    Mesoarchean partial melting of mafic crust and tonalite production during high-T–low-P stagnant tectonism, Akia Terrane, West Greenland

NJG acknowledges support from Australian Research Council grant FL160100168.Different geodynamic models exist for the growth and differentiation of Archean continental crust, ranging from horizontal tectonics with subduction zones to vertical tectonics with foundering of greenstone sequences. U–Pb zircon geochronology, field relationships, and pressure–temperature constraints from granulite-facies metabasite of the Akia Terrane of the North Atlantic Craton in West Greenland show that this terrane grew through two major magmatic growth episodes: an earlier one at c. 3.2 Ga, and a later one at c. 3.05–2.97 Ga. Phase equilibrium modelling for assemblages related to the latter indicates temperatures of >800 °C at 0.8 GPa in the stability field of garnet. U–Pb zircon geochronology and existing Hf isotope data are also consistent with a model involving protracted Mesoarchean magmatic growth with limited mantle addition during a prolonged period of high temperatures in a relatively stagnant tectonic regime prior to Neoarchean compressional tectonism in the Akia Terrane.Publisher PDFPeer reviewe

    Evaluating Brightness and Spectral Properties of Click Beetle and Firefly Luciferases Using Luciferin Analogues: Identification of Preferred Pairings of Luciferase and Substrate for In Vivo Bioluminescence Imaging.

    Currently, a variety of red and green beetle luciferase variants are available for bioluminescence imaging (BLI). In addition, new luciferin analogues providing longer wavelength luminescence have been developed that show promise for improved deep tissue imaging. However, a detailed assessment of these analogues (e.g., Akalumine-HCl, CycLuc1, and amino naphthyl luciferin (NH <sub>2</sub> -NpLH2)) combined with state of the art luciferases has not been performed. The aim of this study was to evaluate for the first time the in vivo brightness and spectral characteristics of firefly (Luc2), click beetle green (CBG99), click beetle red 2 (CBR2), and Akaluc luciferases when paired with different D-luciferin (D-LH2) analogues in vivo. Transduced human embryonic kidney (HEK 293T) cells expressing individual luciferases were analyzed both in vitro and in mice (via subcutaneous injection). Following introduction of the luciferins to cells or animals, the resulting bioluminescence signal and photon emission spectrum were acquired using a sensitive charge-coupled device (CCD) camera equipped with a series of band pass filters and spectral unmixing software. Our in vivo analysis resulted in four primary findings: (1) the best substrate for Luc2, CBG99, and CBR2 in terms of signal strength was D-luciferin; (2) the spectra for Luc2 and CBR2 were shifted to a longer wavelength when Akalumine-HCl was the substrate; (3) CBR2 gave the brightest signal with the near-infrared substrate, NH <sub>2</sub> -NpLH2; and (4) Akaluc was brighter when paired with either CycLuc1 or Akalumine-HCl when paired with D-LH2. We believe that the experimental results described here should provide valuable guidance to end users for choosing the correct luciferin/luciferase pairs for a variety of BLI applications

    Learning Form and Function by Dance-Dramatizing Cultural Legends to Drum Rhythms Wearing Student-Made Animal Masks

    This study examined the self-efficacy in science, art, dance, and music; attitudes concerning contributions of people of various ethnic/cultural groups; and science learning of students involved in an after-school arts-integrated science enrichment project. Students dramatized three traditional animal legends from African, Native American, and Mexican cultures to drum beats while wearing student-made papier-mâché helmet crest masks of the animal characters. They learned the structure and functions of the featured animals through slide shows, embedded explanations in the play scripts, and hands-on form and function analogy materials that related the form and function of animal body parts to manufactured items. Although at least 40 students participated at times in the after-school program, matching pretest and posttest data were only available for 13 students. Results showed positive changes in students’ art self-efficacy with a medium effect size, improvements in knowledge of animal form and function with a large effect size, and a trend toward greater appreciation of the cultural contributions of different ethnic groups.     First comparison of French and Australian OsHV-1 µvars by bath exposure

    Economically devastating mortality events of farmed and wild shellfish due to infectious disease have been reported globally. Currently, one of the most significant disease threats to Pacific oyster Crassostrea gigas culture is the ostreid herpesvirus 1 (OsHV-1), in particular the emerging OsHV-1 microvariant genotypes. OsHV-1 microvariants (OsHV-1 μvars) are spreading globally, and concern is high among growers in areas unaffected by OsHV-1. No study to date has compared the relative virulence among variants. We provide the first challenge study comparing survival of naïve juvenile Pacific oysters exposed to OsHV-1 μvars from Australia (AUS μvar) and France (FRA μvar). Oysters challenged with OsHV-1 μvars had low survival (2.5% exposed toAUS μvar and 10% to FRA μvar), and high viral copy number as compared to control oysters(100% survival and no virus detected). As our study was conducted in a quarantine facility located~320 km from the ocean, we also compared the virulence of OsHV-1 μvars using artificial seawater made from either facility tap water (3782 μmol kg−1seawater total alkalinity) or purchased distilled water (2003 μmol kg−1). Although no differences in survival or viral copy number were detected in oysters exposed to seawater made using tap or distilled water, more OsHV-1 was detected in tanks containing the lower-alkalinity seawater, indicating that water quality may be important for virus transmission, as it may influence the duration of viral viability outside of the hos

    The source of Dalradian detritus in the Buchan Block, NE Scotland: Application of new tools to detrital datasets

    Detrital zircons from four samples of upper Dalradian metasedimentary rocks from the Buchan Block in the NE Grampian Highlands of Scotland were analysed by laser ablation inductively coupled plasma mass spectrometry to establish their U–Pb age and trace element composition. The analysed grains (magmatic cores) mainly yield concordant ages ranging from Neoproterozoic to Eoarchaean. Kernel density plots of the data show pronounced peaks in the late Mesoproterozoic, Palaeoproterozoic and Neoarchaean eras. The data are indistinguishable from detrital zircon age spectra from Dalradian rocks elsewhere, an interpretation supported by application of a non-parametric multidimensional scaling algorithm, and are consistent with a Laurentian source. Similar to existing studies from other Dalradian rocks, the age spectra from the Buchan Block reveal an increase in the relative proportion of older detritus with time, suggesting derivation from late Mesoproterozoic (Grenville) then Palaeoproterozoic orogens before widespread exposure and denudation of their Archaean basement rocks. Application of a novel approach to estimate the most likely time of radiogenic-Pb loss indicates that some detrital zircon grains were affected by element mobility around 470–450 Ma as a result of Grampian orogenesis

    NanoBiT System and Hydrofurimazine for Optimized Detection of Viral Infection in Mice-A Novel in Vivo Imaging Platform.

    Reporter genes are used to visualize intracellular biological phenomena, including viral infection. Here we demonstrate bioluminescent imaging of viral infection using the NanoBiT system in combination with intraperitoneal injection of a furimazine analogue, hydrofurimazine. This recently developed substrate has enhanced aqueous solubility allowing delivery of higher doses for in vivo imaging. The small high-affinity peptide tag (HiBiT), which is only 11 amino-acids in length, was engineered into a clinically used oncolytic adenovirus, and the complementary large protein (LgBiT) was constitutively expressed in tumor cells. Infection of the LgBiT expressing cells with the HiBiT oncolytic virus will reconstitute NanoLuc in the cytosol of the cell, providing strong bioluminescence upon treatment with substrate. This new bioluminescent system served as an early stage quantitative viral transduction reporter in vitro and also in vivo in mice, for longitudinal monitoring of oncolytic viral persistence in infected tumor cells. This platform provides novel opportunities for studying the biology of viruses in animal models

    Building Mesoarchaean crust upon Eoarchaean roots : the Akia Terrane, West Greenland

NJG thanks Curtin University and Australian Research Council grant FL160100168 for financial support.Constraining the source, genesis, and evolution of Archaean felsic crust is key to understanding the growth and stabilization of cratons. The Akia Terrane, part of the North Atlantic Craton, West Greenland, is comprised of Meso-to-Neoarchaean orthogneiss, with associated supracrustal rocks. We report zircon U–Pb and Lu–Hf isotope data, and whole-rock geochemistry, from samples of gneiss and supracrustals from the northern Akia Terrane, including from the Finnefjeld Orthogneiss Complex, which has recently been interpreted as an impact structure. Isotope data record two major episodes of continental crust production at ca. 3.2 and 3.0 Ga. Minor ca. 2.7 and 2.5 Ga magmatic events have more evolved εHf, interpreted as reworking of existing crust perhaps linked to terrane assembly. Felsic rocks from the Finnefjeld Orthogneiss Complex were derived from the same source at the same time as the surrounding tonalites, but from shallower melting, requiring any bolide-driven melting event to have occurred almost simultaneously alongside the production of the surrounding crust. A simpler alternative has the Finnefjeld Complex and surrounding tonalite representing the coeval genesis of evolved crust over a substantial lithospheric depth. Hafnium isotope data from the two major Mesoarchaean crust-forming episodes record a contribution from older mafic Eoarchaean crust. Invoking the involvement of an Eoarchaean root in the growth of younger Mesoarchaean crust puts important constraints on geodynamic models of the formation of the discrete terranes that ultimately assembled to form Earth’s cratons.Publisher PDFPeer reviewe

    U–Pb zircon-rutile dating of the Llangynog Inlier, Wales: constraints on an Ediacaran shallow marine fossil assemblage from East Avalonia

    The Llangynog Inlier of south Wales contains an assemblage of Ediacaran macrofossils from a shallow-marine environment, including discoidal morphs of Aspidella and rare examples of Hiemalora, Palaeopascichnus and Yelovichnus. These are taxa found in other sites in the Avalonian microcontinent (e.g. Charnwood Forest and eastern Newfoundland) and in the younger White Sea Ediacaran assemblages. As the Charnwood fossils reflect a deep-water environment, and no macrofossils have been found in the Ediacaran rocks of the Long Mynd, the fossils of the Llangynog Inlier represent a unique glimpse of shallow marine life in southern Britain (East Avalonia). However, the lack of absolute age constraints has hampered direct comparison with other assemblages. Here, we report in-situ zircon and rutile U–Pb dates from a rhyolitic ash-flow layer of the Coed Cochion Volcaniclastic Member, Llangynog Inlier, which constrains the age of the fossiliferous strata. A weighted mean single grain zircon ID-TIMS U–Pb age of 564.09 ± 0.70 Ma is interpreted as the rhyolite's crystallisation age. This age is consistent with in-situ LA-ICPMS zircon and rutile U–Pb dating. The Llangynog age temporally correlates these fossils to dated horizons within East Avalonia at the Beacon Hill Formation, Charnwood (565.22 ± 0.89 Ma), and the Stretton Shale Formation, Long Mynd (566.6 ± 2.9 Ma). Correlations to West Avalonia include the time-equivalent Fermeuse Formation, St John’s Group, eastern Newfoundland (564.13 ± 0.65 Ma). The data presented here establish the biota of the Llangynog Inlier as a lateral equivalent to the similarly shallow marine, tidally influenced ecosystem of the upper Fermeuse Formation. Intra-terrane depositional environmental variability also affects what is preserved in Avalonian fossil sites. Further, time-constrained geochemical data reinforce the Llangynog Inlier's classification within the Wrekin Terrane
