    On the hygrothermomechanical characterization of polyvinyl acetate

    As a part of a program to understand the mechanisms of failure in time-dependent adhesion and film bonding, the creep compliance of polyvinyl acetate (PVAc) in shear has been determined both as a function of temperature and absorbed moisture. Volumetric expansion as a function of temperature or moisture takeup was also measured. We find that practically realizable changes in moisture content affect both the creep compliance and the swelling of PVAc to a degree comparable to that resulting from realistic changes in temperature. For example, the creep rates (histories) at corresponding times for PVAc subjected to 92% relative humidity storage are accelerated by approximately four orders of magnitude over those found for the dry material. Moreover, we find within reasonable experimental error that water concentration affects the time scale of creep like temperature through a concentration-dependent shift factor.     Long Term Displacement Data of Woven Fabric Webbings Under Constant Load for Inflatable Structures

    Inflatable modules for space applications offer weight and launch volume savings relative to current metallic modules. Limited data exist on the creep behavior of the restraint layer of inflatable modules. Long-term displacement and strain data of two high strength woven fabric webbings, Kevlar and Vectran, under constant load is presented. The creep behavior of webbings is required by designers to help determine service life parameters of inflatable modules. Four groups of different webbings with different loads were defined for this study. Group 1 consisted of 4K Kevlar webbings loaded to 33% ultimate tensile strength and 6K Vectran webbings loaded to 27% ultimate tensile strength, group 2 consisted of 6K Kevlar webbings loaded to 40% and 43% ultimate tensile strength, and 6K Vectran webbings loaded to 50% ultimate tensile strength, group 3 consisted of 6K Kevlar webbings loaded to 52% ultimate tensile strength and 6K Vectran webbings loaded to 60% ultimate tensile strength, and group 4 consisted of 12.5K Kevlar webbings loaded to 22% ultimate tensile strength, and 12.5K Vectran webbings loaded to 22% ultimate tensile strength. The uniquely designed test facility, hardware, displacement measuring devices, and test data are presented. Test data indicate that immediately after loading all webbings stretch an inch or more, however as time increases displacement values significantly decrease to fall within a range of several hundredth of an inch over the remainder of test period. Webbings in group 1 exhibit near constant displacements and strains over a 17-month period. Data acquisition was suspended after the 17th month, however webbings continue to sustain load without any local webbing damage as of the 21st month of testing. Webbings in group 2 exhibit a combination of initial constant displacement and subsequent increases in displacement rates over a 16-month period. Webbings in group 3 exhibit steady increases in displacement rates leading to webbing failure over a 3-month period. Five of six webbings experienced local damage and subsequent failure in group 3. Data from group 4 indicates increasing webbing displacements over a 7-month period. All webbings in groups 1, 2, and 4 remain suspended without any local damage as of the writing of this paper. Variations in facility temperatures over test period seem to have had limited effect on long-term webbing displacement data

    Modification of cell wall polysaccharide guides cell division in <i>Streptococcus mutans</i>

    In ovoid-shaped, Gram-positive bacteria, MapZ guides FtsZ-ring positioning at cell equators. The cell wall of the ovococcus Streptococcus mutans contains peptidoglycan decorated with serotype c carbohydrates (SCCs). In the present study, we identify the major cell separation autolysin AtlA as an SCC-binding protein. AtlA binding to SCC is attenuated by the glycerol phosphate (GroP) modification. Using fluorescently labeled AtlA constructs, we mapped SCC distribution on the streptococcal surface, revealing enrichment of GroP-deficient immature SCCs at the cell poles and equators. The immature SCCs co-localize with MapZ at the equatorial rings throughout the cell cycle. In GroP-deficient mutants, AtlA is mislocalized, resulting in dysregulated cellular autolysis. These mutants display morphological abnormalities associated with MapZ mislocalization, leading to FtsZ-ring misplacement. Altogether, our data support a model in which maturation of a cell wall polysaccharide provides the molecular cues for the recruitment of cell division machinery, ensuring proper daughter cell separation and FtsZ-ring positioning.     PplD Is a De-N-Acetylase of the Cell Wall Linkage Unit of Streptococcal Rhamnopolysaccharides

    The cell wall of the human bacterial pathogen Group A Streptococcus (GAS) consists of peptidoglycan decorated with the Lancefield group A carbohydrate (GAC). GAC is a promising target for the development of GAS vaccines. In this study, employing chemical, compositional, and NMR methods, we show that GAC is attached to peptidoglycan via glucosamine 1-phosphate. This structural feature makes the GAC-peptidoglycan linkage highly sensitive to cleavage by nitrous acid and resistant to mild acid conditions. Using this characteristic of the GAS cell wall, we identify PplD as a protein required for deacetylation of linkage N-acetylglucosamine (GlcNAc). X-ray structural analysis indicates that PplD performs catalysis via a modified acid/base mechanism. Genetic surveys in silico together with functional analysis indicate that PplD homologs deacetylate the polysaccharide linkage in many streptococcal species. We further demonstrate that introduction of positive charges to the cell wall by GlcNAc deacetylation protects GAS against host cationic antimicrobial proteins

    Morphine paradoxically prolongs neuropathic pain in rats by amplifying spinal NLRP3 inflammasome activation

    Pain after disease/damage of the nervous system is predominantly treated with opioids, but without exploration of the long-term consequences. We demonstrate that a short course of morphine after nerve injury doubles the duration of neuropathic pain. Using genetic and pharmacological interventions, and innovative Designer Receptor Exclusively Activated by Designer Drugs disruption of microglia reactivity, we demonstrate that opioid-prolonged neuropathic pain arises from spinal microglia and NOD-like receptor protein 3 inflammasome formation/activation. Inhibiting these processes permanently resets amplified pain to basal levels, an effect not previously reported. These data support the “two-hit hypothesis” of amplification of microglial activation—nerve injury being the first “hit,” morphine the second. The implications of such potent microglial “priming” has fundamental clinical implications for pain and may extend to many chronic neurological disorders

    Where two worlds meet: language policing in mainstream and complementary schools in England

    We compare language policing in two educational contexts in England: mainstream schools and complementary schools. We draw on a varied dataset (policy documents, in-class observations, interviews) collected from mainstream schools and Greek complementary schools in London. We find similarities in how the two types of schools control, regulate, monitor and suppress the language of school students. Both settings hierarchise standardised and non-standardised varieties in institutional policies that delegitimise the non-standardised varieties. Teachers become vehicles for language ideologies in enacting monovarietal policies drawing on discourses around academic success and the primacy of written language over spoken language, including regional varieties such as Cypriot Greek. Our findings suggest that multilingual and multidialectal students in England who attend both mainstream and complementary schools are exposed to similar kinds of prescriptive discourses across the whole spectrum of their educational experiences, which can have a range of negative effects on their learning and the construction of their self-image. We argue that more links need to be forged between the two educational settings and that these should include the development of integrated pedagogies and policies that legitimise students’ whole linguistic repertoires, encompassing both their standardised and their non-standardised varieties as well as their other linguistic resources
