Vigor, vitality and seed dormancy of Avena sativa cultivars in a long-term experiment
Vigor, vitality and seed dormancy of 14 Finnish cultivars of Avena sativa in room temperature were studied in a 22-year laboratory experiment. These parameters were studied by measuring morphological and physical characteristics of seeds and by basic germination and enzymatic tests 1, 4, 6, 11, 16, 21 and 22 years after seed harvesting in 1989. Methylene blue, Congo red and 2,3,5-triphenyltetrazolium chloride (TZ) tests were used to estimate seed quality and changes in vitality over time. Seed vitality clearly decreased in all cultivars during the experiment. The mean vitality declined from 96.3% (one year after harvest) to zero at the end. Vitality according to the TZ test was higher than indicated by the basic germination test. The mean vitality loss was 4.6% per year, but there were clear differences between cultivars. The decrease in vitality correlated with loss in seed weight. Clear signs of deepening dormancy were observed. Seed age is an important factor infl uencing vitality and dormancy. Vitality loss of seeds led to deep dormancy. The appearance, water uptake and imbibition of the seeds remained normal until the end. Ageing, vitality loss and dormancy are concluded to be expressions of genes. It is possible that in the future electronic simulation methods will be developed that will enable accurate estimation of oat seed quality without laboratory tests
Thermal roughening of an SOS-model with elastic interaction
We analyze the effects of a long-ranged step-step interaction on thermal
roughening within the framework of a solid-on-solid model of a crystal surface
by means of Monte Carlo simulation. A repulsive step-step interaction is
modeled by elastic dipoles located on sites adjacent to the steps. In order to
reduce the computational effort involved in calculating interaction energy
based on long-ranged potentials, we employ a multi-grid scheme. As a result of
the long-range character of the step interaction, the roughening temperature
increases drastically compared to a system with short-range cutoff as a
consequence of anti-correlations between surface defects

First Human Model of In Vitro Candida albicans Persistence within Granuloma for the Reliable Study of Host-Fungi Interactions
BACKGROUND: The balance between human innate immune system and Candida albicans virulence signaling mechanisms ultimately dictates the outcome of fungal invasiveness and its pathology. To better understand the pathophysiology and to identify fungal virulence-associated factors in the context of persistence in humans, complex models are indispensable. Although fungal virulence factors have been extensively studied in vitro and in vivo using different immune cell subsets and cell lines, it is unclear how C. albicans survives inside complex tissue granulomas. METHODOLOGY/PRINCIPAL FINDING: We developed an original model of in vitro human granuloma, reproducing the natural granulomatous response to C. albicans. Persistent granulomas were obtained when the ratio of phagocytes to fungi was high. This in vitro fungal granuloma mimics natural granulomas, with infected macrophages surrounded by helper and cytotoxic T lymphocytes. A small proportion of granulomas exhibited C. albicans hyphae. Histological and time-lapse analysis showed that C. albicans blastoconidia were located within the granulomas before hyphae formation. Using staining techniques, fungal load calculations, as well as confocal and scanning electron microscopy, we describe the kinetics of fungal granuloma formation. We provide the first direct evidence that C. albicans are not eliminated by immunocompetent cells inside in vitro human granulomas. In fact, after an initial candicidal period, the remaining yeast proliferate and persist under very complex immune responses. CONCLUSIONS/SIGNIFICANCE: Using an original in vitro model of human fungal granuloma, we herein present the evidence that C. albicans persist and grow into immunocompetent granulomatous structures. These results will guide us towards a better understanding of fungal invasiveness and, henceforth, will also help in the development of better strategies for its control in human physiological conditions
Neodymium isotope constraints on provenance, dispersal, and climate-driven supply of Zambezi sediments along the Mozambique Margin during the past ∼45,000 years
Marine sediments deposited off the Zambezi River that drains a considerable part of the southeast African continent provide continuous records of the continental climatic and environmental conditions.
Here we present time series of neodymium (Nd) isotope signatures of the detrital sediment fraction during the past ~45,000 years, to reconstruct climate-driven changes in the provenance of clays deposited along the Mozambique Margin. Coherent with the surface current regime, the Nd isotope distribution in surface sediments reveals mixing of the alongshore flowing Zambezi suspension load with sediments supplied by smaller rivers located further north. To reconstruct past changes in sediment provenances, Nd isotope signatures
of clays that are not significantly fractionated during weathering processes have been obtained from core 64PE304-80, which was recovered just north of the Zambezi mouth at 1329 m water depth. Distinctly unradiogenic clay signatures (ENd values <214.2) are found during the Last Glacial Maximum, Heinrich Stadial 1, and Younger Dryas. In contrast, the Nd isotope record shows higher, more radiogenic isotope signatures during Marine Isotope Stage 3 and between ~15 and ~5 ka BP, the latter coinciding with the timing of the northern hemisphere African Humid Period. The clay-sized sediment fraction with the least radiogenic Nd isotope signatures was deposited during the Holocene, when the adjacent Mozambique Shelf became completely flooded. In general, the contribution of the distinctly unradiogenic Zambezi suspension load has followed the intensity of precession-forced monsoonal precipitation and enhanced during periods of increased southern hemisphere insolation and high-latitude northern hemispheric climate
