Exosomes neutralize synaptic-plasticity-disrupting activity of AÎČ assemblies in vivo
Background: Exosomes, small extracellular vesicles of endosomal origin, have been suggested to be involved in both the metabolism and aggregation of Alzheimerâs disease (AD)-associated amyloid ÎČ-protein (AÎČ). Despite their ubiquitous presence and the inclusion of components which can potentially interact with AÎČ, the role of exosomes in regulating synaptic dysfunction induced by AÎČ has not been explored. Results: We here provide in vivo evidence that exosomes derived from N2a cells or human cerebrospinal fluid can abrogate the synaptic-plasticity-disrupting activity of both synthetic and AD brain-derived AÎČ. Mechanistically, this effect involves sequestration of synaptotoxic AÎČ assemblies by exosomal surface proteins such as PrPC rather than AÎČ proteolysis. Conclusions: These data suggest that exosomes can counteract the inhibitory action of AÎČ, which contributes to perpetual capability for synaptic plasticity
Structure and Interactions of Human IgG-Fc
This thesis involves structure and interaction studies of the Fc fragment of human IgG. For this purpose, hIgG-Fc of different subclasses were cloned and expressed in the eukaryotic host Pichia pastoris, where relevant protein modification at the post-translational level can be obtained. Sometimes, changes in pH, temperature and salt concentration or addition of moderate amounts of denaturants to a protein solution are associated with the protein forming non-natively folded states, such as the molten globule or the A state. IgG and some parts thereof are capable of forming another, so called alternatively folded state, usually induced by acidification in the presence of anions. This state is in many aspects related to the molten globule and the A state but with distinguishing properties related mainly to chemical stability and formation of oligomeric structures. The first part of this thesis describes two different alternatively folded states of hIgG-Fc of subclass 4. One of them was induced by decreasing the pH of the protein solution. Observed structural changes were highly dependent on the concentration of sodium chloride. The alternatively folded protein showed drastic changes in its secondary structure compared to the native protein and significant tertiary structure was lost. Moreover, it displayed an apparently increased chemical stability and had surface exposed hydrophobic patches resulting in the formation of higher order assemblies. In addition, it was shown for the first time that thermal induction of an alternatively folded state is also possible, with similar, but not identical, properties as the acid-induced state. Heat incubation for 20 hours at neutral pH and at a physiological salt concentration further resulted in the formation of protein aggregates. The dye Congo red had affinity for these aggregates, and when viewed under polarized light, it showed green birefringence. They also displayed binding of Thioflavin T and had a typical fibril appearance in the transmission electron microscope. Hence, the formed aggregates share key properties with structures constituting amyloid. The second part of this thesis is focused on interactions of the Fc-fragment with respect to both Fcγ-receptors on monocytes and the IgG autoantibody rheumatoid factor. Immune complexes and their binding to Fcγ-receptors are of pathogenic interest to rheumatoid arthritis. A surface mimic presenting full IgG molecules was designed as an in vitro immune complex model. Utilizing self-assembled monolayers composed of alkanethiolates with different chemical functionalities, the lateral IgG density could be tuned, enabling control of monocyte interaction with the surface. Importantly, the IgG molecules were homogeneously oriented to expose the Fc-fragment. The protein repellent properties of these surfaces ensured that only differences in IgG concentration determined variations in cellular adhesion. In a separate study the specificities of IgG rheumatoid factor with respect to the different subclasses of hIgG-Fc were investigated, using sera from patients with early rheumatoid arthritis. Strikingly high IgG-RF reactivity against hIgG2-Fc was observed, together with raised levels against hIgG1-Fc and hIgG4-Fc. No reactivity against hIgG3-Fc was found

As Fate Would Have It : A corpus-based study of Fate from an American perspective
Abstract This essay is based on an investigation carried out with the help of the Corpus of Contemporary American English (COCA). Taking the system of transitivity as its theoretical base and using spoken and written discourse as its primary source, this study aspired to find out what kinds of actions Americans perceive Fate to perform, for example physical, mental or verbal, in order to control what happens to people. It also aimed to reveal what actions people are said to perform in their attempts to control Fate. Do Americans deem Fate capable of, for instance, âdecidingâ, âtalkingâ or âconspiringâ and do they say that people, for example, âchallengeâ, âdefyâ or âdefeatâ Fate? Furthermore, a comparison was made in terms of the actions performed by Fate and people between the different domains of discourse represented in the corpus: spoken, fiction, magazine, newspaper and academic. Among other things, this investigation shows that in American discourse both Fate and people are perceived to resort to physical strategies rather than mental or verbal ones in their endeavor to control one another.
En studie av lÀrares uppfattningar av andrasprÄkselevers attityder till skrivande samt deras syn pÄ undervisningav andrasprÄkselever
Uppsatsens syfte var att undersöka vilken uppfattning nÄgra gymnasielÀrare har om andrasprÄkselevers attityder till skriftsprÄket och jÀmföra dessa med de svar sex andrasprÄkselever givit i en tidigare genomförd studie (se Kanmert 2011). Studien syftade Àven till att undersöka lÀrares syn pÄ, och attityder till, undervisning av andrasprÄkselever samt hur de bedömer elevernas förutsÀttningar att uppnÄ angivna kursmÄl. Studien har en kvalitativ ansats med fenomenografisk inriktning. Materialet samlades in genom intervjuenkÀter innehÄllande öppna frÄgor samt flervalsfrÄgor. Resultatet visade att lÀrarnas uppfattning till största delen skilde sig frÄn andrasprÄkselevernas. Det framgick ocksÄ att ÀmneslÀrare tenderar att skjuta över upplevda svÄrigheter pÄ eleverna. LikasÄ föreföll det som om en del ÀmneslÀrare Àr ovilliga att anpassa sin Àmnesundervisning för att kunna möta andrasprÄkselevers behov liksom att majoriteten av informanter upplevde svÄrigheter i samband med undervisning av andrasprÄkselever. Endast ett fÄtal lÀrare ansÄg att deras andrasprÄkselever hade goda förutsÀttningar att uppnÄ angivna kursmÄl i deras respektive Àmnen. De slutsatser som kunde dras av studien var att lÀrare troligen behöver samtala och interagera mer med sina andrasprÄkselever för att kunna skapa bÀttre relationer till dem samt att lÀrare behöver bli medvetna om sin egen roll vad gÀller upplevda svÄrigheter i andrasprÄksundervisningen. LikasÄ verkar andrasprÄkslÀrare och ÀmneslÀrare ha olika syn pÄ vilken kunskapsnivÄ i svenska andrasprÄkselever bör ha uppnÄtt nÀr de börjar gymnasiet. Slutligen konstaterades att det krÀvs en förÀndring om skolan ska kunna leva upp till föreskrivna demokratiska vÀrderingar; skolan bör ges mer resurser i form av personal och skolmateriel. Den ojÀmlika tandstatusens livsförlopp : En longitudinell studie av sambanden mellan socioekonomisk position tidigare i livet och tandlöshet före och efter pensionsÄldern.
Syftet med studien Àr att undersöka om och hur socioekonomisk position samvarierar med tandlöshet före och efter pensionsÄldern samt med försÀmrad tandstatus. Datamaterialet som anvÀnds Àr LevnadsnivÄundersökningen (LNU) 1968 och 1981 och Undersökningen om Àldres levnadsvillkor (SWEOLD) 1991, 2002 och 2004 och i studiepopulationen ingÄr personer som intervjuats i LNU och SWEOLD och som var 69-88 Är vid tiden för uppföljning. Datamaterialet analyseras med logistiska regressioner. Analyserna visar att lÀgre socioekonomisk position ökar risken för att vara tandlös före och efter pensionsÄldern och för att fÄ försÀmrad tandstatus under denna period. Egen utbildning och socialklass har samtidigt samband med tandlöshet före respektive efter pensionsÄldern, med nÄgot starkare samband mellan utbildning och tandlöshet, medan försÀmrad tandstatus enbart uppvisar samband med socialklass. Analyserna visar att socioekonomisk position som vuxen har större betydelse Àr socioekonomisk position som barn med ocksÄ att socioekonomiska förhÄllanden kan ha betydelse för tandstatus under hela livet. Resultatet visar pÄ behovet av insatser för att minska de sociala skillnaderna i tandstatus under hela livsförloppet dÀr en betydelsefull insats kan vara att frÀmja tillgÄngen till tandvÄrd för de med störst behov. Resultatet visar ocksÄ pÄ stora skillnader i tandstatus mellan olika kohorter. För att frÀmja en god tandhÀlsa och ytterligare minska kohortskillnaderna behövs tidiga insater, redan i barndomen. Ytterligare studier behövs för att bÀttre förklara de sociala strukturerna som pÄverkar tandlöshet
The Borderland of Typography : About Gender Norms in Change
The earlier studies of how gender is visually constructed in lifestyle magazines have mainly been done on photographs. But, photographs are only one part of the entire magazine Therefor3, we saw the importance in examining one of the other big parts, typography. We have been studying if gender can be constructed in lifestyle magazines through the shape and color of typography and the color context. It is important to point out that we have not been studying the written word but only the shape and color of the typography. The study has been conducted through a qualitative content analysis with a comparative process of the selected magazines. The analysis began with a semiotic analysis of selected parts of the magazines. The result was then analyzed with a feminist theoretical approach. The result of the study shows that gender can be constructed through typography in a number of different ways. This depends on what feminist approach it is looked upon. In one way gender is constructed in a traditional perspective where the boundaries of masculinity and femininity is constrained to stereotypes. In another way it is constructed through a modern perspective where the boundaries of gender is no longer fixed
Performance measurement in complex projects : â A case study at Saab Aeronautics
Organisationer med komplexa projekt som straÌcker sig oÌver ett flertal aÌr finner svaÌrigheter att foÌrhaÌlla sig till projektens foÌrdefinierade parametrar tid, kostnad och omfattning. Skenande kostnader och foÌrseningar aÌr vanligt foÌrekommande i dessa typer av projekt, daÌ projekten omfattar ett samspel av ett flertal aktiviteter och daÌr projektets omfattning ofta foÌraÌndras. Det finns metoder som verksamheter kan tillaÌmpa foÌr att kontrollera ovannaÌmnda parametrar och i ett tidigt skede kunna identifiera naÌr naÌgon av parametrar tenderar att oÌverskridas. En metod som styrker prestationsmaÌtningar aÌr Earned Value Mangament, EVM. Syftet med studien var att identifiera vart organisationer brister gentemot branschstandarden inom EVM, ANSI/EIA-748. Standarden innehaÌller viktiga steg foÌr att kunna utfoÌra en korrekt prestationsmaÌtning tillsammans med teorin fraÌn Earned Value Management. Resultat studien presentera indikerar paÌ att organisationer maÌste skapa tydliga riktlinjer hur ANSI/EIA-748 skall tillaÌmpas. Till exempel att alla bryter ner arbetet och foÌrdelar sina budgetar paÌ samma saÌtt. Studiens resultat antyder ocksaÌ att Rolling Wave aÌr ett laÌmpligt verktyg foÌr komplexa och laÌnga projekt. Av den anledningen att projektledare inte planerar foÌr mycket i onoÌdan eftersom projektets omfattning staÌndigt foÌraÌndras
