    Agonistic Interventions into Public Commemorative Art:An Innovative Form of Counter-memorial Practice?

    In light of recent controversies around the removal or modification of public commemorative art, such as memorials and monuments, this paper interrogates the value of competing approaches to counter-memorial practice using the framework of agonistic memory. It argues that much counter-memorial practice today, as it relates to historical memory, is dominated by a “cosmopolitan” mode that fails to offer a convincing response to the rise of right-wing populism and its instrumentalization of conflicts over public commemorative art. The article investigates two case studies of counter-memorial interventions that focus on the memory of fascism in Europe today and seeks to identify and assess emergent agonistic practices

    Genome-Wide Distribution of RNA-DNA Hybrids Identifies RNase H Targets in tRNA Genes, Retrotransposons and Mitochondria

    During transcription, the nascent RNA can invade the DNA template, forming extended RNA-DNA duplexes (R-loops). Here we employ ChIP-seq in strains expressing or lacking RNase H to map targets of RNase H activity throughout the budding yeast genome. In wild-type strains, R-loops were readily detected over the 35S rDNA region, transcribed by Pol I, and over the 5S rDNA, transcribed by Pol III. In strains lacking RNase H activity, R-loops were elevated over other Pol III genes, notably tRNAs, SCR1 and U6 snRNA, and were also associated with the cDNAs of endogenous TY1 retrotransposons, which showed increased rates of mobility to the 5'-flanking regions of tRNA genes. Unexpectedly, R-loops were also associated with mitochondrial genes in the absence of RNase H1, but not of RNase H2. Finally, R-loops were detected on actively transcribed protein-coding genes in the wild-type, particularly over the second exon of spliced ribosomal protein genes

    Most eukaryotic protein-coding transcripts contain introns, which vary in number and position along the transcript body. Intron removal through pre-mRNA splicing is tightly linked to transcription by RNA polymerase II as it translocates along each gene. Here, we review recent evidence that transcription and splicing are functionally coupled. We focus on how RNA polymerase II elongation rates impact splicing through local regulation and transcriptional pausing within genes. Emerging concepts of how splicing-related changes in elongation might be achieved are highlighted. We place the interplay between transcription and splicing in the context of chromatin where nucleosome positioning influences elongation, and histone modifications participate directly in the recruitment of splicing regulators to nascent transcripts

    Service-Oriented Process Control with Grafchart and the Devices Profile for Web Services

    To fulfill increasing requirements within the manufacturing sector, highly flexible and adaptable automation systems are needed. It is desirable to have one integrated approach that stretches from the process planning phase, through the implementation phase and all the way to the phase for execution of the process control logics. One promising approach is to use the concepts of service-oriented architectures within automation, here referred to as SOA-AT. As service technology, DPWS has proved to be the most suitable for realizing service based communication on device level. The paper shows how Grafchart, a graphical language aimed for sequential control applications, can support the development of DPWS applications, and how Grafchart can be used for process modeling and execution in the planning and execution phase. This constitutes a unique framework for the development and execution of SOA applications in accordance with the requirements for automatic control tasks. The paper also presents an industry-related experimental setup in which the SOA-AT concepts are demonstrated through the use of Grafchart
