    The Role of Growth Retardation in Lasting Effects of Neonatal Dexamethasone Treatment on Hippocampal Synaptic Function

    BACKGROUND: Dexamethasone (DEX), a synthetic glucocorticoid, is commonly used to prevent or lessen the morbidity of chronic lung disease in preterm infants. However, evidence is now increasing that this clinical practice negatively affects somatic growth and may result in long-lasting neurodevelopmental deficits. We therefore hypothesized that supporting normal somatic growth may overcome the lasting adverse effects of neonatal DEX treatment on hippocampal function. METHODOLOGY/PRINCIPAL FINDINGS: To test this hypothesis, we developed a rat model using a schedule of tapering doses of DEX similar to that used in premature infants and examined whether the lasting influence of neonatal DEX treatment on hippocampal synaptic plasticity and memory performance are correlated with the deficits in somatic growth. We confirmed that neonatal DEX treatment switched the direction of synaptic plasticity in hippocampal CA1 region, favoring low-frequency stimulation- and group I metabotropic glutamate receptor agonist (S)-3,5,-dihydroxyphenylglycine-induced long-term depression (LTD), and opposing the induction of long-term potentiation (LTP) by high-frequency stimulation in the adolescent period. The effects of DEX on LTP and LTD were correlated with an increase in the autophosphorylation of Ca(2+)/calmodulin-dependent protein kinase II at threonine-286 and a decrease in the protein phosphatase 1 expression. Neonatal DEX treatment resulted in a disruption of memory retention subjected to object recognition task and passive avoidance learning. The adverse effects of neonatal DEX treatment on hippocampal synaptic plasticity and memory performance of the animals from litters culled to 4 pups were significantly less than those for the 8-pup litters. However, there was no significant difference in maternal care between groups. CONCLUSION/SIGNIFICANCE: Our results demonstrate that growth retardation plays a crucial role in DEX-induced long-lasting influence of hippocampal function. Our findings suggest that therapeutic strategies designed to support normal development and somatic growth may exert beneficial effects to reduce lasting adverse effects following neonatal DEX treatment

    Psychological Functioning and Disease-Related Quality of Life in Pediatric Patients With an Implantable Cardioverter Defibrillator

    The objective of this multicenter study was to evaluate psychological functioning and disease-related quality of life (DRQoL) in pediatric patients with an implantable cardioverter defibrillator (ICD) in The Netherlands. Thirty patients were investigated; the mean age was 16.3Ā years, and the mean duration of implantation was 3.6Ā years. To assess psychological problems, three domains of the Symptom Checklist (SCL-90-R) were administered to the 25 patientsĀ >13Ā years old. DRQoL was assessed with a disease-specific pediatric questionnaire, the short-form 11-item Worries About (WA)ICDs Scale. PatientsĀ ā‰„13Ā years old scored significantly higher than the reference group on the domains of anxiety, depression, and sleeping problems of the SCL-90-R (TĀ =Ā 7.5, pĀ <Ā 0.001; TĀ =Ā 5.4, pĀ <Ā 0.001; and TĀ =Ā 7.8, pĀ <Ā 0.001, respectively). Patients who had received an (in)appropriate shock reported more depressive symptoms (TĀ =Ā 2.1, pĀ <Ā 0.03). Patients withĀ >2Ā years implant duration (NĀ =Ā 19) or who had received an (in)appropriate shock (NĀ =Ā 13) showed lower DRQoL scores on the modified WAICD (TĀ =Ā 2.1, pĀ <Ā 0.04; TĀ =Ā 2.1, pĀ <Ā 0.5, respectively). Age at implantation or underlying disease did not influence psychological problems or DRQoL. Young ICD patients showed more anxiety, depression, and sleeping disorders. Worries were increased among patients with ICD shocks and in those who had their ICD implanted forĀ >2Ā years. To determine psychological problems and help children to learn to cope with shocks, proper guidance and monitoring of young ICD patients are recommended

    Although type I interferons (IFNs) were first described almost 60 years ago, the ability to monitor and modulate the functional activities of the individual IFN subtypes that comprise this family has been hindered by a lack of reagents. The major type I IFNs, IFN-Ī² and the multiple subtypes of IFN-Ī±, are expressed widely and induce their effects on cells by interacting with a shared heterodimeric receptor (IFNAR). In the mouse, the physiologic actions of IFN-Ī± and IFN-Ī² have been defined using polyclonal anti-type I IFN sera, by targeting IFNAR using monoclonal antibodies or knockout mice, or using Ifnb-/- mice. However, the corresponding analysis of IFN-Ī± has been difficult because of its polygenic nature. Herein, we describe two monoclonal antibodies (mAbs) that differentially neutralize murine IFN-Ī² or multiple subtypes of murine IFN-Ī±. Using these mAbs, we distinguish specific contributions of IFN-Ī² versus IFN-Ī± in restricting viral pathogenesis and identify IFN-Ī± as the key mediator of the antiviral response in mice infected with West Nile virus. This study thus suggests the utility of these new reagents in dissecting the antiviral and immunomodulatory roles of IFN-Ī² versus IFN-Ī± in murine models of infection, immunity, and autoimmunity

    Dopaminergic modulation of affective and social deficits induced by prenatal glucocorticoid exposure

    Prenatal stress or exposure to elevated levels of glucocorticoids (GCs) can impair specific neurobehavioral circuits leading to alterations in emotional processes later in life. In turn, emotional deficits may interfere with the quality and degree of social interaction. Here, by using a comprehensive behavioral approach in combination with the measurement of ultrasonic vocalizations, we show that in utero GC (iuGC)-exposed animals present increased immobility in the forced swimming test, pronounced anhedonic behavior (both anticipatory and consummatory), and an impairment in social interaction at different life stages. Importantly, we also found that social behavioral expression is highly dependent on the affective status of the partner. A profound reduction in mesolimbic dopaminergic transmission was found in iuGC animals, suggesting a key role for dopamine (DA) in the etiology of the observed behavioral deficits. Confirming this idea, we present evidence that a simple pharmacological approachā€”acute L-3,4-dihydroxyphenylacetic acid (L-DOPA) oral administration, is able to normalize DA levels in iuGC animals, with a concomitant amelioration of several dimensions of the emotional and social behaviors. Interestingly, L-DOPA effects in control individuals were not so straightforward; suggesting that both hypo- and hyperdopaminergia are detrimental in the context of such complex behaviors.This work was supported by a grant of Institute for the Study of Affective Neuroscience (ISAN) and Janssen Neurosciences Prize.     A histone acetylome-wide association study of Alzheimerā€™s disease identifies disease-associated H3K27ac differences in the entorhinal cortex

    We quantified genome-wide patterns of lysine H3K27 acetylation (H3K27ac) in entorhinal cortex samples from Alzheimerā€™s disease (AD) cases and matched controls using chromatin immunoprecipitation and highly parallel sequencing. We observed widespread acetylomic variation associated with AD neuropathology, identifying 4,162 differential peaks (false discovery rateā€‰<ā€‰0.05) between AD cases and controls. Differentially acetylated peaks were enriched in disease-related biological pathways and included regions annotated to genes involved in the progression of amyloid-Ī² and tau pathology (for example, APP, PSEN1, PSEN2, and MAPT), as well as regions containing variants associated with sporadic late-onset AD. Partitioned heritability analysis highlighted a highly significant enrichment of AD risk variants in entorhinal cortex H3K27ac peak regions. AD-associated variable H3K27ac was associated with transcriptional variation at proximal genes including CR1, GPR22, KMO, PIM3, PSEN1, and RGCC.     In het rapport zijn de resultaten voor 1997 weergegeven van het monitoringprogramma voor prioritaire afvalstoffen dat in opdracht van de Directie Afvalstoffen van het Directoraat-Generaal Milieubeheer door het RIVM wordt uitgevoerd. In het rapport zijn de rapportages van 27 prioritaire afvalstoffen gebundeld. Per afvalstof worden de belangrijkste gegevens betreffende de hoeveelheden (onder meer naar herkomst en verwerkingsmethode) en samenstelling volgens een standaard opzet gepresenteerd en toegelicht. In de toelichting bij de gegevens zijn de gebruikte bronnen en werkwijzen (eerder vastgelegd in meetprotocollen) beschreven.The report presents the results for the year 1997 of the monitoring programme on priority waste streams in the Netherlands. This programme is being carried out by RIVM by order of the Directorate of Waste Management Policy of the Ministry of Housing, Spatial Planning and the Environment. Basic data and additional information on the quantities, specified into origin and method of disposal, as well as the composition, are presented in a standard format for 27 priority waste streams. The sources of information and procedures used to determine the data are explaned.DG
