    Design and Preliminary Testing of Demand-Responsive Transverse Rumble Strips

    Transverse rumble strips are common practice to alert drivers by engaging their auditory and tactile senses in addition to visual senses by traffic signals. However, continuous exposure to noise and vibration by transverse rumble strips often results in diminished effectiveness and erratic behaviors, leading to additional safety challenges. In response, demand-responsive transverse rumble strips were developed as traffic safety countermeasures that reduce unnecessary noise and vibration associated with transverse rumble strips by incorporating active control of the rumble strips. Rather than staying static, demand-responsive transverse rumble strips are activated based on the presence of pedestrians, at predesignated times, or in response to abrupt changes in traffic flow. To evaluate the effectiveness of demand-responsive transverse rumble strips, the research team assessed noise and vibration data, both inside the vehicles and on the roadside, for various types of vehicles traveling at different speeds. The test data indicate that demand-responsive transverse rumble strips produced noticeable in-vehicle noise and vibration that could alert drivers to downstream events. Furthermore, demand-responsive transverse rumble strips generated sufficient noise to alert roadside pedestrians to vehicle presence but at low enough level to be considered as acceptable for a residential neighborhood use. Accordingly, demand-responsive transverse rumble strips could address the challenges that static transverse rumble strips face, by providing a design with relatively limited noise while enhancing safety

    Real-Time Anticipatory Suspension Control for Single Event Disturbances

    Most commercial vehicles currently on the market are still equipped with a passive suspension system, while some luxury brands may already use an adaptive suspension. Active suspension systems on the other hand are rarely found, however, they offer great opportunities to close the gap of the well-known trade-off between ride comfort and handling. Besides that, they can also be used to mitigate single event disturbances, an objective of the USA army as announced in a solicitation which initiated and motivated this research. In addition to that, several studies were found stating the impact and danger of potholes and their impact on the vehicle and passenger. Reviewing the literature, several control strategies for controlling active suspension systems were found. However, most of these approaches used feedback control and did not try to mitigate single event disturbances. Since literature also suggested making use of look ahead preview, research at the Performance Engineering Research Lab at Virginia Tech was started in 2015 combining look ahead preview and an adaptive system to generate optimal force profiles. This introductory research succeeded and proved the used approach to be very promising. However, the used adaptive system was not designed to operate in real-time and did not show any correlation between different road profiles. Therefore, the main objective of this research project is to evaluate and analyze each of the adaptive systems by searching for correlations in their solutions. The results then should be used in order to design a control law which emulates the adaptive system and can be used in a real-time environment. First, an overall research methodology was derived. According to this a software application was developed which extracts ideal force profiles from single event disturbance signals in order to miti- gate their impact to the vehicle. The application uses a quarter car model with a partially loaded active suspension system, a set of predefined road profiles, a road profile preprocessor, and an adaptive algorithm. The preprocessing includes geometric filtering using a Tandem-Cam Model and the adaptive processor used an iterative version of the Filtered-X Last-Mean-Square algorithm. During evaluation and analysis of several generated data sets, high correlations in the generated and adjusted adaptive systems were discovered. From these an empirical and theoretical universal filter model was derived, which was then used to design an open-loop control law named Optimal Force Control. The original control law and an adjusted version designed for a real-time environment were tested for all predefined road profiles over all considered vehicle velocities and prove to perform much better than the offline solution using the adaptive system. In summary, a control law named Optimal Force Control was designed which can be used and implemented in a vehicle to extract an analytical and ideal force profile given a road profile input. Implementing an active suspension system with tracking controller, this approach can be used in order to mitigate single event disturbance signals by reducing the vertical vehicle acceleration

    Comparative Analysis of the Glycosylation Profiles of Membrane-Anchored HIV-1 Envelope Glycoprotein Trimers and Soluble gp140

    The human immunodeficiency virus type 1 (HIV-1) envelope glycoprotein (Env) trimer, which consists of the gp120 and gp41 subunits, is the focus of multiple strategies for vaccine development. Extensive Env glycosylation provides HIV-1 with protection from the immune system, yet the glycans are also essential components of binding epitopes for numerous broadly neutralizing antibodies. Recent studies have shown that when Env is isolated from virions, its glycosylation profile differs significantly from that of soluble forms of Env (gp120 or gp140) predominantly used in vaccine discovery research. Here we show that exogenous membrane-anchored Envs, which can be produced in large quantities in mammalian cells, also display a virion-like glycan profile, where the glycoprotein is extensively decorated with high-mannose glycans. Additionally, because we characterized the glycosylation with a high-fidelity profiling method, glycopeptide analysis, an unprecedented level of molecular detail regarding membrane Env glycosylation and its heterogeneity is presented. Each glycosylation site was characterized individually, with about 500 glycoforms characterized per Env protein. While many of the sites contain exclusively high-mannose glycans, others retain complex glycans, resulting in a glycan profile that cannot currently be mimicked on soluble gp120 or gp140 preparations. These site-level studies are important for understanding antibody-glycan interactions on native Env trimers. Additionally, we report a newly observed O-linked glycosylation site, T606, and we show that the full O-linked glycosylation profile of membrane-associated Env is similar to that of soluble gp140. These findings provide new insight into Env glycosylation and clarify key molecular-level differences between membrane-anchored Env and soluble gp140

    Secreted nuclear protein DEK regulates hematopoiesis through CXCR2 signaling

    The nuclear protein DEK is an endogenous DNA-binding chromatin factor regulating hematopoiesis. DEK is one of only 2 known secreted nuclear chromatin factors, but whether and how extracellular DEK regulates hematopoiesis is not known. We demonstrated that extracellular DEK greatly enhanced ex vivo expansion of cytokine-stimulated human and mouse hematopoietic stem cells (HSCs) and regulated HSC and hematopoietic progenitor cell (HPC) numbers in vivo and in vitro as determined both phenotypically (by flow cytometry) and functionally (through transplantation and colony formation assays). Recombinant DEK increased long-term HSC numbers and decreased HPC numbers through a mechanism mediated by the CXC chemokine receptor CXCR2 and heparan sulfate proteoglycans (HSPGs) (as determined utilizing Cxcr2-/- mice, blocking CXCR2 antibodies, and 3 different HSPG inhibitors) that was associated with enhanced phosphorylation of ERK1/2, AKT, and p38 MAPK. To determine whether extracellular DEK required nuclear function to regulate hematopoiesis, we utilized 2 mutant forms of DEK: one that lacked its nuclear translocation signal and one that lacked DNA-binding ability. Both altered HSC and HPC numbers in vivo or in vitro, suggesting the nuclear function of DEK is not required. Thus, DEK acts as a hematopoietic cytokine, with the potential for clinical applicability

    Observation of High-Energy Astrophysical Neutrinos in Three Years of IceCube Data

    A search for high-energy neutrinos interacting within the IceCube detector between 2010 and 2012 provided the first evidence for a high-energy neutrino flux of extraterrestrial origin. Results from an analysis using the same methods with a third year (2012-2013) of data from the complete IceCube detector are consistent with the previously reported astrophysical flux in the 100 TeV - PeV range at the level of 10−8 GeV cm−2 s−1 sr−110^{-8}\, \mathrm{GeV}\, \mathrm{cm}^{-2}\, \mathrm{s}^{-1}\, \mathrm{sr}^{-1} per flavor and reject a purely atmospheric explanation for the combined 3-year data at 5.7σ5.7 \sigma. The data are consistent with expectations for equal fluxes of all three neutrino flavors and with isotropic arrival directions, suggesting either numerous or spatially extended sources. The three-year dataset, with a livetime of 988 days, contains a total of 37 neutrino candidate events with deposited energies ranging from 30 to 2000 TeV. The 2000 TeV event is the highest-energy neutrino interaction ever observed.Comment: 8 pages, 5 figures. Accepted by PRL.     Differential limit on the extremely-high-energy cosmic neutrino flux in the presence of astrophysical background from nine years of IceCube data

    We report a quasi-differential upper limit on the extremely-high-energy (EHE) neutrino flux above 5×1065\times 10^{6} GeV based on an analysis of nine years of IceCube data. The astrophysical neutrino flux measured by IceCube extends to PeV energies, and it is a background flux when searching for an independent signal flux at higher energies, such as the cosmogenic neutrino signal. We have developed a new method to place robust limits on the EHE neutrino flux in the presence of an astrophysical background, whose spectrum has yet to be understood with high precision at PeV energies. A distinct event with a deposited energy above 10610^{6} GeV was found in the new two-year sample, in addition to the one event previously found in the seven-year EHE neutrino search. These two events represent a neutrino flux that is incompatible with predictions for a cosmogenic neutrino flux and are considered to be an astrophysical background in the current study. The obtained limit is the most stringent to date in the energy range between 5×1065 \times 10^{6} and 5×10105 \times 10^{10} GeV. This result constrains neutrino models predicting a three-flavor neutrino flux of $E_\nu^2\phi_{\nu_e+\nu_\mu+\nu_\tau}\simeq2\times 10^{-8}\ {\rm GeV}/{\rm cm}^2\ \sec\ {\rm sr}at at 10^9\ {\rm GeV}$. A significant part of the parameter-space for EHE neutrino production scenarios assuming a proton-dominated composition of ultra-high-energy cosmic rays is excluded.Comment: The version accepted for publication in Physical Review

    The IceCube Data Acquisition System: Signal Capture, Digitization, and Timestamping

    IceCube is a km-scale neutrino observatory under construction at the South Pole with sensors both in the deep ice (InIce) and on the surface (IceTop). The sensors, called Digital Optical Modules (DOMs), detect, digitize and timestamp the signals from optical Cherenkov-radiation photons. The DOM Main Board (MB) data acquisition subsystem is connected to the central DAQ in the IceCube Laboratory (ICL) by a single twisted copper wire-pair and transmits packetized data on demand. Time calibration is maintained throughout the array by regular transmission to the DOMs of precisely timed analog signals, synchronized to a central GPS-disciplined clock. The design goals and consequent features, functional capabilities, and initial performance of the DOM MB, and the operation of a combined array of DOMs as a system, are described here.
