Electronic structure and Jahn-Teller effect in GaN:Mn and ZnS:Cr
We present an ab-initio and analytical study of the Jahn-Teller effect in two
diluted magnetic semiconductors (DMS) with d4 impurities, namely Mn-doped GaN
and Cr-doped ZnS. We show that only the combined treatment of Jahn-Teller
distortion and strong electron correlation in the 3d shell may lead to the
correct insulating electronic structure. Using the LSDA+U approach we obtain
the Jahn-Teller energy gain in reasonable agreement with the available
experimental data. The ab-initio results are completed by a more
phenomenological ligand field theory.Comment: 15 pages, 5 figure

Fire debris analysis by Raman spectroscopy and chemometrics
A paper reporting the use of Raman Spectroscopy in fire debris analysis is presented. Five polymer based samples, namely carpet (polypropylene), nylon stockings (nylon), foam packaging (polystyrene), CD cases (polystyrene) and DVD cases (polypropylene) were burnt with each one of the following ignitable liquids: petrol, diesel, kerosene and ethanol. Raman shifts were obtained and, in some cases, peaks were identified to correspond to pyrolysis products in the form of alkanes, aromatic or polyaromatic compounds. All pyrolysis peaks were used to produce a Principal Component Analysis (PCA) of the burned samples with the different ignitable liquids. The change in the Raman spectra made it possible to identify some of the pyrolysis products produced in the combustion and also to identify the different plastic materials in fire debris, even when different fuels have been used and the chemical and structural identity of the plastic has been altered in the fire
Andersen-Tawil syndrome: report of 3 novel mutations and high risk of symptomatic cardiac involvement.
IntroductionAndersen-Tawil syndrome (ATS) is a potassium channelopathy affecting cardiac and skeletal muscle. Periodic paralysis is a presenting symptom in some patients, whereas, in others, symptomatic arrhythmias or prolongation of QT in echocardiographic recordings will lead to diagnosis of ATS. Striking intrafamilial variability of expression of KCNJ2 mutations and rarity of the syndrome may lead to misdiagnosis.MethodsWe report 15 patients from 8 Polish families with ATS, including 3 with novel KCNJ2 mutations.ResultsAll patients had dysmorphic features; periodic paralysis affected males more frequently than females (80% vs. 20%), and most attacks were normokalemic. Two patients (with T75M and T309I mutations) had aborted sudden cardiac death. An implantable cardioverter-defibrillator was utilized in 40% of cases.ConclusionsKCNJ2 mutations cause a variable phenotype, with dysmorphic features seen in all patients studied, a high penetrance of periodic paralysis in males and ventricular arrhythmia with a risk of sudden cardiac death
Amino acid substitution equivalent to human chorea-acanthocytosis I2771R in yeast Vps13 protein affects its binding to phosphatidylinositol 3-phosphate
The rare human disorder chorea-acanthocytosis (ChAc) is caused by mutations in hVPS13A gene. The hVps13A protein interacts
with actin and regulates the level of phosphatidylinositol 4-phosphate (PI4P) in the membranes of neuronal cells. Yeast
Vps13 is involved in vacuolar protein transport and, like hVps13A, participates in PI4P metabolism. Vps13 proteins are conserved
in eukaryotes, but their molecular function remains unknown. One of the mutations found in ChAc patients causes
amino acids substitution I2771R which affects the localization of hVps13A in skeletal muscles. To dissect the mechanism of
pathogenesis of I2771R, we created and analyzed a yeast strain carrying the equivalent mutation. Here we show that in yeast,
substitution I2749R causes dysfunction of Vps13 protein in endocytosis and vacuolar transport, although the level of the protein
is not affected, suggesting loss of function. We also show that Vps13, like hVps13A, influences actin cytoskeleton organization
and binds actin in immunoprecipitation experiments. Vps13-I2749R binds actin, but does not function in the actin
cytoskeleton organization. Moreover, we show that Vps13 binds phospholipids, especially phosphatidylinositol 3-phosphate
(PI3P), via its SHR_BD and APT1 domains. Substitution I2749R attenuates this ability. Finally, the localization of Vps13-GFP is
altered when cellular levels of PI3P are decreased indicating its trafficking within the endosomal membrane system. These results
suggest that PI3P regulates the functioning of Vps13, both in protein trafficking and actin cytoskeleton organization.
Attenuation of PI3P-binding ability in the mutant hVps13A protein may be one of the reasons for its mislocalization and disrupted
function in cells of patients suffering from ChAc
