    Dissecting the cellular landscape and transcriptome network in viral myocarditis by single-cell RNA sequencing

    Coxsackievirus B3 (CVB3)-induced myocarditis is commonly employed to study viral pathogenesis in mice.Chronically affected micemaydevelop dilated cardiomyopathy, which may involve the mediation of immune and nonimmune cells. To dissect this complexity, we performed single-cell RNA sequencing on heart cells from healthy and myocarditic mice, leading us to note significant proportions of myeloid cells, T cells, and fibroblasts. Although the transcriptomes of myeloid cells were mainly of M2 phenotype, the Th17 cells, CTLs, and Treg cells had signatures critical for cytotoxic functions. Fibroblasts were heterogeneous expressing genes important in fibrosis and regulation of inflammation and immune responses. The intercellular communication networks revealed unique interactions and signaling pathways in the cardiac cellulome, whereas myeloid cells and T cells had upregulated unique transcription factors modulating cardiac remodeling functions. Together, our data suggest thatM2cells, T cells, and fibroblasts may cooperatively or independently participate in the pathogenesis of viral myocarditis

    Manila clams Ruditapes philippinarum spreading north and establishing in the European Wadden Sea

    In an era of accelerating biological globalization and climatic warming, it is vital to understand how introduced species integrate. Pacific Manila clams Ruditapes philippinarum were introduced for aquaculture to Europe in the 1970s, spread and became harvested from the Mediterranean Sea to British coasts. From the Dutch Rhine Delta, Manila clams immigrated stepwise into the southern Wadden Sea, and by long distance jump dispersal they arrived in the northern Wadden Sea in the 2010s. Encounters remained few until 2021, when live clams were often found in intertidal seagrass beds and around mixed beds of Pacific oysters with native mussels. Shell lengths reached up to 74 mm with a longevity of 7–8 years. Adult abundances remained low (<10 clams m−2), although larval numbers substantially increased in 2022 and 2023. Strong predation on spat may constitute ecological resistance to the immigrant. Haplotype diversity at mtDNA COI gene fragments is high and its composition is mixed from dominant haplotypes of other European sites, suggesting multiple introductions or an unknown source with an already mixed population. Currently, this is the most northern (55◦ N) population of R. philippinarum in continental Europe, but with the rapid expansion of this genetically variable population, further northward spread can be expected

In general, spending is inspired by ideological and opportunistic motives: all government expenditure categories show an upward drift during election times and the partisan motives behind government spending are clearly revealed: left-wing cabinets attach greater importance to social security and health care than right-wing cabinets and right-wing cabinets value expenditure on infrastructure and defense more than left-wing parties.     Targeting Conservation Investments in Heterogeneous Landscapes: A distance function approach and application to watershed management

    To achieve a given level of an environmental amenity at least cost, decision-makers must integrate information about spatially variable biophysical and economic conditions. Although the biophysical attributes that contribute to supplying an environmental amenity are often known, the way in which these attributes interact to produce the amenity is often unknown. Given the difficulty in converting multiple attributes into a unidimensional physical measure of an environmental amenity (e.g., habitat quality), analyses in the academic literature tend to use a single biophysical attribute as a proxy for the environmental amenity (e.g., species richness). A narrow focus on a single attribute, however, fails to consider the full range of biophysical attributes that are critical to the supply of an environmental amenity. Drawing on the production efficiency literature, we introduce an alternative conservation targeting approach that relies on distance functions to cost-efficiently allocate conservation funds across a spatially heterogeneous landscape. An approach based on distance functions has the advantage of not requiring a parametric specification of the amenity function (or cost function), but rather only requiring that the decision-maker identify important biophysical and economic attributes. We apply the distance-function approach empirically to an increasingly common, but little studied, conservation initiative: conservation contracting for water quality objectives. The contract portfolios derived from the distance-function application have many desirable properties, including intuitive appeal, robust performance across plausible parametric amenity measures, and the generation of ranking measures that can be easily used by field practitioners in complex decision-making environments that cannot be completely modeled.
