První metoda byla vyvíjena pro stanovení léčiva ze skupiny inhibitorů protonové pumpy omeprazolu a druhá pro stanovení retinolu jako nového potencionálního biomarkeru poškození ledvin. V teoretické části práce jsou blíže popsány jednotlivé analyty, jejich klinický význam, možnosti stanovení a charakteristiky matric, ve kterých byly cílové analyty stanovovány. Dále je zde diskutována úprava vzorku před analýzou a rozebrány validační normy vztahující se k chromatografickým metodám používaným jak ve farmaceutické analýze, tak v bioanalýze. V experimentální části je blíže uveden vývoj, optimalizace a validace UHPLC metod a jejich následné využití. Ve farmaceuticko-analytické části práce je popsáno stanovení omeprazolu v nových suspenzních přípravcích zahrnující využití UHPLC systému, kolon s částicemi s porézním povrchem a velmi jednoduchou úpravu vzorku. RNDr. Lenka Kujovská Krčmová, Ph.D. Title of Dissertation Thesis: Usage of HPLC and UHPLC - MS/MS techniques for clinical research The dissertation thesis is dealing with the extraction techniques and with development, optimization and validation of chromatographic methods for the determination of omeprazol which is drug from the group of proton pump inhibitors, and retinol as a new potential biomarker of kidney damage. The theoretical part of the thesis is focused on detailed description of the individual analytes, their clinical significance, options of determination and the matrix in which the target analytes were determined. Furthermore, the preparation of the sample before the analysis is discussed and validation recommendations related to chromatographic methods used in both pharmaceutical analysis and bioanalysis are also mentioned. In the experimental part the development, optimization and validation of UHPLC methods and their subsequent use are described in detail. Key words: Liriodendron tulipifera, alkaloids, biological activity. Alkaloids isolated from the leaves of Liriodendron tulipifera L. have been described to have a various biological activity. For Alzheimer's disease (AD) treatment is significant primarily the alkaloid's inhibitory activity towards enzymes acetylcholinesterase and butyrylcholinesterase, which contribute to the pathogenesis of AD. Substances affecting these enzymes represent the main accessible approach to AD treatment, therefore plant-based drug screening remains a relevant topic. An alkaloid extract from the leaves of Liriodendron tulipifera was chosen for an isolation and identification of alkaloids due to the positive results of preliminary tests on its cholinesterase inhibitory activity. The extract was separated by using a flash chromatography. The isolation of alkaloids from selected fractions (no. 81-89 and no. 68-89) was done by the use of preparative thin-layer chromatography. Pro léčbu Alzheimerovy choroby (AD) je významná především jejich inhibiční aktivita vůči acetylcholinesterase a butyrylcholinesterase, tedy enzymům podílejících se na patogenezi AD. Látky ovlivňující tyto enzymy v současnosti představují hlavní dostupný přístup k léčbě AD, screening rostlinných drog proto zůstává aktuálním tématem. Alkaloidní extrakt z listů Liriodendron tulipifera byl zvolen k izolaci alkaloidů a k jejich dalšímu zkoumání z důvodu pozitivních výsledků předběžných testů na jeho inhibiční cholinesterasovou aktivitu. Zvolenou metodou pro separaci extraktu byla flash chromatografie. K izolaci alkaloidů ze zvolených frakcí (č. 81-89 a č. 68-80) byla využita preparativní tenkovrstvá chromatografie. U izolovaných látek byla vyhodnocena strukturní analýza (pomocí NMR a MS) a byla měřena jejich optická otáčivost. Burn out syndrom at healthcare social workers

The subject of this bachelor is ,,Burnout syndrome Health social workers" This work was created on the basis of my interest, because I am interested in burnout syndrome and in health social work, which I would like to work in after graduating from this study programme. The thesis deals with the problem of burnout syndrome between health social workers and appeals to critical information about prevention of his rise. The thesis is divided into two parts. In the theoretical part is describes the term burnout syndrome, his symptoms, phases, diagnostics and prevention. Helper syndrome is described here too and caddish with burnout syndrome and stress. Further in this part it describes the profession of a health social worker, his education, competency, ethic, types of clients, multidisciplinarity and at least risks for beginning burnout syndrome in health social workers. The practical part is to utilize the quantity method of research with questionnaire. Participants of this research were 123 people and dates were analyzed by standardized method MBI - Maslach Burnout Inventory.Tématem této bakalářské práce je: ,,Syndrom vyhoření u zdravotně sociálních pracovníků". Práce vznikla na základě mého zájmu o problematiku syndromu vyhoření a také o profesi zdravotně sociálního pracovníka, které bych se po absolvování tohoto oboru ráda věnovala. Práce se zabývá představením problému vyhoření mezi zdravotně sociálními pracovníky a apelováním na důležitost prevence jeho vzniku. Je rozdělena do dvou částí. V teoretické části je vysvětlen pojem syndrom vyhoření, popsány jeho příznaky, stádia, diagnostika a prevence. Také je zde popsán syndrom pomáhajících a souvislost mezi vyhořením a stresem. Dále je v této části objasněna profese zdravotně sociálního pracovníka, jeho vzdělání, kompetence, etika, typologie jeho klientů, multidisciplinarita a nakonec jsou zde uvedena rizika vzniku syndromu vyhoření u zdravotně sociálních pracovníků. V praktické části je využita kvantitativní metoda sběru dat, pomocí dotazníkového šetření. A violin sings and talks - a musical education program for elementary students

Diplomová práce je zaměřena na housle a výchovné pořady. Teoretická část se zabývá historií houslí, stavbou a hrou na tento nástroj a slavnými houslisty. Dále hudebními pořady a výchovnými koncerty, jejich organizátory Leonardem Bernsteinem a Františkem Blahou a jejich dramaturgií, společně s tvorbou koncertu. Také se věnuje výběru houslových poslechových skladeb, hudebně výrazovým prostředkům houslové hry a houslových skladeb, kde je vysvětleno, jak housle zpívají a o čem housle vyprávějí, charakteristickým hudebně vyjadřovacím prostředkům skladeb pro housle - melodii, rytmu a dynamice. Praktická část představuje návrh hudebního pořadu pro žáky na 1. stupni základní školy, jenž se jmenuje: Housle zpívají a vyprávějí. Tento pořad by byl vhodný pro žáky 1. - 3. ročníku. Cílem práce je vytvořit výchovný pořad a přiblížit housle dětem. Představit jim tento nástroj a ukázat, že housle nejen zpívají, ale také vyprávějí.ObhájenoThe diploma thesis is focused on violin and educational programs. The theoretical part deals with the history of the violin, the design and playing of this instrument and famous violinists. Furthermore, it considers music programs and educational concerts, their organizers Leonard Bernstein and František Blaha and their dramaturgy, along with the creation of the concerts. It also deals with the selection of violin instrumentals/compositions; musical means of violin playing and violin compositions; which explain how a violin can sing and how a violin conveys its musical characteristics - melody, rhythm, and dynamics. The practical part is a music program proposal for primary school students called: Violins sing and narrate. This program would be for 1st - 3rd year students. Implementation of Reading and Writing for Critical Thinking Programme in the Teaching of Interwar Czechoslovak History (1918 - 1938) at Lower Secondary Schools: Case Study

Tato diplomová práce seznamuje s konstruktivistickým pojetím výuky, které sleduje a podporuje přirozené způsoby učení. Popisuje komplexní vzdělávací program Čtením a psaním ke kritickému myšlení a jeho aplikaci ve výuce moderních dějin se zvláštním zaměřením na dějiny Československa v letech 1918 - 1938. Přípravy hodin respektují třífázový model učení E - U - R (evokace - uvědomění si významu - reflexe) a využívají různé druhy školních historických pramenů. V praktické části práce se nachází pět návrhů vyučovacích hodin zaměřených na dějiny Československa v letech 1918-1938, z nichž některé byly částečně ověřeny v praxi. Cílem práce je sloužit jako praktický průvodce pro učitele, kteří hledají inspiraci pro výuku daného historického období, či hledají nové cesty, jak svou výuku zlepšit a obohatit. This diploma thesis introduces the concept of the constructivism, which follows and supports the natural ways of learning, as a paradigm for teaching and learning. First, the text describes a comprehensive educational programme Reading and Writing for Critical Thinking (RWCT). Second, application of the programme in the teaching of modern history is discussed with the special reference to the history of Czechoslovakia in the years 1918-1938.
