Optical detection of single non-absorbing molecules using the surface plasmon of a gold nanorod
Current optical detection schemes for single molecules require light
absorption, either to produce fluorescence or direct absorption signals. This
severely limits the range of molecules that can be detected, because most
molecules are purely refractive. Metal nanoparticles or dielectric resonators
detect non-absorbing molecules by a resonance shift in response to a local
perturbation of the refractive index, but neither has reached single-protein
sensitivity. The most sensitive plasmon sensors to date detect single molecules
only when the plasmon shift is amplified by a highly polarizable label or by a
localized precipitation reaction on the particle's surface. Without
amplification, the sensitivity only allows for the statistical detection of
single molecules. Here we demonstrate plasmonic detection of single molecules
in realtime, without the need for labeling or amplification. We monitor the
plasmon resonance of a single gold nanorod with a sensitive photothermal assay
and achieve a ~ 700-fold increase in sensitivity compared to state-of-the-art
plasmon sensors. We find that the sensitivity of the sensor is intrinsically
limited due to spectral diffusion of the SPR. We believe this is the first
optical technique that detects single molecules purely by their refractive
index, without any need for photon absorption by the molecule. The small size,
bio-compatibility and straightforward surface chemistry of gold nanorods may
open the way to the selective and local detection of purely refractive proteins
in live cells
Controlling Curie temperature in (Ga,Ms)As through location of the Fermi level within the impurity band
The ferromagnetic semiconductor (Ga,Mn)As has emerged as the most studied
material for prototype applications in semiconductor spintronics. Because
ferromagnetism in (Ga,Mn)As is hole-mediated, the nature of the hole states has
direct and crucial bearing on its Curie temperature TC. It is vigorously
debated, however, whether holes in (Ga,Mn)As reside in the valence band or in
an impurity band. In this paper we combine results of channeling experiments,
which measure the concentrations both of Mn ions and of holes relevant to the
ferromagnetic order, with magnetization, transport, and magneto-optical data to
address this issue. Taken together, these measurements provide strong evidence
that it is the location of the Fermi level within the impurity band that
determines TC through determining the degree of hole localization. This finding
differs drastically from the often accepted view that TC is controlled by
valence band holes, thus opening new avenues for achieving higher values of TC.Comment: 5 figures, supplementary material include
Nanoantenna-enhanced ultrafast nonlinear spectroscopy of a single gold nanoparticle
Optical nanoantennas are a novel tool to investigate previously unattainable
dimensions in the nanocosmos. Just like their radio-frequency equivalents,
nanoantennas enhance the light-matter interaction in their feed gap. Antenna
enhancement of small signals promises to open a new regime in linear and
nonlinear spectroscopy on the nanoscale. Without antennas especially the
nonlinear spectroscopy of single nanoobjects is very demanding. Here, we
present for the first time antenna-enhanced ultrafast nonlinear optical
spectroscopy. In particular, we utilize the antenna to determine the nonlinear
transient absorption signal of a single gold nanoparticle caused by mechanical
breathing oscillations. We increase the signal amplitude by an order of
magnitude which is in good agreement with our analytical and numerical models.
Our method will find applications in linear and nonlinear spectroscopy of
nanoobjects, ranging from single protein binding events via nonlinear tensor
elements to the limits of continuum mechanics
Refractive-index sensing with ultra-thin plasmonic nanotubes
We study the refractive-index sensing properties of plasmonic nanotubes with
a dielectric core and ultra-thin metal shell. The few-nm thin metal shell is
described by both the usual Drude model and the nonlocal hydrodynamic model to
investigate the effects of nonlocality. We derive an analytical expression for
the extinction cross section and show how sensing of the refractive index of
the surrounding medium and the figure-of-merit are affected by the shape and
size of the nanotubes. Comparison with other localized surface plasmon
resonance sensors reveals that the nanotube exhibits superior sensitivity and
comparable figure-of-merit
Home parenteral nutrition with an omega-3-fatty-acid-enriched MCT/LCT lipid emulsion in patients with chronic intestinal failure (the HOME study):study protocol for a randomized, controlled, multicenter, international clinical trial
BACKGROUND: Home parenteral nutrition (HPN) is a life-preserving therapy for patients with chronic intestinal failure (CIF) indicated for patients who cannot achieve their nutritional requirements by enteral intake. Intravenously administered lipid emulsions (ILEs) are an essential component of HPN, providing energy and essential fatty acids, but can become a risk factor for intestinal-failure-associated liver disease (IFALD). In HPN patients, major effort is taken in the prevention of IFALD. Novel ILEs containing a proportion of omega-3 polyunsaturated fatty acids (n-3 PUFA) could be of benefit, but the data on the use of n-3 PUFA in HPN patients are still limited. METHODS/DESIGN: The HOME study is a prospective, randomized, controlled, double-blind, multicenter, international clinical trial conducted in European hospitals that treat HPN patients. A total of 160 patients (80 per group) will be randomly assigned to receive the n-3 PUFA-enriched medium/long-chain triglyceride (MCT/LCT) ILE (Lipidem/Lipoplus® 200 mg/ml, B. Braun Melsungen AG) or the MCT/LCT ILE (Lipofundin® MCT/LCT/Medialipide® 20%, B. Braun Melsungen AG) for a projected period of 8 weeks. The primary endpoint is the combined change of liver function parameters (total bilirubin, aspartate transaminase and alanine transaminase) from baseline to final visit. Secondary objectives are the further evaluation of the safety and tolerability as well as the efficacy of the ILEs. DISCUSSION: Currently, there are only very few randomized controlled trials (RCTs) investigating the use of ILEs in HPN, and there are very few data at all on the use of n-3 PUFAs. The working hypothesis is that n-3 PUFA-enriched ILE is safe and well-tolerated especially with regard to liver function in patients requiring HPN. The expected outcome is to provide reliable data to support this thesis thanks to a considerable number of CIF patients, consequently to broaden the present evidence on the use of ILEs in HPN. Transit Timing and Duration Variations for the Discovery and Characterization of Exoplanets
Transiting exoplanets in multi-planet systems have non-Keplerian orbits which
can cause the times and durations of transits to vary. The theory and
observations of transit timing variations (TTV) and transit duration variations
(TDV) are reviewed. Since the last review, the Kepler spacecraft has detected
several hundred perturbed planets. In a few cases, these data have been used to
discover additional planets, similar to the historical discovery of Neptune in
our own Solar System. However, the more impactful aspect of TTV and TDV studies
has been characterization of planetary systems in which multiple planets
transit. After addressing the equations of motion and parameter scalings, the
main dynamical mechanisms for TTV and TDV are described, with citations to the
observational literature for real examples. We describe parameter constraints,
particularly the origin of the mass/eccentricity degeneracy and how it is
overcome by the high-frequency component of the signal. On the observational
side, derivation of timing precision and introduction to the timing diagram are
given. Science results are reviewed, with an emphasis on mass measurements of
transiting sub-Neptunes and super-Earths, from which bulk compositions may be

The potential impact of expanding target age groups for polio immunization campaigns
BACKGROUND: Global efforts to eradicate wild polioviruses (WPVs) continue to face challenges due to uninterrupted endemic WPV transmission in three countries and importation-related outbreaks into previously polio-free countries. We explore the potential role of including older children and adults in supplemental immunization activities (SIAs) to more rapidly increase population immunity and prevent or stop transmission. METHODS: We use a differential equation-based dynamic poliovirus transmission model to analyze the epidemiological impact and vaccine resource implications of expanding target age groups in SIAs. We explore the use of older age groups in SIAs for three situations: alternative responses to the 2010 outbreak in Tajikistan, retrospective examination of elimination in two high-risk states in northern India, and prospective and retrospective strategies to accelerate elimination in endemic northwestern Nigeria. Our model recognizes the ability of individuals with waned mucosal immunity (i.e., immunity from a historical live poliovirus infection) to become re-infected and contribute to transmission to a limited extent. RESULTS: SIAs involving expanded age groups reduce overall caseloads, decrease transmission, and generally lead to a small reduction in the time to achieve WPV elimination. Analysis of preventive expanded age group SIAs in Tajikistan or prior to type-specific surges in incidence in high-risk areas of India and Nigeria showed the greatest potential benefits of expanded age groups. Analysis of expanded age group SIAs in outbreak situations or to accelerate the interruption of endemic transmission showed relatively less benefit, largely due to the circulation of WPV reaching individuals sooner or more effectively than the SIAs. The India and Nigeria results depend strongly on how well SIAs involving expanded age groups reach relatively isolated subpopulations that sustain clusters of susceptible children, which we assume play a key role in persistent endemic WPV transmission in these areas. CONCLUSIONS: This study suggests the need to carefully consider the epidemiological situation in the context of decisions to use expanded age group SIAs. Subpopulations of susceptible individuals may independently sustain transmission, which will reduce the overall benefits associated with using expanded age group SIAs to increase population immunity to a sufficiently high level to stop transmission and reduce the incidence of paralytic cases
Konstruktivistische Ansätze in der Erwachsenenbildung und Weiterbildung
Theoretical approaches in the field of further education and advanced vocational training have to face multifaceted demands: the analysis of knowledge aquisition and knowledge transfer and its instructional support as well as the revealment of the mechanisms which influence further education on an organisational level in companies. This article describes, that herefore especially moderate constructivistic approaches are useful. After an introduction to the philosophical tradition of these approaches and important characteristics of adult learning, two examples of constructivistic models are being described particularly: The theory of situated learning environments and career counseling. Concluding it is shown, that a moderate constructive perspective fulfils important criteria for the theoretic modelling of further education processes.Theoretische Ansätze in der Erwachsenen- und insbesondere in der beruflichen Weiterbildung müssen sich vielfältigen Ansprüchen stellen: der Analyse des Wissenserwerbs und Wissenstransfers und seiner instruktionalen Unterstützung ebenso wie der Aufdeckung der Mechanismen, die in den Betrieben auf organisatorischer Ebene die Weiterbildung beeinflussen. In diesem Beitrag wird die Auffassung vertreten, daß dafür insbesondere liberalisierte konstruktivistische Ansätze gut geeignet sind. Nach einer Einführung in die philosophische Tradition dieser Ansätze und wichtiger Merkmale des Lernens im Erwachsenenalter werden zwei Beispiele konstruktivistischer Modelle genauer beschrieben: die Theorie situierter Lernumgebungen und das career counseling. Abschließend wird gezeigt, daß eine liberalisierte konstruktivistische Perspektive wichtige Kriterien für die theoretische Modellierung von Weiterbildungsprozessen erfüllt
Accurate Prediction of DnaK-Peptide Binding via Homology Modelling and Experimental Data
Molecular chaperones are essential elements of the protein quality control machinery that governs translocation and folding of nascent polypeptides, refolding and degradation of misfolded proteins, and activation of a wide range of client proteins. The prokaryotic heat-shock protein DnaK is the E. coli representative of the ubiquitous Hsp70 family, which specializes in the binding of exposed hydrophobic regions in unfolded polypeptides. Accurate prediction of DnaK binding sites in E. coli proteins is an essential prerequisite to understand the precise function of this chaperone and the properties of its substrate proteins. In order to map DnaK binding sites in protein sequences, we have developed an algorithm that combines sequence information from peptide binding experiments and structural parameters from homology modelling. We show that this combination significantly outperforms either single approach. The final predictor had a Matthews correlation coefficient (MCC) of 0.819 when assessed over the 144 tested peptide sequences to detect true positives and true negatives. To test the robustness of the learning set, we have conducted a simulated cross-validation, where we omit sequences from the learning sets and calculate the rate of repredicting them. This resulted in a surprisingly good MCC of 0.703. The algorithm was also able to perform equally well on a blind test set of binders and non-binders, of which there was no prior knowledge in the learning sets.
