    A simulated investigation on the machining instability and dynamic surface generation

    In this paper, the authors propose the generic concept of machining instability based on the analysis of all kinds of machining instable behaviors and their features. The investigation covers all aspects of the machining process, including the machine tool structural response, cutting process variables, tooling geometry and workpiece material property in a full dynamic scenario. The paper presents a novel approach for coping with the sophisticated machining instability and enabling better understanding of its effect on the surface generation through a combination of the numerical method with the characteristic equations and using block diagrams/functions to represent implicit equations and nonlinear factors. It therefore avoids the lengthy algebraic manipulations in deriving the outcome and the solution scheme is thus simple, robust and intuitive. Several machining case studies and their simulation results demonstrate the proposed approach is feasible for shop floor CNC machining optimisation in particular. The results also indicate the proposed approach is useful to monitor the machining instability and surface topography and to be potentially applied in adaptive control of the instability in real time

    New interleukin-15 superagonist (IL-15SA) significantly enhances graft-versus-tumor activity.

    Interleukin-15 (IL-15) is a potent cytokine that increases CD8+ T and NK cell numbers and function in experimental models. However, obstacles remain in using IL-15 therapeutically, specifically its low potency and short in vivo half-life. To help overcome this, a new IL-15 superagonist complex comprised of an IL-15N72D mutation and IL-15RαSu/Fc fusion (IL-15SA, also known as ALT-803) was developed. IL-15SA exhibits a significantly longer serum half-life and increased in vivo activity against various tumors. Herein, we evaluated the effects of IL-15SA in recipients of allogeneic hematopoietic stem cell transplantation. Weekly administration of IL-15SA to transplant recipients significantly increased the number of CD8+ T cells (specifically CD44+ memory/activated phenotype) and NK cells. Intracellular IFN-γ and TNF-α secretion by CD8+ T cells increased in the IL-15SA-treated group. IL-15SA also upregulated NKG2D expression on CD8+ T cells. Moreover, IL-15SA enhanced proliferation and cytokine secretion of adoptively transferred CFSE-labeled T cells in syngeneic and allogeneic models by specifically stimulating the slowly proliferative and nonproliferative cells into actively proliferating cells.We then evaluated IL-15SA\u27s effects on anti-tumor activity against murine mastocytoma (P815) and murine B cell lymphoma (A20). IL-15SA enhanced graft-versus-tumor (GVT) activity in these tumors following T cell infusion. Interestingly, IL-15 SA administration provided GVT activity against A20 lymphoma cells in the murine donor leukocyte infusion (DLI) model without increasing graft versus host disease. In conclusion, IL-15SA could be a highly potent T- cell lymphoid growth factor and novel immunotherapeutic agent to complement stem cell transplantation and adoptive immunotherapy

    Environmental versus Social Externalities and Life Satisfaction: A Cross-Country Investigation

    U ovom se radu istražuju determinante životnog zadovoljstva na uzorku od 135 zemalja. Osnovni se doprinos rada sastoji u ispitivanju osjetljivosti životnog zadovoljstva na okolišne i društvene eksternalije. Rezultati analize, pritom, imaju implikacije i za nosioce javnih politika. Naime, oni pokazuju da neki faktori konzistentno utječu na razinu životnog zadovoljstva građana, bez obzira jesu li okolišne ili društvene eksternalije uključene u analizu. Postoji, međutim, i skup faktora čiji utjecaj na razinu životnog zadovoljstva oscilira ako se spomenute eksternalije uključe u analizu. Rezultati analize također pokazuju da je razina životnog zadovoljstva u tranzicijskim zemljama, uz ostale nepromijenjene uvjete, niža u odnosu na druge zemlje iz uzorka, upućujući na zaključak da u ovim zemljama nisu ostvarena ranija očekivanja građana.Using recent cross-country data for 135 nations, this paper examines the determinants of life satisfaction. The main contribution lies in examining the sensitivity of satisfaction to externalities, both social and environmental. Besides contributing to the literature, the findings also have some implications for public policy. The results show that some factors consistently affect life satisfaction whether or not externalities are considered, while others are sensitive to such considerations. Even under each category of spillovers, the influence in satisfaction varies. Further, other things being the same, satisfaction was lower in transition nations, perhaps signifying that these countries were somehow failing to meet the expectations of their citizens

    Development of Novel Dual Inhibitor of Chemokine Receptor 4 and Mcl-1 Against Multiple Myeloma

    Multiple myeloma (MM) is a neoplastic plasma-cell disorder. This is characterized by clonal proliferation of malignant plasma cells in the bone-marrow (BM) microenvironment, monoclonal protein in blood or urine, and associated organ dysfunction. The treatment options approved by FDA are immune-modulatory agents, proteasome inhibitors, and autologous stem cell transplantation (ASCT). Unfortunately, MM remains uniformly fatal owing to intrinsic or acquired drug resistance and the median survival time is 3 to 5 years. Thus, there is a great need for novel strategies to combat MM. The intimate relationship of myeloma cells to BM microenvironment is “hallmark of myeloma”. The homing of MM cells to the BM, mediated by the chemokine stromal cell-derived factor-1α (SDF-1α) and its receptor CXCR4 has important functional sequelae. The BM microenvironment constituting cells secrete chemokines, cytokines, and growth factors such as interleukin 6 (IL6), vascular endothelial growth factor (VEGF), SDF-1α, and tumor necrosis factor α (TNFα) etc. These growth factors either secreted by MM or BM microenvironment cells (e.g. stromal cells) contribute in activation of several signaling pathways including nuclear factor-κB (NF-κB); phosphatidylinositol 3-kinase (PI3K)-Akt; Ras-Raf-MAPK kinase (MEK)-extracellular signal regulated kinase (ERK); and the Janus kinase 2 (JAK2)-signal transducer and activator of transcription 3 (STAT3). Activation of these pathways has been associated with increased expression of several anti-apoptotic proteins such as Bcl-2, Bcl-xL, Mcl-1, and XIAP. Collectively, these discoveries highlight that interaction of MM cells to BM microenvironment not only promote growth, survival and migration of MM cells, but also confer resistance to conventional chemotherapy. We hypothesized that an agent capable of inhibiting the migration of myeloma cells to bone marrow and suppressing the expression of survival protein Mcl-1 would be a better option for MM treatment.We have synthesized a novel dual inhibitor of CXCR4 and Mcl-1. Our data suggests that this molecule inhibits the expression of CXCR4 and Mcl-1 and survival of MM cells

    Transferability and polymorphism of barley EST-SSR markers used for phylogenetic analysis in Hordeum chilense

    Background Hordeum chilense, a native South American diploid wild barley, is a potential source of useful genes for cereal breeding. The use of this wild species to increase genetic variation in cereals will be greatly facilitated by marker-assisted selection. Different economically feasible approaches have been undertaken for this wild species with limited direct agricultural use in a search for suitable and cost-effective markers. The availability of Expressed Sequence Tags (EST) derived microsatellites or simple sequence repeat (SSR) markers, commonly called as EST-SSRs, for barley (Hordeum vulgare) represents a promising source to increase the number of genetic markers available for the H. chilense genome. Results All of the 82 barley EST-derived SSR primer pairs tested for transferability to H. chilense amplified products of correct size from this species. Of these 82 barley EST-SSRs, 21 (26%) showed polymorphism among H. chilense lines. Identified polymorphic markers were used to test the transferability and polymorphism in other Poaceae family species with the aim of establishing H. chilense phylogenetic relationships. Triticum aestivum-H. chilense addition lines allowed us to determine the chromosomal localizations of EST-SSR markers and confirm conservation of the linkage group. Conclusion From the present study a set of 21 polymorphic EST-SSR markers have been identified to be useful for diversity analysis of H. chilense, related wild barleys like H. murinum, and for wheat marker-assisted introgression breeding. Across-genera transferability of the barley EST-SSR markers has allowed phylogenetic inference within the Triticeae complex

    Modeling Islamist Extremist Communications on Social Media using Contextual Dimensions: Religion, Ideology, and Hate

    Terror attacks have been linked in part to online extremist content. Although tens of thousands of Islamist extremism supporters consume such content, they are a small fraction relative to peaceful Muslims. The efforts to contain the ever-evolving extremism on social media platforms have remained inadequate and mostly ineffective. Divergent extremist and mainstream contexts challenge machine interpretation, with a particular threat to the precision of classification algorithms. Our context-aware computational approach to the analysis of extremist content on Twitter breaks down this persuasion process into building blocks that acknowledge inherent ambiguity and sparsity that likely challenge both manual and automated classification. We model this process using a combination of three contextual dimensions -- religion, ideology, and hate -- each elucidating a degree of radicalization and highlighting independent features to render them computationally accessible. We utilize domain-specific knowledge resources for each of these contextual dimensions such as Qur'an for religion, the books of extremist ideologues and preachers for political ideology and a social media hate speech corpus for hate. Our study makes three contributions to reliable analysis: (i) Development of a computational approach rooted in the contextual dimensions of religion, ideology, and hate that reflects strategies employed by online Islamist extremist groups, (ii) An in-depth analysis of relevant tweet datasets with respect to these dimensions to exclude likely mislabeled users, and (iii) A framework for understanding online radicalization as a process to assist counter-programming. Given the potentially significant social impact, we evaluate the performance of our algorithms to minimize mislabeling, where our approach outperforms a competitive baseline by 10.2% in precision.Comment: 22 page
