    Meiotic nuclear divisions in budding yeast require PP2ACdc55-mediated antagonism of Net1 phosphorylation by Cdk

    During meiosis, one round of deoxyribonucleic acid replication is followed by two rounds of nuclear division. In Saccharomyces cerevisiae, activation of the Cdc14 early anaphase release (FEAR) network is required for exit from meiosis I but does not lead to the activation of origins of replication. The precise mechanism of how FEAR regulates meiosis is not understood. In this paper, we report that premature activation of FEAR during meiosis caused by loss of protein phosphatase PP2ACdc55 activity blocks bipolar spindle assembly and nuclear divisions. In cdc55 meiotic null (cdc55-mn) cells, the cyclin-dependent kinase (Cdk)–counteracting phosphatase Cdc14 was released prematurely from the nucleolus concomitant with hyperphosphorylation of its nucleolar anchor protein Net1. Crucially, a mutant form of Net1 that lacks six Cdk phosphorylation sites rescued the meiotic defect of cdc55-mn cells. Expression of a dominant mutant allele of CDC14 mimicked the cdc55-mn phenotype. We propose that phosphoregulation of Net1 by PP2ACdc55 is essential for preventing precocious exit from meiosis I

    Stability and function of adult vasculature is sustained by Akt/Jagged1 signalling axis in endothelium.

    The signalling pathways operational in quiescent, post-development vasculature remain enigmatic. Here we show that unlike neovascularization, endothelial Akt signalling in established vasculature is crucial not for endothelial cell (EC) survival, but for sustained interactions with pericytes and vascular smooth muscle cells (VSMCs) regulating vascular stability and function. Inducible endothelial-specific Akt1 deletion in adult global Akt2KO mice triggers progressive VSMC apoptosis. In hearts, this causes a loss of arteries and arterioles and, despite a high capillary density, diminished vascular patency and severe cardiac dysfunction. Similarly, endothelial Akt deletion induces retinal VSMC loss and basement membrane deterioration resulting in vascular regression and retinal atrophy. Mechanistically, the Akt/mTOR axis controls endothelial Jagged1 expression and, thereby, Notch signalling regulating VSMC maintenance. Jagged1 peptide treatment of Akt1ΔEC;Akt2KO mice and Jagged1 re-expression in Akt-deficient endothelium restores VSMC coverage. Thus, sustained endothelial Akt1/2 signalling is critical in maintaining vascular stability and homeostasis, thereby preserving tissue and organ function

    Finite time singularities in a class of hydrodynamic models

    Models of inviscid incompressible fluid are considered, with the kinetic energy (i.e., the Lagrangian functional) taking the form L∌∫kα∣vk∣2d3k{\cal L}\sim\int k^\alpha|{\bf v_k}|^2d^3{\bf k} in 3D Fourier representation, where α\alpha is a constant, 0<α<10<\alpha< 1. Unlike the case α=0\alpha=0 (the usual Eulerian hydrodynamics), a finite value of α\alpha results in a finite energy for a singular, frozen-in vortex filament. This property allows us to study the dynamics of such filaments without the necessity of a regularization procedure for short length scales. The linear analysis of small symmetrical deviations from a stationary solution is performed for a pair of anti-parallel vortex filaments and an analog of the Crow instability is found at small wave-numbers. A local approximate Hamiltonian is obtained for the nonlinear long-scale dynamics of this system. Self-similar solutions of the corresponding equations are found analytically. They describe the formation of a finite time singularity, with all length scales decreasing like (t∗−t)1/(2−α)(t^*-t)^{1/(2-\alpha)}, where t∗t^* is the singularity time.Comment: LaTeX, 17 pages, 3 eps figures.     An updated radiocarbon-based ice margin chronology for the last deglaciation of the North American Ice Sheet Complex

    The North American Ice Sheet Complex (NAISC; consisting of the Laurentide, Cordilleran and Innuitian ice sheets) was the largest ice mass to repeatedly grow and decay in the Northern Hemisphere during the Quaternary. Understanding its pattern of retreat following the Last Glacial Maximum is critical for studying many facets of the Late Quaternary, including ice sheet behaviour, the evolution of Holocene landscapes, sea level, atmospheric circulation, and the peopling of the Americas. Currently, the most up-to-date and authoritative margin chronology for the entire ice sheet complex is featured in two publications (Geological Survey of Canada Open File 1574 [Dyke et al., 2003]; ‘Quaternary Glaciations – Extent and Chronology, Part II’ [Dyke, 2004]). These often-cited datasets track ice margin recession in 36 time slices spanning 18 ka to 1 ka (all ages in uncalibrated radiocarbon years) using a combination of geomorphology, stratigraphy and radiocarbon dating. However, by virtue of being over 15 years old, the ice margin chronology requires updating to reflect new work and important revisions. This paper updates the aforementioned 36 ice margin maps to reflect new data from regional studies. We also update the original radiocarbon dataset from the 2003/2004 papers with 1541 new ages to reflect work up to and including 2018. A major revision is made to the 18 ka ice margin, where Banks and Eglinton islands (once considered to be glacial refugia) are now shown to be fully glaciated. Our updated 18 ka ice sheet increased in areal extent from 17.81 to 18.37 million km2, which is an increase of 3.1% in spatial coverage of the NAISC at that time. Elsewhere, we also summarize, region-by-region, significant changes to the deglaciation sequence. This paper integrates new information provided by regional experts and radiocarbon data into the deglaciation sequence while maintaining consistency with the original ice margin positions of Dyke et al. (2003) and Dyke (2004) where new information is lacking; this is a pragmatic solution to satisfy the needs of a Quaternary research community that requires up-to-date knowledge of the pattern of ice margin recession of what was once the world’s largest ice mass. The 36 updated isochrones are available in PDF and shapefile format, together with a spreadsheet of the expanded radiocarbon dataset (n = 5195 ages) and estimates of uncertainty for each interval

    Simulação do rendimento de grãos de arroz irrigado em cenårios de mudanças climåticas

    O objetivo deste trabalho foi simular o rendimento de grĂŁos de arroz em cenĂĄrios de mudanças climĂĄticas com aumento da atual concentração de CO2 na atmosfera e da temperatura mĂ©dia do ar em Santa Maria, RS, e verificar as implicaçÔes nas recomendaçÔes de Ă©poca de semeadura. Foram utilizados cenĂĄrios de mudanças climĂĄticas, para os prĂłximos 100 anos, com o dobro da quantidade de CO2 e com aumentos de temperatura do ar de 1, 2, 3, 4 e 5°C. O rendimento de grĂŁos da cultura do arroz foi simulado com o modelo InfoCrop. As simulaçÔes foram realizadas para trĂȘs cultivares de arroz (IRGA 421, IRGA 417 e EPAGRI 109) em sete datas de semeadura, com intervalos mensais, de 20 de julho atĂ© 20 de janeiro. Um aumento no rendimento de grĂŁos de arroz irrigado foi observado nos cenĂĄrios de mudanças climĂĄticas simulado para as trĂȘs cultivares, com maior incremento na muito precoce (IRGA 421) e menor na de ciclo longo (EPAGRI 109).
