Kiedy po wystawie w Muzeum Saturn, upamiętniającej losy czeladzkich żołnierzy walczących pod Monte Cassino w 70. rocznicę bitwy, opracowywaliśmy projekt niniejszego przedsięwzięcia, zależało nam również na
sprawdzeniu, czy, a jeśli tak, to w jakim stopniu wydarzenia sprzed tak wielu lat cokolwiek znaczą dla najmłodszych czeladzian. Dla ich dziadków, a nawet pradziadków były pokoleniowym przeżyciem; bezdyskusyjnym elementem składowym ich tożsamości, podobnie jak bezdyskusyjne było ich przywiązanie do kraju, miasta, rodziny — nakazujące im (w przeważającej większości) powroty po II wojnie światowej. Pathogenesis of non-alcoholic fatty liver disease
Non-alcoholic fatty liver disease (NAFLD) represents a spectrum of disease ranging from hepatocellular steatosis through steatohepatitis to fibrosis and irreversible cirrhosis. The prevalence of NAFLD has risen rapidly in parallel with the dramatic rise in obesity and diabetes, and is rapidly becoming the most common cause of liver disease in Western countries. Indeed, NAFLD is now recognized to be the aetiology in many cases previously labelled as cryptogenic cirrhosis
A Switch in Hepatic Cortisol Metabolism across the Spectrum of Non Alcoholic Fatty Liver Disease
Context: Non alcoholic fatty liver disease (NAFLD) is the hepatic manifestation of the metabolic syndrome. NAFLD represents a spectrum of liver disease ranging from reversible hepatic steatosis, to non alcoholic steato-hepatitis (NASH) and cirrhosis. The potential role of glucocorticoids (GC) in the pathogenesis of NAFLD is highlighted in patients with GC excess, Cushing's syndrome, who develop central adiposity, insulin resistance and in 20% of cases, NAFLD. Although in most cases of NAFLD, circulating cortisol levels are normal, hepatic cortisol availability is controlled by enzymes that regenerate cortisol (F) from inactive cortisone (E) (11β-hydroxysteroid dehydrogenase type 1, 11β-HSD1), or inactivate cortisol through A-ring metabolism (5α- and 5β-reductase, 5αR and 5βR). Objective and Methods: In vitro studies defined 11β-HSD1 expression in normal and NASH liver samples. We then characterised hepatic cortisol metabolism in 16 patients with histologically proven NAFLD compared to 32 obese controls using gas chromatographic analysis of 24 hour urine collection and plasma cortisol generation profile following oral cortisone. Results: In patients with steatosis 5αR activity was increased, with a decrease in hepatic 11β-HSD1 activity. Total cortisol metabolites were increased in this group consistent with increased GC production rate. In contrast, in patients with NASH, 11β-HSD1 activity was increased both in comparison to patients with steatosis, and controls. Endorsing these findings, 11β-HSD1 mRNA and immunostaining was markedly increased in NASH patients in peri septal hepatocytes and within CD68 positive macrophages within inflamed cirrhotic septa. Conclusion: Patients with hepatic steatosis have increased clearance and decreased hepatic regeneration of cortisol and we propose that this may represent a protective mechanism to decrease local GC availability to preserve hepatic metabolic phenotype. With progression to NASH, increased 11β-HSD1 activity and consequent cortisol regeneration may serve to limit hepatic inflammation
Possible pathological mechanisms in the development of non-alcoholic steatohepatitis (NASH)
Nicht-alkoholische Fettleber (NAFLD) und nicht-akoholische Steatohepatitis
(NASH) als deren progressive Form sind häufig auftretende Fehlfunktionen der
Leber. Es besteht ein starker Zusammenhang zum metabolischen Syndrom und vor
allem die NASH birgt ein erhöhtes Risiko für progressive Fibrose, Zirrhose,
ein Leberversagen oder die Entstehung eines hepatozellulären Karzinoms (HCC).
In Hinblick auf mögliche pathogenetische Faktoren der Fettleber- oder NASH-
Entstehung sind 73 Patienten mit histologisch nachgewiesener gesunder,
verfetteter oder NASH-Leber mittels Leberbiopsie, Blut- und Urinanalysen
untersucht worden. Anhand von real-time PCR wurde die mRNA-Expression der Gene
11β-Hydroxysteroid Dehydrogenase 1 (11β-HSD1), Hexose-6-Phosphat Dehydrogenase
(H6PDH), Hairy Enhancer of Split 1 (HES-1), Glukose-6-Phosphatase (G6Pase),
Glukose-6-Phosphat Transporter (G6PT) und 5α-Reduktase untersucht. Im 24h-Urin
der Studienpatienten sowie 60 gesunder Kontrollen wurden Kortisolmetaboliten
mittels Gas chromatographischer Massenspektrometrie (GC-MS) gemessen. Die 11β-
HSD1, die in der Leber Kortison zu Kortisol aktiviert, scheint keine Rolle in
der Entwicklung einer NAFLD oder in der Progression zur NASH zu spielen, zeigt
aber eine enge Beziehung zu H6PDH, seinem Substratliferanten. Die fehlende
Korrelation von 11β-HSD1 zur Fettleber steht im Gegensatz zu im Mäusemodell
erhobenen Daten [14]. Die hepatische 11β-HSD1 mRNA-Expression wurde bei
adipösen Patienten erniedrigt vorgefunden und stützt bereits vorhandene Daten,
dass adipöse Patienten oral eingenommenes Kortison schlechter aktivieren als
schlanke Individuen [48-50]. Die aus dem Quotienten THF+5α-THF/THE errechnete
Gesamtaktivität der 11β-HSD jedoch, zeigte keinen Zusammenhang mit Adipositas,
NAFLD oder NASH. Da dies aber die Gesamtaktivität beider 11β-HSD-Enzyme Typ 1
und 2 gleichzeitig aufzeigt, kann es nicht in direkten Zusammenhang mit einer
ausschließlichen 11β-HSD1-Aktivität gebracht werden. Die hepatische mRNA-
Expression von HES-1 steht in keiner Korrelation zum Leberfettgehalt, jedoch
aber zum Hüft-Taillen-Index, zu Cholesterin und zu LDL. Da die hier
untersuchten Fettlebern auch keinen Zusammenhang mit lokal erhöhten
Glukokortikoiden zu haben scheinen (mRNA-Expression von 11β-HSD1 in der Leber
war nicht erhöht), steht dies nicht im Gegensatz zu bereits bekannten Daten
aus Studien, die eine Fettleberentstehung durch Stimulation der hepatischen GR
mit konsekutiver HES-1 Minderexpression beschreiben [25]. Obwohl die
Urinanalyse hier untersuchter Patienten eine erhöhte 5α-Reduktaseaktivität bei
gesunden Männern, sowie bei weiblicher NAFLD und NASH aufzeigt, spiegeln sich
diese Funde nicht auf Transkriptionsebene in einer erhöhten hepatische mRNA-
Exprimierung von 5α-Reduktase in der Leber wieder. Eine Erklärung wäre eine
erhöhte 5α-Reduktase-Aktivität im extrahepatischen Gewebe. In Patienten mit
histologisch nachgewiesener NAFLD konnte eine signifikante Erniedrigung der
hepatischen mRNA-Expression von G6Pase und G6PT aufgezeigt werden. Dies könnte
sowohl Ursache als auch Grund für die Fettakkumulation in der Leber sein und
muss in weiteren Studien untersucht werden.Non-alcoholic fatty liver disease (NAFLD) and its progressive form non-
alcoholic steatohepatitis (NASH) are common liver disorders. They are strongly
associated with the metabolic syndrom. NASH especially bears the risk of
fibrosis, cirrhosis, end stage liver disease as well as development of a
hepatocellular carcinoma (HCC). In order to investigate possible pathogenetic
factors of development of NAFLD or NASH, liver biopsies, blood and urin
analysis from 73 patients with histologically proven healthy liver, NAFLD or
NASH have been evaluated. MRNA-Expressions of 11β-Hydroxysteroid Dehydrogenase
1 (11β-HSD1), Hexose-6-Phosphate Dehydrogenase (H6PDH), Hairy Enhancer of
Split 1 (HES-1), Glukose-6-Phosphatase (G6Pase), Glukose-6-Phosphate
Transporter (G6PT) und 5α-Reductase have been measured by means of real-time
PCR. Cortisol metabolites were measured via gas chromatography-mass
spektrometry (GC-MS) from patients' 24-hour urine collections. 11β-HSD1 which
activates cortisone to cortisol within the liver does not seem to play a role
in the development and progression of NAFLD and NASH, but shows a strong
interrelation with H6PDH. The missing correlation between 11β-HSD1 and the
development of NAFLD contradicts findings in mouse models [14]. Hepatic 11β-
HSD1 mRNA-expression was lower in patients with abdominal obesity and
therefore supports data showing decreased activation of oral cortisone in
those patients compared to lean controls [48-50]. The overall activity of 11β-
HSD1 calculated by the THF+5α-THF/THE ratio, however, did not show any
correlation with obesity, NAFLD or NASH. Hence this ratio reflects the overall
activity of 11β-HSD Type 1 and Type 2 simultaniously it cannot be used to
conclude on 11β-HSD1 activity only. The mRNA-expression of HES-1 showed no
correlation with liver fat content, but with hip-to-waist-ratio, cholesterol
and low-density lipoprotein (LDL) levels. Since there was no evidence for
locally elevated glucocorticoid levels (no increase in 11β-HSD1 mRNA-
expression) in patients with fatty liver there is no contradiction to previous
studies showing development of fatty liver due to stimulation of hepatic
glucocorticoid receptors with consecutive decrease in HES-1 mRNA-expression
[25]. Even though increased activity of 5α-reductase (urine analysis) was
shown in healthy men, female NAFLD and NASH there were no corresponding
results in the analysis of hepatic 5α-reductase mRNA-expression. Changes in
activity of extrahepatic 5α-reductase could be an explanation for this missing
link. There was a significant decrease in G6Pase and G6PT mRNA-Expression in
patients with NAFLD. This could be either cause for or result of fat
accumulation within the liver and must be investigated further in following
studies
The transfer of a humorous effect in wordplays in Polish audiovisual translation of an American animated television series The Penguins of Madagascar
Niniejsza praca analizuje mechanizmy gier językowych, które wywołują efekt komiczny w serialu animowanym Penguins of Madagascar/Pingwiny z Madagaskaru. Analiza obejmuje gry językowe na trzech płaszczyznach: leksykalnej, semantycznej oraz stylistycznej. W części teoretycznej przybliżona została specyfika przekładu audiowizualnego oraz definicje takich pojęć jak humor, komizm, gra językowa, elementy nacechowane kulturowo. Zrozumienie tych pojęć, których granice są często nieostre, było kluczowe dla dalszej analizy. Niniejsza praca nie bada ekwiwalencji formalnej czy też funkcjonalnej tekstu wyjściowego i docelowego, a raczej porównuje mechanizmy gier językowych, dzięki którym możliwe było uzyskanie efektu komicznego w dialogach angielskich i polskich.The thesis analyzes the mechanisms of the wordplays that evoke a humorous effect in the animated series Penguins of Madagascar/Pingwiny z Madagaskaru. The analysis encompasses wordplays on the three linguistic fields: lexical, semantic and stylistic. Specificity of the audiovisual translation as well as the definitions of terms like humor, the comic, wordplays and cultural specific items were described in the theoretical part of the work. A good understanding of these terms, the scope of which is often unclear, played a pivotal role in further analysis. The thesis does not analyze any equivalence, neither formal nor functional of the source and target texts. The aim of the thesis was rather to analyze the mechanisms of wordplays thanks to which it was possible to evoke a comic effect in English and Polish dialogues

Myślą przewodnią pracy magisterskiej jest wyjaśnienie koncepcji odpowiedzialności przebijającej, powszechnej w niektórych systemach prawnych. Celem rozważań zawartych w pracy jest przedstawienie odpowiedzialności przebijającej jako możliwego rozwiązania na negatywne konsekwencje zasady prawnej odrębności podmiotów tworzących grupę spółek oraz zasady wyłączonej odpowiedzialności wspólników zależnych spółek kapitałowych.W pierwszym rozdziale pracy, wyjaśnione zostały podstawowe zagadnienia związane z grupą spółek oraz planowane regulacje w tym zakresie na gruncie polskiego systemu prawnego. Kolejne rozdziały pracy poświęcone zostały przedstawieniu koncepcji odpowiedzialności przebijającej jako odpowiedź na istniejącą potrzebę wzmocnienia pozycji wierzycieli kapitałowych spółek zależnych. Celem ostatniego rozdziały pracy jest ocena projektu nowelizacji Kodeksu spółek handlowych w zakresie odpowiedzialności spółki dominującej w grupie spółek oraz rozważnie postulatów wprowadzenia koncepcji odpowiedzialności przebijającej, jako odrębnego przepisu prawa, do polskiego systemu prawnego.The main idea of the thesis is to explain the concept of piercing the corporate veil, which is widely used in some legal systems. The aim of the work is to present the issue of piercing the corporate veil as a possible solution to the negative consequences of the principle of separate legal personality and limited liability of the entities of the group of companies.The first chapter explains the basic concepts of the group of companies and the planned legislative changes related to this matter on the basis of the Polish legal system. The following chapters present the concept of piercing the corporate veil as a response to the need to empower the creditors of limited liability subsidiaries. The purpose of the last chapter is to consider the legislative changes in the Polish Commercial Code concerning the parent entity in the group of companies and the proposal to introduce the piercing of the corporate veil into the Polish legal system as a special provision
Blood pressure does not rise before the onset of microalbuminuria in children followed from diagnosis of type 1 diabetes. Oxford Regional Prospective Study Group.
OBJECTIVE: To examine whether a rise in blood pressure could be detected before the onset of microalbuminuria (MA) in a cohort of children followed from diagnosis of type 1 diabetes. RESEARCH DESIGN AND METHODS: The Oxford Regional Prospective Study is an incident cohort study of children with type 1 diabetes aged (mean +/- SD) 9.8 +/- 3.7 years at diagnosis. Subjects were assessed annually from diagnosis, with measurement of HbA1c, arterial blood pressure (random zero), and three urine samples for estimation of the albumin/creatinine ratio. During follow-up, 63 of 494 children developed MA at one or more annual assessments and were designated as cases for a nested case-control study. Each case was matched for sex and age at diagnosis with two normoalbuminuric control subjects. Blood pressure (BP) data were compared at corresponding years of diabetes duration. RESULTS: Cases with MA were similar to normoalbuminuric control subjects with respect to age and BMI, but they had higher mean HbA1c levels (mean difference 1.1%, P < 0.001). In the years before the onset of MA, the diastolic BP standard deviation score (SDS) was significantly higher than zero in cases (mean 0.49, P < 0.001) and in control subjects (0.50, P < 0.001). No difference could be detected between cases and control subjects before the onset of MA in either systolic or diastolic BP (mean difference systolic -1.2 mmHg [95% CI -4.7 to 2.7], mean difference diastolic 0.1 mmHg [-2.4 to 2.6]). However, within the cases, the onset of MA was associated with elevations in systolic and diastolic BP SDSs (F = 16.1, P < 0.001; and F = 18.0, P < 0.001). BMI, but not HbA1c, was associated with systolic and diastolic BP SDSs in the subjects with MA (F = 0.6, P = 0.4; and F = 12.3, P = 0.001). However, the association of BP with MA remained signifcant for systolic BP (P = 0.001) and for diastolic BP (P < 0.001) after adjusting for BMI. CONCLUSIONS: A rise in systemic BP cannot be detected before the first appearance of MA in children with type 1 diabetes. BP rises concurrently with the onset of MA and is also closely related to BMI
