    MKM: Multiple Kernel Memory for Protecting Page Table Switching Mechanism Against Memory Corruption

    Countermeasures against kernel vulnerability attacks on an operating system (OS) are highly important kernel features. Some kernels adopt several kernel protection methods such as mandatory access control, kernel address space layout randomization, control flow integrity, and kernel page table isolation; however, kernel vulnerabilities can still be exploited to execute attack codes and corrupt kernel memory. To accomplish this, adversaries subvert kernel protection methods and invoke these kernel codes to avoid administrator privileges restrictions and gain complete control of the target host. To prevent such subversion, we present Multiple Kernel Memory (MKM), which offers a novel security mechanism using an alternative design for kernel memory separation that was developed to reduce the kernel attack surface and mitigate the effects of illegal data manipulation in the kernel memory. The proposed MKM is capable of isolating kernel memory and dedicates the trampoline page table for a gateway of page table switching and the security page table for kernel protection methods. The MKM encloses the vulnerable kernel code in the kernel page table. The MKM mechanism achieves complete separation of the kernel code execution range of the virtual address space on each page table. It ensures that vulnerable kernel code does not interact with different page tables. Thus, the page table switching of the trampoline and the kernel protection methods of the security page tables are protected from vulnerable kernel code in other page tables. An evaluation of MKM indicates that it protects the kernel code and data on the trampoline and security page tables from an actual kernel vulnerabilities that lead to kernel memory corruption. In addition, the performance results show that the overhead is 0.020μs to 0.5445μs, in terms of the system call latency and the application overhead average is 196.27 μs to 6,685.73 μs , for each download access of 100,000 Hypertext Transfer Protocol sessions

    Η εργασία έχει ως σκοπό την διερεύνηση της επίδρασης, στο επίπεδο προστασίας των προσφύγων, των πρόσφατων πολιτικών εξελίξεων και αποφάσεων, σε διεθνή, ευρωπαϊκή και εθνική κλίμακα, όπως αυτές εμφανίζονται στις διακηρύξεις των επίσημων επιμέρους θέσεων και πρακτικών, αλλά και στις διάφορες πρωτοβουλίες, νομοθετικές και άλλες. Σημαντικό ρόλο διαδραματίζουν η ανάλυση της πολυεπίπεδης διάρθρωσης του νομικού πλαισίου, που ρυθμίζει το καθεστώς χορήγησης ασύλου, καθώς και η μελέτη ευρύτερων παραμέτρων που συμβάλουν στη διαμόρφωσή του όπως η εθνική ασφάλεια και η κρατική κυριαρχία, ο αναδυόμενος εθνικισμός και η ξενοφοβία, αλλά και η οικονομική ύφεση (2008-13). Η διόγκωση των μεταναστευτικών ρευμάτων από τις χώρες της Μέσης Ανατολής, της Ασίας και της Βόρειας Αφρικής προς την Ευρώπη, ιδίως μέσω της Ελλάδας, με ορόσημο τα έτη 2015 και 2016, αλλά και η εκτίναξη του αριθμού των εν δυνάμει προσφύγων, προερχόμενων από χώρες που δημιουργούν ζητήματα διεθνούς προστασίας, τίθενται στο επίκεντρο της σχετικής προβληματικής. Η διαχείριση της προσφυγικής κρίσης και η επακόλουθη λήψη πολιτικών αποφάσεων τόσο σε επίπεδο ΕΕ όσο και σε διεθνές επίπεδο, επισφράγισε την επικράτηση ως προτεραιότητας της συνοριακής ασφάλειας, με την ταυτόχρονη εισαγωγή όρων γεωπολιτικής και διεθνών σχέσεων στο ήδη πολύπλοκο ζήτημα της διεθνούς προστασίας. Συμπερασματικά, θα λέγαμε, πως ενόψει της σύγκρουσης μεταξύ των κανονιστικών αρχών του διεθνούς και ενωσιακού δικαίου αφενός και, αφετέρου, της υιοθετούμενης πραγματιστικής προσέγγισης των κρατών της ΕΕ, τα δικαιώματα των προσφύγων φαίνεται ότι σχετικοποιούνται και σημαντικές διαστάσεις τους αποδυναμώνονται.This thesis aims to examine the impact of recent political developments and decisions at international, european and national level, as they appear in the official declarations and practices, but also in the various initiatives, legislative and other, on the level of refugee protection. Of great importance is the analysis of the multilevel structure of the legal framework regulating the asylum system, and the study of further parameters contributing to its formation, such as national security and state sovereignty, emerging nationalism and xenophobia, as well as the economic recession (2008-13). The expansion of migration flows from the Middle East, Asia and North Africa to Europe, in particular through Greece, with a milestone in 2015 and 2016, as well as the escalation of the number of potential refugees coming from countries which create issues of international protection, are at the heart of the problem. The management of the refugee crisis and the consequent political decision-making at both EU and international level has highlighted the prevalence of border security as a priority, with the introduction -at the same time- of geopolitical and international relations in the already complex issue of international protection. In conclusion, we would say that, in view of the conflict between the normative principles of international and EU legislation and the adopted pragmatist approach of the EU states, the rights of refugees seem to be relativised and their dimensions are weakening

    [Pemetrexed + Sorafenib] lethality is increased by inhibition of ERBB1/2/3-PI3K-NFκB compensatory survival signaling

    In the completed phase I trial NCT01450384 combining the anti-folate pemetrexed and the multi-kinase inhibitor sorafenib it was observed that 20 of 33 patients had prolonged stable disease or tumor regression, with one complete response and multiple partial responses. The pre-clinical studies in this manuscript were designed to determine whether [pemetrexed + sorafenib] –induced cell killing could be rationally enhanced by additional signaling modulators. Multiplex assays performed on tumor material that survived and re-grew after [pemetrexed + sorafenib] exposure showed increased phosphorylation of ERBB1 and of NFκB and IκB; with reduced IκB and elevated G-CSF and KC protein levels. Inhibition of JAK1/2 downstream of the G-CSF/KC receptors did not enhance [pemetrexed + sorafenib] lethality whereas inhibition of ERBB1/2/4 using kinase inhibitory agents or siRNA knock down of ERBB1/2/3 strongly promoted killing. Inhibition of ERBB1/2/4 blocked [pemetrexed + sorafenib] stimulated NFκB activation and SOD2 expression; and expression of IκB S32A S36A significantly enhanced [pemetrexed + sorafenib] lethality. Sorafenib inhibited HSP90 and HSP70 chaperone ATPase activities and reduced the interactions of chaperones with clients including c-MYC, CDC37 and MCL-1. In vivo, a 5 day transient exposure of established mammary tumors to lapatinib or vandetanib significantly enhanced the anti-tumor effect of [pemetrexed + sorafenib], without any apparent normal tissue toxicities. Identical data to that in breast cancer were obtained in NSCLC tumors using the ERBB1/2/4 inhibitor afatinib. Our data argue that the combination of pemetrexed, sorafenib and an ERBB1/2/4 inhibitor should be explored in a new phase I trial in solid tumor patients

    An RNA structure-mediated, posttranscriptional model of human α-1-antitrypsin expression

    Protein and mRNA expression are in most cases poorly correlated, which suggests that the posttranscriptional regulatory program of a cell is an important component of gene expression. This regulatory network is still poorly understood, including how RNA structure quantitatively contributes to translational control. We present here a series of structural and functional experiments that together allow us to derive a quantitative, structure-dependent model of translation that accurately predicts translation efficiency in reporter assays and primary human tissue for a complex and medically important protein, α-1-antitrypsin. Our model demonstrates the importance of accurate, experimentally derived RNA structural models partnered with Kozak sequence information to explain protein expression and suggests a strategy by which α-1-antitrypsin expression may be increased in diseased individuals
