    Modelling and Simulation: Helping Students Acquire This Skill Using a Stock and Flow Approach With Mathbench

    Computational and modelling skills are vital to most fields of biological research, yet traditional biology majors have no or little opportunity to develop these skills during their undergraduate education. We describe an approach, which can address this issue by a synergy of online resources called MathBench modules and Stock and Flow modelling. Using a step-by-step method starting with a MathBench ‘bootcamp’, we were able to achieve a significant gain in quantitative skills of students with no previous experience with model building. At the end of the course, the students were able to construct and analyse complex models and gained confidence in mathematical skills

    Building Community-Based Approaches to Systemic Reform in Mathematical Biology Education

    Starting in the early 2000’s, several reports were released recognizing the convergence of mathematics, biology and computer science, and calling for a rethinking of how undergraduates are prepared for careers in research and the science and technology workforce. This call for change requires careful consideration of the mathematical biology education system to identify key components and leverage points for change. This paper demonstrates the wide range of resources and approaches available to the mathematical biology education community to create systemic change by highlighting the efforts of four community-based education reform organizations. A closer look at these organizations provides an opportunity to examine how to leverage components of the education system including faculty, academic institutions, students, access to resources, and the power of community

    Optical dating of K-feldspar grains from Middle Pleistocene lacustrine sediment at Marathousa 1 (Greece)

    Post-infrared infrared stimulated luminescence (pIRIR) measurements are reported for multiple aliquots of potassium-rich feldspar grains from sedimentary deposits at Marathousa 1 and Choremi Mine in the Megalopolis Basin in southern Greece. Ages were obtained for 9 samples from the deposits that over- and underlie as well as include the archaeological and palaeontological deposits at Marathousa 1. These sediments are sandwiched between lignite seams II and III and thought to represent sediment deposition during a single glacial period. A single age was obtained for a sample from Choremi mine. The equivalent dose estimates are based on a newly developed method presented elsewhere, and environmental dose rate determinations followed standard procedures. A specific focus of this paper is the determination of a representative estimate of time-averaged palaeo-water content of the organic and sand-rich deposits and the impact of porosity and compaction on these estimates. Ages are presented using two water content scenarios. These final ages have relatively large uncertainties, making it difficult to accurately assign deposition to a single oxygen isotope stage (OIS). Taking uncertainty into account, sediment deposition at Marathousa 1 occur sometime during OIS 12 and 11 and at Choremi mine during MIS 8. When combined with other proxy information, these results support the interpretation that the peat deposits (represented by lignite seams II and III) were deposited during warm interglacial periods, and that the intervening clastic materials were deposited during glacial periods. In this case, the ages are best interpreted as supporting deposition of sediments during MIS12. This is consistent with one of the proposed age models, but younger than the other

    This evaluation covers the work of the CIDVA from July 2021-June 2022, and sought to answer 3 key questions: 1. How well is the project working in term of outcomes and process? 2. Has the project experienced any barriers and have these impacted outcomes? 3. How could CIDVA activities be improved? 4. What learning has emerged for future work in the field of domestic abuse in children and young people? These questions were answered by analysing quarterly monitoring data for 2020-21 provided by the VAWG commissioning service as well as in-depth qualitative interviews with the CIDVAs and their managers, 2 young people who had used the service, and a range of other statutory and non-statutory service providers who work with children and young people in Islington

    Toward Integration: From Quantitative Biology to Mathbio-Biomath?

    In response to the call of BIO2010 for integrating quantitative skills into undergraduate biology education, 30 Howard Hughes Medical Institute (HHMI) Program Directors at the 2006 HHMI Program Directors Meeting established a consortium to investigate, implement, develop, and disseminate best practices resulting from the integration of math and biology. With the assistance of an HHMI-funded mini-grant, led by Karl Joplin of East Tennessee State University, and support in institutional HHMI grants at Emory and University of Delaware, these institutions held a series of summer institutes and workshops to document progress toward and address the challenges of implementing a more quantitative approach to undergraduate biology education. This report summarizes the results of the four summer institutes (2007–2010). The group developed four draft white papers, a wiki site, and a listserv. One major outcome of these meetings is this issue of CBE—Life Sciences Education, which resulted from proposals at our 2008 meeting and a January 2009 planning session. Many of the papers in this issue emerged from or were influenced by these meetings

    Structure-based design, synthesis and evaluation <em>in vitro</em> of arylnaphthyridinones, arylpyridopyrimidinones and their tetrahydro derivatives as inhibitors of the tankyrases

    The tankyrases are members of the PARP superfamily; they poly(ADP-ribosyl)ate their target proteins using NAD+ as a source of electrophilic ADP-ribosyl units. The three principal protein substrates of the tankyrases (TRF1, NuMA and axin) are involved in replication of cancer cells; thus inhibitors of the tankyrases may have anticancer activity. Using structure-based drug design and by analogy with known 3-arylisoquinolin-1-one and 2-arylquinazolin-4-one inhibitors, series of arylnaphthyridinones, arylpyridinopyrimidinones and their tetrahydro-derivatives were synthesised and evaluated in vitro. 7-Aryl-1,6-naphthyridin-5-ones, 3-aryl-2,6-naphthyridin-1-ones and 3-aryl-2,7-naphthyridin-1-ones were prepared by acid-catalysed cyclisation of the corresponding arylethynylpyridinenitriles or reaction of bromopyridinecarboxylic acids with ÎČ-diketones, followed by treatment with NH3. The 7-aryl-1,6-naphthyridin-5-ones were methylated at 1-N and reduced to 7-aryl-1-methyl-1,2,3,4-tetrahydro-1,6-naphthyridin-5-ones. Cu-catalysed reaction of benzamidines with bromopyridinecarboxylic acids furnished 2-arylpyrido[2,3-d]pyrimidin-4-ones. Condensation of benzamidines with methyl 1-benzyl-4-oxopiperidine-3-carboxylate and deprotection gave 2-aryl-5,6,7,8-tetrahydropyrido[4,3-d]pyrimidin-4-ones, aza analogues of the known inhibitor XAV939. Introduction of the ring-N in the arylnaphthyridinones and the arylpyridopyrimidinones caused &gt;1000-fold loss in activity, compared with their carbocyclic isoquinolinone and quinazolinone analogues. However, the 7-aryl-1-methyl-1,2,3,4-tetrahydro-1,6-naphthyridin-5-ones showed excellent inhibition of the tankyrases, with some examples having IC50 = 2 nM. One compound (7-(4-bromophenyl)-1-methyl-1,2,3,4-tetrahydro-1,6-naphthyridin-5-one) showed 70-fold selectivity for inhibition of tankyrase-2 versus tankyrase-1.
