    Intracellular labeling of live cells via transient membrane permeabilization for fluorescence and super resolution microscopy

    Fluorescence imaging of intracellular proteins is often achieved by using transfection-induced expression of fluorescent protein. This can potentially impose artifacts such as loss of function or over-expression of target proteins. Direct labeling of the intracellular protein is an alternative to transfection, but is largely limited by permeability of the fluorescent probes. Here, we have developed a high-throughput technique for labeling intracellular proteins of living cells. The technique makes use of Streptolysin O (SLO), a bacterial enzyme that permeabilizes cells to DNA, RNA, proteins. We show that SLO can be used to deliver a variety of fluorescent probes; ranging from organic dyes (<1 kDa in size) to fluorescent immunoglobulin antibody (~150 kDa) for specific labeling of intracellular proteins. We demonstrate in numerous ways that after permeabilization the cells remained viable and responded normally to cell signaling protein. We applied this technique to observe the dynamic motion of labeled actin and mitochondria using super-resolution fluorescence microscopy (dSTORM). Furthermore, we show the ability to image single proteins inside the living cell by tracking single molecules of kinesin using photostable probes

    Soft-sediment deformation structures related to volcanic earthquakes of the Lower Cretaceous Qingshan Group in Lingshan Island, Shandong Province, East China

    Abstract The study on soft-sediment deformation structures (SSDS) of Lingshan Island has been one of the hot topics of sedimentology researches in China in recent years, and SSDS developed in turbidite system in the Laiyang Group are widely known by domestic researchers. However, few studies were conducted on the SSDS in fan delta system in the Qingshan Group, Lingshan Island. This study analyzes the classification and characteristics of SSDS especially their lithofacies association and lithologic characteristics through field outcrops investigation and thin section analysis as well. A conclusion was acquired that the paleoenvironment was a fan delta system with occurrence of several volcanic eruptions, where the water became gradually shallower. The SSDS types in the Qingshan Group includes load and flame structure, ball and pillow structure, water-escape structure, hydroplastic deformation structure, plastic sandstone breccia structure, volcanic drop stone and V-shaped ground fissure mainly caused by volcanic earthquakes of three types: (1) seismic waves, (2) gravity and inertia effect of pyroclastic flows, (3) instant differential air pressure; which is different from slumping and tectonic earthquakes occurred in the Laiyang Group. In addition, with the lithofacies association analysis between pyroclastic flow and SSDS beds, a distribution model of SSDS related to volcanic earthquakes can be established: SSDS types changed gradually with their distance further away from the volcanic activity core. Brittle deformation which was common in the proximal zone disappeared gradually; liquefied and plastic SSDS continued to dominate in the medial zone; and slightly liquefied SSDS were developed in the distal zone. Meanwhile, the scale and size of SSDS is negatively correlated with the distance of SSDS depositional locations from the volcanic vent

    Estuarine plastisphere as an overlooked source of N2O production

    “Plastisphere”, microbial communities colonizing plastic debris, has sparked global concern for marine ecosystems. Microbiome inhabiting this novel human-made niche has been increasingly characterized; however, whether the plastisphere holds crucial roles in biogeochemical cycling remains largely unknown. Here we evaluate the potential of plastisphere in biotic and abiotic denitrification and nitrous oxide (N2O) production in estuaries. Biofilm formation provides anoxic conditions favoring denitrifiers. Comparing with surrounding bulk water, plastisphere exhibits a higher denitrifying activity and N2O production, suggesting an overlooked N2O source. Regardless of plastisphere and bulk water, bacterial and fungal denitrifications are the main regulators for N2O production instead of chemodenitrification. However, the contributions of bacteria and fungi in the plastisphere are different from those in bulk water, indicating a distinct N2O production pattern in the plastisphere. These findings pinpoint plastisphere as a N2O source, and provide insights into roles of the new biotope in biogeochemical cycling in the Anthropocene

    Clonal spread of multidrug-resistant Acinetobacter baumannii in eastern Taiwan

    Background and PurposeThis study was conducted to investigate the molecular epidemiology and antimicrobial susceptibility of multidrug-resistant (MDR) Acinetobacter baumannii to three types of antibiotics.MethodsOne hundred and thirty-four specimens of MDR A baumannii were collected from three branches (Taipei, Dalin, and Hualien branches) of Buddhist Tzu Chi Hospital, which are located in northern, southern, and eastern Taiwan, during 2007. Genotyping was performed by pulsed-field gel electrophoresis. Antibiotic susceptibilities to colistin, rifampicin, and tigecycline were determined. The synergistic effects of rifampin and colistin were also evaluated.ResultsAntibiotic susceptibility testing showed that 10.4%, 47.8% and 45.5% of the MDR A baumannii isolates are resistant to colistin, rifampicin, and tigecycline, respectively. A majority of the rifampicin-resistant isolates (62.7%) were found in the Haulien branch, whereas 62.2% of tigecycline-resistant isolates were found in the Taipei branch. The combination of colistin and rifampicin had a synergistic effect on all of the isolates. Genotyping by pulsed-field gel electrophoresis identified 17, 23, and 11 pulsotypes in the Taipei, Dalin, and Haulien branches, respectively. Furthermore, 74.5% of isolates in the Haulien branch were identified as one of three pulsotypes. Among 37 rifampicin-resistant and 22 tigecycline-resistant MDR A baumannii isolates found in the Haulien branch, 51.3% (19/37) and 50% (11/22) of the isolates belonged to the same clone, respectively.ConclusionThis study confirms the high prevalence of resistance to rifampicin and tigecycline in MDR A baumannii in the three hospitals that were studied, and the high proportion of identical strains that exist in eastern Taiwan

    Metabolic syndrome and abdominal fat are associated with inflammation, but not with clinical outcomes, in peritoneal dialysis patients

    BACKGROUND: In the general population, metabolic syndrome (MetS) is correlated with visceral fat and a risk factor for cardiovascular disease (CVD); however, little is known about the significance of abdominal fat and its association with inflammation and medication use in peritoneal dialysis (PD) patients. We investigated the relationship of visceral fat area (VFA) with C-reactive protein (CRP) levels and medication use in PD patients and followed their clinical outcomes. METHODS: In a prospective study from February 2009 to February 2012, we assessed diabetes mellitus (DM) status, clinical and PD-associated characteristics, medication use, CRP levels, components of MetS, and VFA in 183 PD patients. These patients were categorized into 3 groups based on MetS and DM status: non-MetS (group 1, n = 73), MetS (group 2, n = 65), and DM (group 3, n = 45). VFA was evaluated by computed tomography (CT) and corrected for body mass index (BMI). RESULTS: Patients in group 1 had smaller VFAs than patients in groups 2 and 3 (3.2 ± 1.8, 4.6 ± 1.9, and 4.9 ± 2.0 cm(2)/[kg/m(2)], respectively, P < 0.05) and lower CRP levels (0.97 ± 2.31, 1.27 ± 2.57, and 1.11 ± 1.35 mg/dL, respectively, P < 0.05). VFA increased with the number of criteria met for MetS. After adjusting for age, body weight, and sex, CRP and albumin levels functioned as independent positive predictors of VFA; on other hand, the use of renin-angiotensin system blockers was inversely correlated with VFA in PD patients without DM. In the survival analysis, DM patients (group 3) had the poorest survival among the 3 groups, but no significant differences were found between groups 1 and 2. CONCLUSION: This study showed that VFA and MetS are associated with CRP levels but cannot predict survival in PD patients without DM. The complex relationship of nutritional parameters to VFA and MetS may explain these results. The type of antihypertensive medication used was also associated with the VFA. The mechanisms behind these findings warrant further investigation

    Field test of a practical secure communication network with decoy-state quantum cryptography

    We present a secure network communication system that operated with decoy-state quantum cryptography in a real-world application scenario. The full key exchange and application protocols were performed in real time among three nodes, in which two adjacent nodes were connected by approximate 20 km of commercial telecom optical fiber. The generated quantum keys were immediately employed and demonstrated for communication applications, including unbreakable real-time voice telephone between any two of the three communication nodes, or a broadcast from one node to the other two nodes by using one-time pad encryption.Comment: 10 pages, 2 figures, 2 tables, typos correcte
