    High resolution physical mapping of single gene fragments on pachytene chromosome 4 and 7 of Rosa

    Background: Rosaceae is a family containing many economically important fruit and ornamental species. Although fluorescence in situ hybridization (FISH)-based physical mapping of plant genomes is a valuable tool for map-based cloning, comparative genomics and evolutionary studies, no studies using high resolution physical mapping have been performed in this family. Previously we proved that physical mapping of single-copy genes as small as 1.1 kb is possible on mitotic metaphase chromosomes of Rosa wichurana using Tyramide-FISH. In this study we aimed to further improve the physical map of Rosa wichurana by applying high resolution FISH to pachytene chromosomes. Results: Using high resolution Tyramide-FISH and multicolor Tyramide-FISH, 7 genes (1.7-3 kb) were successfully mapped on pachytene chromosomes 4 and 7 of Rosa wichurana. Additionally, by using multicolor Tyramide-FISH three closely located genes were simultaneously visualized on chromosome 7. A detailed map of heterochromatine/euchromatine patterns of chromosome 4 and 7 was developed with indication of the physical position of these 7 genes. Comparison of the gene order between Rosa wichurana and Fragaria vesca revealed a poor collinearity for chromosome 7, but a perfect collinearity for chromosome 4. Conclusions: High resolution physical mapping of short probes on pachytene chromosomes of Rosa wichurana was successfully performed for the first time. Application of Tyramide-FISH on pachytene chromosomes allowed the mapping resolution to be increased up to 20 times compared to mitotic metaphase chromosomes. High resolution Tyramide-FISH and multicolor Tyramide-FISH might become useful tools for further physical mapping of single-copy genes and for the integration of physical and genetic maps of Rosa wichurana and other members of the Rosaceae

    Towards a FISH-based karyotype of Rosa L. (Rosaceae)

    The genus Rosa Linnaeus, 1753 has important economic value in ornamental sector and many breeding activities are going on supported by molecular studies. However, the cytogenetic studies of rose specks are scarce and mainly focused on chromosome counting and chromosome morphology-based karyotyping. Due to the small size of the chromosomes and a high frequency of polyploidy in the genus, karyotyping is very challenging for rose species and requires FISH-based cytogenetic markers to be applied. Therefore, in this work the aim is to establish a FISH-based karyotype for Rosa wichurana (Crepin, 1888), a rose species with several benefits for advanced molecular cytogenetic studies of genus Rosa (Kirov et al. 2015a). It is shown that FISH signals from 5S, 45S and an Arabidopsis-type telomeric repeat are distributed on five (1, 2, 4, 5 and 7) of seven chromosome pairs. In addition, it is demonstrated that the interstitial telomeric repeat sequences (ITR) are located in the centromeric regions of four chromosome pairs. Using low hybridization stringency for ITR visualization, we showed that the number of ITR signals increases four times (1-4 signals). This study is the first to propose a FISH-based R. wichurana katyotype for the reliable identification of chromosomes. The possible origin of R wichurana ITR loci is discussed

    Percutaneous management of acute necrotizing pancreatitis

    OBJECTIVES: The study aims to evaluate the efficacy of percutaneous necrosectomy (PN) performed under ultrasound control and of endoscopic necrosectomy through secondary sinus track (ENTSST) using nephroscope and cystoscope.MATERIAL AND METHOD: Puncture of fluid collections in the pancreas was performed under ultrasonographic control to 23 patients with acute necrotizing pancreatitis (ANP). ENTSST using nephroscope and cystoscope was performed to 47 patients after open or percutaneous necrosectomy and persistent sepsis (without satellite collection of CT).RESULTS: Seventeen patients (74%) treated with percutaneous necrosectomy recovered without open surgery. Two of this group died. The average hospital stay was 42 days. Twenty-three patients required an average of two (range 1-4) ENTSST.CONCLUSIONS: Based on our initial results we believe that the percutaneous necrosectomy and ENTSST in well selected patients might be the better choice than open necrosectomy and postoperative lavage. Common solution for these methods has not been reached yet

    Comparison of Goal-Directed Hemodynamic Optimization Using Pulmonary Artery Catheter and Transpulmonary Thermodilution in Combined Valve Repair: A Randomized Clinical Trial

    Our aim was to compare the effects of goal-directed therapy guided either by pulmonary artery catheter (PAC) or by transpulmonary thermodilution (TTD) combined with monitoring of oxygen transport on perioperative hemodynamics and outcome after complex elective valve surgery. Measurements and Main Results. Forty patients were randomized into two equal groups: a PAC group and a TTD group. In the PAC group, therapy was guided by mean arterial pressure (MAP), cardiac index (CI) and pulmonary artery occlusion pressure (PAOP), whereas in the TTD group we additionally used global end-diastolic volume index (GEDVI), extravascular lung water index (EVLWI), and oxygen delivery index (DO2I). We observed a gradual increase in GEDVI, whereas EVLWI and PAOP decreased by 20–30% postoperatively (P < 0.05). The TTD group received 20% more fluid accompanied by increased stroke volume index and DO2I by 15–20% compared to the PAC group (P < 0.05). Duration of mechanical ventilation was increased by 5.2 hrs in the PAC group (P = 0.04). Conclusions. As compared to the PAC-guided algorithm, goal-directed therapy based on transpulmonary thermodilution and oxygen transport increases the volume of fluid therapy, improves hemodynamics and DO2I, and reduces the duration of respiratory support after complex valve surgery

    Эмоциональное состояние врачей – анестезиологов-реаниматологов в разные периоды пандемии COVID-19

    Objective: identifcation of the peculiarities of the emotional states of anesthesiologists and intensivists in different periods of the COVID-19 Pandemic.Material and Methods. The study was conducted using an anonymous questionnaire survey in the frst and second waves of the pandemic COVID-19 among anesthesiologists and intensivists of the Arkhangelsk region: in May 2020 (in the frst wave of the COVID-19 pandemic), 58 doctors participated; the average age was 32.7 ± 1.7 years (M ± SD); in October 2020 (during the second wave of the COVID-19 pandemic), 43 doctors were involved; the average age was 28.0 ± 1.2 years (M ± SD). The repeated questionnaire survey was conducted among the same doctors.Results. During the second wave of COVID-19, male doctors more often noted poor health, signifcant heaviness and intensity of their work versus the frst wave, while women on the contrary during the frst wave had a poorer emotional state and higher level of emotional burnout. During the second wave of COVID-19, anesthesiologists and intensivists involved in caring for patients with COVID-19 were more likely to experience poor emotional state, anxiety, low mood, irritability, and a high level of burnout compared to the frst wave.Summary. The second wave of COVID-19 is accompanied by further exhaustion of emotional resources of anesthesiologists and intensivists.Цель: выявление особенностей эмоционального состояния врачей ‒ анестезиологов-реаниматологов в разные периоды пандемии COVID-19.Материал и методы. Исследование проводилось в форме анонимного анкетирования во время первой и второй волн пандемии среди врачей ‒ анестезиологов-реаниматологов Архангельской области: в мае 2020 г. (в первую волну пандемии COVID-19) приняли участие 58 врачей, средний возраст 32,7 ± 1,7 года (M ± SD); в октябре 2020 г. (во время второй волны пандемии COVID-19) опрошено 43 врача, средний возраст 28,0 ± 1,2 года (M ± SD).  Повторное анкетирование проводили среди тех же врачей.Результаты. Во время второй волны COVID-19 врачи-мужчины чаще отмечали у себя плохое самочувствие, высокую степень тяжести и напряженности работы, а у женщин, наоборот, во время первой волны отмечались более плохое эмоциональное состояние, более высокий уровень эмоционального «выгорания». Врачи ‒ анестезиологи-реаниматологи, участвующие в оказании помощи пациентам с COVID-19, во время второй волны COVID-19 чаще отмечали у себя плохое эмоциональное состояние, тревогу, сниженное настроение, раздражительность и высокий уровень «выгорания» по сравнению с первой волной.Заключение. Вторая волна COVID-19 сопровождается дальнейшим истощением эмоциональных ресурсов анестезиологов-реаниматологов
