    Submarine slope failures in gassy soils

    Submarine slope stability has become an important concern and a subject of research with increasing demand for offshore developments and technological advancement for harsh and challenging environments. The consequences of submarine slope failure adjacent to oil and gas facilities would have a large financial, safety and regulatory impact. This current research work investigates potential failure of submarine gassy slopes triggered by tidal variations. Due to tidal variations, failure of an unsaturated slope may occur under specific combinations of increasing degree of saturation and soil permeability, and decreasing tidal period. Novel physical model tests in a geotechnical centrifuge were undertaken to examine submarine slope failure mechanisms containing gassy sediments. The model preparation techniques, measurement systems and results are presented. The response observed in the model test is discussed and further developments proposed. The buried PPT’s response of the submarine slope are comparable in terms of attenuation and phase lag with Nagaswaran (1983) and with field measurements of Atigh and Byrne (2004) in terms of phase lag

    Knowledge, practices and gender discrepancies of migrant labours concerning malaria in Bhubaneswar, Odisha, India

    Background: Bhubaneswar, the capital of Odisha is an up-coming city since 1960s. The capital is undergoing various developmental activities and rapid industrialization. This is leading to migration of labours from various districts of Odisha including malaria endemic districts. The records available in central malaria laboratory, Bhubaneswar revealed high malaria incidence (ranged from 5% to 11% during last five years).Methods: The study was undertaken in Bhubaneswar, Odisha, India, on migrant laboures (300 samples) through a pre-structured schedule.Results: The study revealed the migrated respondents were from malaria endemic districts of Odisha. The malaria endemicity of the districts ranged from 2.2-9.8. It was also found that only 6% of the migrants were from low endemic areas i.e. SPR5. Study of the awareness of the respondents showed 31% males and 9% females knew mosquito bite as a cause of malaria. Rapid Diagnostic Test showed twenty seven numbers of respondents tested positive for malaria (24 positive for PF and 3 positive for PV), out of them 6 were males and 21 were females. The study of their sleeping habits and personal protection methods showed that 32% respondents slept outdoor and 73% did not use anything as protection against mosquito bite. The study regarding knowledge of malaria signs and symptoms among respondents showed that the most commonly recognized signs and symptoms of malaria were headache (23%).  Vector incrimination study showed that per man hour density of an. stephensi is 5.7.Conclusions: Thus presence of efficient malaria vectors and inadequate knowledge in the migrant labour community and emphatically among female migrant labours regarding cause, symptoms, prevention, control measures and risks associated with malaria is the major reason of malaria transmission.

    Epigenetic suppression of human telomerase ( hTERT ) is mediated by the metastasis suppressor NME2 in a G-quadruplex–dependent fashion

    Transcriptional activation of the human telomerase reverse transcriptase (hTERT) gene, which remains repressed in adult somatic cells, is critical during tumorigenesis. Several transcription factors and the epigenetic state of the hTERT promoter are known to be important for tight control of hTERT in normal tissues, but the molecular mechanisms leading to hTERT reactivation in cancer are not well-understood. Surprisingly, here we found occupancy of the metastasis suppressor non-metastatic 2 (NME2) within the hTERT core promoter in HT1080 fibrosarcoma cells and HCT116 colon cancer cells and NME2-mediated transcriptional repression of hTERT in these cells. We also report that loss of NME2 results in up-regulated hTERT expression. Mechanistically, additional results indicated that the RE1-silencing transcription factor (REST)–lysine-specific histone demethylase 1 (LSD1) co-repressor complex associates with the hTERT promoter in an NME2-dependent way and that this assembly is required for maintaining repressive chromatin at the hTERT promoter. Interestingly, a G-quadruplex motif at the hTERT promoter was essential for occupancy of NME2 and the REST repressor complex on the hTERT promoter. In light of this mechanistic insight, we studied the effects of G-quadruplex–binding ligands on hTERT expression and observed that several of these ligands repressed hTERT expression. Together, our results support a mechanism of hTERT epigenetic control involving a G-quadruplex promoter motif, which potentially can be targeted by tailored small molecules

    Metastases suppressor NME2 associates with telomere ends and telomerase and reduces telomerase activity within cells

    Analysis of chromatin-immunoprecipitation followed by sequencing (ChIP-seq) usually disregards sequence reads that do not map within binding positions (peaks). Using an unbiased approach, we analysed all reads, both that mapped and ones that were not included as part of peaks. ChIP-seq experiments were performed in human lung adenocarcinoma and fibrosarcoma cells for the metastasis suppressor non-metastatic 2 (NME2). Surprisingly, we identified sequence reads that uniquely represented human telomere ends in both cases. In vivo presence of NME2 at telomere ends was validated using independent methods and as further evidence we found intranuclear association of NME2 and the telomere repeat binding factor 2. Most remarkably, results demonstrate that NME2 associates with telomerase and reduces telomerase activity in vitro and in vivo, and sustained NME2 expression resulted in reduced telomere length in aggressive human cancer cells. Anti-metastatic function of NME2 has been demonstrated in human cancers, however, mechanisms are poorly understood. Together, findings reported here suggest a novel role for NME2 as a telomere binding protein that can alter telomerase function and telomere length. This presents an opportunity to investigate telomere-related interactions in metastasis suppression

    Metastases suppressor NM23-H2 interaction with G-quadruplex DNA within c-MYC promoter nuclease hypersensitive element induces c-MYC expression

    Regulatory influence of the G-quadruplex or G4 motif present within the nuclease hypersensitive element (NHE) in the promoter of c-MYC has been noted. On the other hand, association of NM23-H2 to the NHE leads to c-MYC activation. Therefore, NM23-H2 interaction with the G4 motif within the c-MYC NHE presents an interesting mechanistic possibility. Herein, using luciferase reporter assay and chromatin immunoprecipitation we show NM23-H2 mediated c-MYC activation involves NM23-H2-G4 motif binding within the c-MYC NHE. G4 motif complex formation with recombinant NM23-H2 was independently confirmed using fluorescence energy transfer, which also indicated that the G4 motif was resolved to an unfolded state within the protein-bound complex. Taken together, this supports transcriptional role of NM23-H2 via a G4 motif

    Domestic violence against women in eastern India: a population-based study on prevalence and related issues

    <p>Abstract</p> <p>Background</p> <p>Violence against women is now widely recognised as an important public health problem, owing to its health consequences. Violence against women among many Indian communities on a regularly basis goes unreported. The objective of this study is to report the prevalence and other related issues of various forms of domestic violence against women from the eastern zone of India.</p> <p>Methods</p> <p>It is a population-based study covering both married women (n = 1718) and men (n = 1715) from three of the four states of Eastern India selected through a systematic multistage sampling strategy. Interviews were conducted using separate pre-piloted structured questionnaires for women (victimization) and men (perpetration). Women were asked whether their husband or any other family members committed violent acts against them. And men were asked whether they had ever perpetrated violent acts against their wives. Three principle domestic violence outcome variables (physical, psychological and sexual violence) were determined by response to a set of questions for each variable. In addition, data on socio-economic characteristics were collected. Descriptive statistics, bi- and multivariate analyses were done.</p> <p>Results</p> <p>The overall prevalence of physical, psychological, sexual and any form of violence among women of Eastern India were 16%, 52%, 25% and 56% respectively. These rates reported by men were 22%, 59%, 17% and 59.5% respectively. Men reported higher prevalence of all forms of violence apart from sexual violence. Husbands were mostly responsible for violence in majority of cases and some women reported the involvement of husbands' parents. It is found that various acts of violence were continuing among majority of women who reported violence. Some socio-economic characteristics of women have significant association with the occurrence of domestic violence. Urban residence, older age, lower education and lower family income are associated with occurrence of domestic violence. Multivariate logistic regressions revealed that the physical violence has significant association with state, residence (rural or urban), age and occupation of women, and monthly family income. Similar associations are found for psychological violence (with residence, age, education and occupation of the women and monthly family income) and sexual violence (with residence, age and educational level of women).</p> <p>Conclusion</p> <p>The prevalence of domestic violence in Eastern India is relatively high compared to majority of information available from India and confirms that domestic violence is a universal phenomenon. The primary healthcare institutions in India should institutionalise the routine screening and treatment for violence related injuries and trauma. Also, these results provide vital information to assess the situation to develop public health interventions, and to sensitise the concerned agencies to implement the laws related to violence against women.</p

    CURATION AND MANAGEMENT OF CULTURAL HERITAGE THROUGH LIBRARIES

    Libraries, museums and archives hold valuable collections in a variety of media, presenting a vast body of knowledge rooted in the history of human civilisation. These form the repository of the wisdom of great works by thinkers of past and the present. The holdings of these institutions are priceless heritage of the mankind as they preserve documents, ideas, and the oral and written records. To value the cultural heritage and to care for it as a treasure bequeathed to us by our ancestors is the major responsibility of libraries. The past records constitute a natural resource and are indispensable to the present generation as well as to the generations to come. Libraries preserve the documentary heritage resources for which they are primarily responsible. Any loss of such materials is simply irreplaceable. Therefore, preserving this intellectual, cultural heritage becomes not only the academic commitment but also the moral responsibility of the librarians/information scientists, who are in charge of these repositories. The high quality of the papers and the discussion represent the thinking and experience of experts in their particular fields. The contributed papers also relate to the methodology used in libraries in Asia to provide access to manuscripts and cultural heritage. The volume discusses best practices in Knowledge preservation and how to collaborate and preserve the culture. The book also deals with manuscript and archives issues in the digital era. The approach of this book is concise, comprehensively, covering all major aspects of preservation and conservation through libraries. The readership of the book is not just limited to library and information science professionals, but also for those involved in conservation, preservation, restoration or other related disciplines. The book will be useful for librarians, archivists and conservators.
