    Can differences in phosphorus uptake kinetics explain the distribution of cattail and sawgrass in the Florida Everglades?

    <p>Abstract</p> <p>Background</p> <p>Cattail (<it>Typha domingensis</it>) has been spreading in phosphorus (P) enriched areas of the oligotrophic Florida Everglades at the expense of sawgrass (<it>Cladium mariscus </it>spp. <it>jamaicense</it>). Abundant evidence in the literature explains how the opportunistic features of <it>Typha </it>might lead to a complete dominance in P-enriched areas. Less clear is how <it>Typha </it>can grow and acquire P at extremely low P levels, which prevail in the unimpacted areas of the Everglades.</p> <p>Results</p> <p>Apparent P uptake kinetics were measured for intact plants of <it>Cladium </it>and <it>Typha </it>acclimated to low and high P at two levels of oxygen in hydroponic culture. The saturated rate of P uptake was higher in <it>Typha </it>than in <it>Cladium </it>and higher in low-P acclimated plants than in high-P acclimated plants. The affinity for P uptake was two-fold higher in <it>Typha </it>than in <it>Cladium</it>, and two- to three-fold higher for low-P acclimated plants compared to high-P acclimated plants. As <it>Cladium </it>had a greater proportion of its biomass allocated to roots, the overall uptake capacity of the two species at high P did not differ. At low P availability, <it>Typha </it>increased biomass allocation to roots more than <it>Cladium</it>. Both species also adjusted their P uptake kinetics, but <it>Typha </it>more so than <it>Cladium</it>. The adjustment of the P uptake system and increased biomass allocation to roots resulted in a five-fold higher uptake per plant for <it>Cladium </it>and a ten-fold higher uptake for <it>Typha</it>.</p> <p>Conclusions</p> <p>Both <it>Cladium </it>and <it>Typha </it>adjust P uptake kinetics in relation to plant demand when P availability is high. When P concentrations are low, however, <it>Typha </it>adjusts P uptake kinetics and also increases allocation to roots more so than <it>Cladium</it>, thereby improving both efficiency and capacity of P uptake. <it>Cladium </it>has less need to adjust P uptake kinetics because it is already efficient at acquiring P from peat soils (e.g., through secretion of phosphatases, symbiosis with arbuscular mycorrhizal fungi, nutrient conservation growth traits). Thus, although <it>Cladium </it>and <it>Typha </it>have qualitatively similar strategies to improve P-uptake efficiency and capacity under low P-conditions, <it>Typha </it>shows a quantitatively greater response, possibly due to a lesser expression of these mechanisms than <it>Cladium</it>. This difference between the two species helps to explain why an opportunistic species such as <it>Typha </it>is able to grow side by side with <it>Cladium </it>in the P-deficient Everglades.</p

    Mangrove Peat Collapse Following Mass Tree Mortality: Implications for Forest Recovery from Hurricane Mitch

    1 We measured sediment elevation and accretion dynamics in mangrove forests on the islands of Guanaja and Roatan, Honduras, impacted by Hurricane Mitch in 1998 to determine if collapse of underlying peat was occurring as a result of mass tree mortality. Little is known about the balance between production and decomposition of soil organic matter in the maintenance of sediment elevation of mangrove forests with biogenic soils. 2 Sediment elevation change measured with the rod surface elevation table from 18 months to 33 months after the storm differed significantly among low, medium and high wind impact sites. Mangrove forests suffering minimal to partial mortality gained elevation at a rate (5 mm year−1) greater than vertical accretion (2 mm year−1) measured from artificial soil marker horizons, suggesting that root production contributed to sediment elevation. Basin forests that suffered mass tree mortality experienced peat collapse of about 11 mm year−1 as a result of decomposition of dead root material and sediment compaction. Low soil shear strength and lack of root growth accompanied elevation decreases. 3 Model simulations using the Relative Elevation Model indicate that peat collapse in the high impact basin mangrove forest would be 37 mm year−1 for the 2 years immediately after the storm, as root material decomposed. In the absence of renewed root growth, the model predicts that peat collapse will continue for at least 8 more years at a rate (7 mm year−1) similar to that measured (11 mm year−1). 4 Mass tree mortality caused rapid elevation loss. Few trees survived and recovery of the high impact forest will thus depend primarily on seedling recruitment. Because seedling establishment is controlled in large part by sediment elevation in relation to tide height, continued peat collapse could further impair recovery rates

    Evaporative loss from irrigated interrows in a highly advective semi-arid agricultural area

    Agricultural productivity has increased in the Texas High Plains at the cost of declining water tables, putting at risk the sustainability of the Ogallala Aquifer as a principal source of water for irrigated agriculture. This has led area producers to seek alternative practices that can increase water use efficiency (WUE) through more careful management of water. One potential way of improving WUE is by reducing soil evaporation (E), thus reducing overall evapotranspiration (ET). Before searching for ways to reduce E, it is first important to quantify E and understand the factors that determine its magnitude. The objectives of this study were (1) to quantify E throughout part of the growing season for irrigated cotton in a strongly advective semi-arid region; (2) to study the effects of LAI, days after irrigation, and measurement location within the row on the E/ET fraction; and (3) to study the ability of microlysimeter (ML) measures of E combined with sap flow gage measures of transpiration (T) to accurately estimate ET when compared with weighing lysimeter ET data and to assess the E/T ratio. The research was conducted in an irrigated cotton field at the Conservation & Production Research Laboratory of the USDA-ARS, Bushland, TX. ET was measured by a large weighing lysimeter, and E was measured by 10 microlysimeters that were deployed in two sets of 5 across the interrow. In addition, 10 heat balance sap flow gages were used to determine T. A moderately good agreement was found between the sum E + T and ET (SE = 1 mm or ~10% of ET). It was found that E may account for \u3e50% of ET during early stages of the growing season (LAI \u3c 0.2), significantly decreasing with increase in LAI to values near 20% at peak LAI of three. Measurement location within the north-south interrows had a distinct effect on the diurnal pattern of E, with a shift in time of peak E from west to east, a pattern that was governed by the solar radiation reaching the soil surface. However, total daily E was unaffected by position in the interrow. Under wet soil conditions, wind speed and direction affected soil evaporation. Row orientation interacted with wind direction in this study such that aerodynamic resistance to E usually increased when wind direction was perpendicular to row direction; but this interaction needs further study because it appeared to be lessened under higher wind speeds

    A laminin 511 matrix is regulated by TAZ and functions as the ligand for the alpha6Bbeta1 integrin to sustain breast cancer stem cells

    Understanding how the extracellular matrix impacts the function of cancer stem cells (CSCs) is a significant but poorly understood problem. We report that breast CSCs produce a laminin (LM) 511 matrix that promotes self-renewal and tumor initiation by engaging the alpha6Bbeta1 integrin and activating the Hippo transducer TAZ. Although TAZ is important for the function of breast CSCs, the mechanism is unknown. We observed that TAZ regulates the transcription of the alpha5 subunit of LM511 and the formation of a LM511 matrix. These data establish a positive feedback loop involving TAZ and LM511 that contributes to stemness in breast cancer

    Phase I Study of Safety and Immunogenicity of an Escherichia coli-Derived Recombinant Protective Antigen (rPA) Vaccine to Prevent Anthrax in Adults

    The fatal disease caused by Bacillus anthracis is preventable with a prophylactic vaccine. The currently available anthrax vaccine requires a lengthy immunization schedule, and simpler and more immunogenic options for protection against anthrax are a priority for development. In this report we describe a phase I clinical trial testing the safety and immunogenicity of an anthrax vaccine using recombinant Escherichia coli-derived, B. anthracis protective antigen (rPA).A total of 73 healthy adults ages 18-40 were enrolled and 67 received 2 injections separated by 4 weeks of either buffered saline placebo, or rPA formulated with or without 704 µg/ml Alhydrogel® adjuvant in increasing doses (5, 25, 50, 100 µg) of rPA. Participants were followed for one year and safety and immunologic data were assessed. Tenderness and warmth were the most common post-injection site reactions. No serious adverse events related to the vaccine were observed. The most robust humoral immune responses were observed in subjects receiving 50 µg of rPA formulated with Alhydrogel® with a geometric mean concentration of anti-rPA IgG antibodies of 283 µg/ml and a toxin neutralizing geometric 50% reciprocal geometric mean titer of 1061. The highest lymphoproliferative peak cellular response (median Lymphocyte Stimulation Index of 29) was observed in the group receiving 25 µg Alhydrogel®-formulated rPA.The vaccine was safe, well tolerated and stimulated a robust humoral and cellular response after two doses.ClinicalTrials.gov NCT00057525

    Patterns and Perceptions of Climate Change in a Biodiversity Conservation Hotspot

    Quantifying local people's perceptions to climate change, and their assessments of which changes matter, is fundamental to addressing the dual challenge of land conservation and poverty alleviation in densely populated tropical regions To develop appropriate policies and responses, it will be important not only to anticipate the nature of expected changes, but also how they are perceived, interpreted and adapted to by local residents. The Albertine Rift region in East Africa is one of the world's most threatened biodiversity hotspots due to dense smallholder agriculture, high levels of land and resource pressures, and habitat loss and conversion. Results of three separate household surveys conducted in the vicinity of Kibale National Park during the late 2000s indicate that farmers are concerned with variable precipitation. Many survey respondents reported that conditions are drier and rainfall timing is becoming less predictable. Analysis of daily rainfall data for the climate normal period 1981 to 2010 indicates that total rainfall both within and across seasons has not changed significantly, although the timing and transitions of seasons has been highly variable. Results of rainfall data analysis also indicate significant changes in the intra-seasonal rainfall distribution, including longer dry periods within rainy seasons, which may contribute to the perceived decrease in rainfall and can compromise food security. Our results highlight the need for fine-scale climate information to assist agro-ecological communities in developing effective adaptive management

    Effects of antiplatelet therapy on stroke risk by brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases: subgroup analyses of the RESTART randomised, open-label trial

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy

    Inequalities in the use of secondary prevention of cardiovascular disease by socioeconomic status: evidence from the PURE observational study

    Background: There is little evidence on the use of secondary prevention medicines for cardiovascular disease by socioeconomic groups in countries at different levels of economic development. Methods: We assessed use of antiplatelet, cholesterol, and blood-pressure-lowering drugs in 8492 individuals with self-reported cardiovascular disease from 21 countries enrolled in the Prospective Urban Rural Epidemiology (PURE) study. Defining one or more drugs as a minimal level of secondary prevention, wealth-related inequality was measured using the Wagstaff concentration index, scaled from −1 (pro-poor) to 1 (pro-rich), standardised by age and sex. Correlations between inequalities and national health-related indicators were estimated. Findings: The proportion of patients with cardiovascular disease on three medications ranged from 0% in South Africa (95% CI 0–1·7), Tanzania (0–3·6), and Zimbabwe (0–5·1), to 49·3% in Canada (44·4–54·3). Proportions receiving at least one drug varied from 2·0% (95% CI 0·5–6·9) in Tanzania to 91·4% (86·6–94·6) in Sweden. There was significant (p<0·05) pro-rich inequality in Saudi Arabia, China, Colombia, India, Pakistan, and Zimbabwe. Pro-poor distributions were observed in Sweden, Brazil, Chile, Poland, and the occupied Palestinian territory. The strongest predictors of inequality were public expenditure on health and overall use of secondary prevention medicines. Interpretation: Use of medication for secondary prevention of cardiovascular disease is alarmingly low. In many countries with the lowest use, pro-rich inequality is greatest. Policies associated with an equal or pro-poor distribution include free medications and community health programmes to support adherence to medications.
