXMEX) and the Horn of Africa (HOA). Our results demonstrate that GEOS-5 precipitation forecasts show skill globally at 1-month lead, and can show up to 3 months skill regionally in the TEXMEX and HOA areas. Our results also demonstrate that the CLSM soil moisture percentiles are a goof indicator of drought, as compared to the North American Drought Monitor of TEXMEX and a combination of Famine Early Warning Systems Network (FEWS NET) data and Moderate Resolution Imaging Spectrometer (MODIS)'s Normalizing Difference Vegetation Index (NDVI) anomalies over HOA. The data assimilation experiments produced mixed results. GRACE terrestrial water storage (TWS) assimilation was found to significantly improve soil moisture and evapotransportation, as well as drought monitoring via soil moisture percentiles, while AMSR-E soil moisture assimilation produced marginal benefits. We carried out 1-3 month lead-time forecast experiments using GEOS-5 forecasts as input to LIS/CLSM. Based on these forecast experiments, we find that the expected skill in GEOS-5 forecasts from 1-3 months is present in the soil moisture percentiles used to indicate drought. In the case of the HOA drought, the failure of the long rains in April appears in the February 1, March 1 and April 1 initialized forecasts, suggesting that for this case, drought forecasting would have provided some advance warning about the drought conditions observed in 2011. Three key recommendations for follow-up work include: (1) carry out a comprehensive analysis of droughts observed over the entire period of record for GEOS-5 forecasts; (2) continue to analyze the GEOS-5 forecasts in HOA stratifying by anomalies in long and short rains; and (3) continue to include GRACE TWS, Soil Moisture/Ocean Salinity (SMOS) and the upcoming NASA Soil Moisture Active/Passive (SMAP) soil moisture products in a routine activity building on this prototype to further quantify the benefits for drought assessment and prediction

    P38 mitogen activated protein kinase regulates endothelial VCAM-1 expression at the post-transcriptional level

    The cytokine tumor necrosis factor (TNF) alpha was found to stimulate the p38 mitogen activated protein (MAP) kinase signalling cascade in human umbilical vein endothelial cells. TNFalpha increased the activity of the p38 substrate MAP kinase-activated-protein (MAPKAP) kinase 2 and the subsequent phosphorylation of the small heat shock protein Hsp27 about two to three fold. This stimulation was blocked almost completely by the specific p38 MAP kinase inhibitor SB203580. This inhibitor also suppressed the TNFalpha-induced surface expression of the endothelial adhesion molecule vascular cell adhesion molecule (VCAM)-1. In contrast, inhibition of p38 MAP kinase had no effect on the stimulated surface expression of the intercellular cell adhesion molecule (ICAM)-1. VCAM-1 mRNA accumulation induced by TNFalpha was not affected by SB203580, suggesting that the p38 MAP kinase signalling cascade regulates the endothelial expression of VCAM-1 at the post-transcriptional level

    Out-of-hospital cardiac arrest survival in international airports

    Background  The highest achievable survival rate following out-of-hospital cardiac arrest is unknown. Data from airports serving international destinations (international airports) provide the opportunity to evaluate the success of pre-hospital resuscitation in a relatively controlled but real-life environment.  Methods  This retrospective cohort study included all cases of out-of-hospital cardiac arrest at international airports with resuscitation attempted between January 1st, 2013 and December 31st, 2015. Crude incidence, patient, event characteristics and survival to hospital discharge/survival to 30 days (survival) were calculated. Mixed effect logistic regression analyses were performed to identify predictors of survival. Variability in survival between airports/countries was quantified using the median odds ratio.  Results  There were 800 cases identified, with an average of 40 per airport. Incidence was 0.024/100,000 passengers per year. Percentage survival for all patients was 32%, and 58% for patients with an initial shockable heart rhythm.  In adjusted analyses, initial shockable heart rhythm was the strongest predictor of survival (odds ratio, 36.7; 95% confidence interval [CI], 15.5 to 87.0). In the bystander-witnessed subgroup, delivery of a defibrillation shock by a bystander was a strong predictor of survival (odds ratio 4.8; 95% CI, 3.0 to 7.8). Grouping of cases was significant at country level and survival varied between countries.  Conclusions  In international airports, there was 32% of patients survived an out-of-hospital cardiac arrest, substantially more than in the general population. Our analysis suggested similarity between airports within countries, but differences between countries. Systematic data collection and reporting is essential to ensure international airports continually maximise activities to increase survival
