    In Favor of ā€œLeader Proofingā€

    Get PDF
Although it is widely assumed that successful polities require strong leaders, something like the opposite is probably the case. A successful political system may well be one that has no need of strong leaders and may even eschew them. Strong leaders may occasionally be desirable in any polity, but those occasions areā€“or should beā€“rare. As often as not (possibly more often than not) strong leaders pose substantial risks. They are liable to do as much damage as good, possibly more. There is a lot to be said for any polity's political culture and institutions having built into them a fair amount of ā€œleader proofing.ā€ </jats:p

The Future of Cell Biology: Emerging Model Organisms

    Get PDF
Most current research in cell biology uses just a handful of model systems including yeast, Arabidopsis, Drosophila, Caenorhabditis elegans, zebrafish, mouse, and cultured mammalian cells. And for good reason - for many biological questions, the best system for the question is likely to be found among these models. However, in some cases, and particularly as the questions that engage scientists broaden, the best system for a question may be a little-studied organism. Modern research tools are facilitating a renaissance for unusual and interesting organisms as emerging model systems. As a result, we predict that an ever-expanding breadth of model systems may be a hallmark of future cell biology

Design and Construction of Compaction Grouting for Foundation Soil Improvements

    Get PDF
This paper presents the design and construction of compaction grouting work completed for a tank replacement project in Portland, Oregon. The project site is located along the west bank of the Willamette River. The subsurface soils at the project site were determined to be highly susceptible to soil liquefaction and lateral spreading under a design earthquake event per the building code. Compaction grouting was designed and constructed to strengthen the foundation soils supporting the new steel tank that is 115 feet in diameter and 40 feet in height. The design of the compaction grouting was completed using the design guidelines outlined in ASCE/G-I Standard 53-10. Detailed quality assurance/quality control processes were implemented during grouting operations to account for the variability in soil conditions being grouted. Real time monitoring was also completed to evaluate the ground movement induced by the grouting process and its impact to adjacent structures and critical utilities. Pre- and post-grouting CPTs were completed to verify that the intended ground densification was achieved. A hydrostatic test was also completed with the tank filled with water. The tank foundation settlement under the hydrostatic test was found to range between Ā¼ to Ā¾ inches and met the acceptance criteria per API-650 and API-653 Standards

The Lion, the Witch, and the Wormhole: Ensemble averaging the symmetric product orbifold

We consider the ensemble average of two dimensional symmetric product orbifold CFTs SymN(TD)\text{Sym}^N(\mathbb{T}^D) over the Narain moduli space. We argue for a bulk dual given by NN copies of an abelian Chern-Simons theory coupled to topological gravity, endowed with a discrete gauge symmetry exchanging the NN copies. As a check of this proposal, we calculate the ensemble average of various partition and correlation functions of the symmetric product orbifold theory and compare the resulting expressions to gauge theory quantities in the bulk. We comment on the ensemble average of the tensionless string partition function on AdS3ƗS3ƗT4\text{AdS}_3 \times \text{S}^3 \times \mathbb T^4 by considering the specific case of D=4D=4 with the addition of supersymmetry.Comment: 84 pages, 25 figure

Common mental disorders and ethnicity in England : the EMPIRIC Study

    Get PDF
Background. There is little population-based evidence on ethnic variation in the most common mental disorders (CMD), anxiety and depression. We compared the prevalence of CMD among representative samples of White, Irish, Black Caribbean, Bangladeshi, Indian and Pakistani individuals living in England using a standardized clinical interview. Method. Cross-sectional survey of 4281 adults aged 16ā€“74 years living in private households in England. CMD were assessed using the Revised Clinical Interview Schedule (CIS-R), a standardized clinical interview. Results. Ethnic differences in the prevalence of CMD were modest, and some variation with age and sex was noted. Compared to White counterparts, the prevalence of CMD was higher to a statistically significant degree among Irish [adjusted rate ratios (RR) 2.09, 95% CI 1.16ā€“2.95, p=0.02] and Pakistani (adjusted RR 2.38, 95% CI 1.25ā€“3.53, p=0.02) men aged 35ā€“54 years, even after adjusting for differences in socio-economic status. Higher rates of CMD were also observed among Indian and Pakistani women aged 55ā€“74 years, compared to White women of similar age. The prevalence of CMD among Bangladeshi women was lower than among White women, although this was restricted to those not interviewed in English. There were no differences in rates between Black Caribbean and White samples. Conclusions. Middle-aged Irish and Pakistani men, and older Indian and Pakistani women, had significantly higher rates of CMD than their White counterparts. The very low prevalence of CMD among Bangladeshi women contrasted with high levels of socio-economic deprivation among this group. Further study is needed to explore reasons for this variation

Improving formaldehyde consumption drives methanol assimilation in engineered E. coli

    Get PDF
Due to volatile sugar prices, the food vs fuel debate, and recent increases in the supply of natural gas, methanol has emerged as a promising feedstock for the bio-based economy. However, attempts to engineer Escherichia coli to metabolize methanol have achieved limited success. Here, we provide a rigorous systematic analysis of several potential pathway bottlenecks. We show that regeneration of ribulose 5-phosphate in E. coli is insufficient to sustain methanol assimilation, and overcome this by activating the sedoheptulose bisphosphatase variant of the ribulose monophosphate pathway. By leveraging the kinetic isotope effect associated with deuterated methanol as a chemical probe, we further demonstrate that under these conditions overall pathway flux is kinetically limited by methanol dehydrogenase. Finally, we identify NADH as a potent kinetic inhibitor of this enzyme. These results provide direction for future engineering strategies to improve methanol utilization, and underscore the value of chemical biology methodologies in metabolic engineering
