Synthesis, screening, and sequencing of cysteine-rich one-bead one-compound peptide libraries.
Cysteine-rich peptides are valued as tags for biarsenical fluorophores and as environmentally important reagents for binding toxic heavy metals. Due to the inherent difficulties created by cysteine, the power of one-bead one-compound (OBOC) libraries has never been applied to the discovery of short cysteine-rich peptides. We have developed the first method for the synthesis, screening, and sequencing of cysteine-rich OBOC peptide libraries. First, we synthesized a heavily biased cysteine-rich OBOC library, incorporating 50% cysteine at each position (Ac-X8-KM-TentaGel). Then, we developed conditions for cysteine alkylation, cyanogen bromide cleavage, and direct MS/MS sequencing of that library at the single bead level. The sequencing efficiency of this library was comparable to a traditional cysteine-free library. To validate screening of cysteine-rich OBOC libraries, we reacted a library with the biarsenical FlAsH and identified beads bearing the known biarsenical-binding motif (CCXXCC). These results enable OBOC libraries to be used in high-throughput discovery of cysteine-rich peptides for protein tagging, environmental remediation of metal contaminants, or cysteine-rich pharmaceuticals
Spectroscopic Characterization of i-motif Forming c-myc Derived Sequences Double-Labeled with Pyrene
In current studies we use the oligonucleotides based on c-myc sequence: CCC CAC CCT CCC CAC CCT CCC C (cmyc22) and CCC CAC CCT CCC CAC CCT CCC CA (cmyc22A) functionalized by pyrene moieties at both termini. Results of the circular dichroism (CD), UV absorption melting experiments, and steady-state fluorescence measurements of pyrene-modified i-motifs as well as their unlabeled precursors are presented and discussed here. The pyrene labels have a remarkable influence on i-motif stability which was deduced from CD spectra and confirmed by UV melting experiments. Both probes emit fluorescence band of pyrene monomer with intensity decreasing upon pH lowering. O twórczej destrukcji przestrzeni – Marks i sieci logistyczne
Główny cel artykułu stanowi reinterpretacja najistotniejszych przemian sektora logistycznego (nowe formy i funkcje infrastruktury, innowacje w oprogramowaniu i kształtowaniu przestrzeni, specyfika miast logistycznych itp.) z perspektywy krytycznych studiów miejskich. Wykorzystuję do tego inspiracje płynące z Marksowskich koncepcji cyrkulacji kapitału i niszczenia przestrzeni przez czas. W pierwszej części artykułu zwracam uwagę na obecny w myśli Marksa metodologiczny prymat ruchu oraz wyróżnioną rolę transportu w realizacji, ale również zwiększaniu wartości dodatkowej. Tworzę tym samym główne punkty oparcia dla tezy, w której ujmuję logistykę jako logiczną wypadkową przestrzennej dynamiki kapitału i kapitalistycznej logiki akceleracji, analizowanych przez Marksa w drugim tomie Kapitału i w Zarysie krytyki ekonomii politycznej. Druga część tekstu stanowi rozbudowaną interpretację Marksowskiego pojęcia niszczenia przestrzeni przez czas. Proponując trzy możliwe odczytania tego konceptu (jako kompresję, synchronizację i abstrakcję), wiążę je z typową dla logistyki strategią twórczej destrukcji przestrzeni, która materializuje się na gruncie i za sprawą infrastruktury akceleracji. W trzeciej części omawiam te innowacje sektora, które pozwalają w nim widzieć obszar tendencyjny z punktu widzenia całego kapitalizmu. Podejmuję w tym celu zagadnienie fabrykowania świata (Mezzadra i Neilson 2013a), które umożliwia eksplorowanie nowego porządku geoekonomicznego – z uwzględnieniem zmieniających się postaci władzy infrastruktury i pracowniczej podmiotowości – w terminach globalnej fabryki (Cowen 2014b) i miasta logistycznego
Wywłaszczająca urbanizacja. Miejski marksizm wobec problemu akumulacji pierwotnej
The article is a complex attempt to present the achievements of the broadly understood urban Marxism in the field of reinterpretation of the classic concept from the cards of Capital – “primitive accumulation”. Drawing on theories of such scholars as David Harvey (“accumulation by dispossession”) or Saskia Sassen (“expulsions”), the author points to the possible ways to operationalize the concept of “dispossessing urbanization”. Inspired by the theorizations of autonomist Marxists (Antonio Negri, Sandro Mezzadra, Massimo De Angelis), he proposes to see it as a relatively new and increasingly important dispostif that allows not only to improve traditional methods of enclosures and construct entirely new ones, but also to instrumentalize urban cultures which seems unlikely for the historical versions of primitive accumulation. This theoretical inspiration leads him also to highlight another gap in the accumulation by dispossession theory, namely the lack of sufficient reflection on the production of subjectivity processes which seem constitutive for the broad set of dispossessing practices.Artykuł stanowi próbę kompleksowego przedstawienia dokonań szeroko rozumianego miejskiego marksizmu na polu reinterpretacji klasycznego pojęcia z kart Kapitału – „akumulacji pierwotnej”. Opierając się na teoriach takich autorów jak David Harvey („akumulacja przez wywłaszczenie”) czy Saskia Sassen („wysiedlenia”), autor wskazuje na możliwe sposoby operacjonalizacji wyrastającego z nich konceptu „wywłaszczającej urbanizacji”. Inspirując się dociekaniami marksistów autonomistycznych (Antonio Negri, Sandro Mezzadra, Massimo De Angelis), proponuje widzieć w niej stosunkowo nowe i stale zyskujące na znaczeniu urządzenie, pozwalające nie tylko na usprawnienie tradycyjnych i konstruowanie nowych metod grodzenia, ale i szczególny, nietypowy dla klasycznych postaci akumulacji pierwotnej rodzaj instrumentalizacji miejskich kultur. Wspomniana inspiracja teoretyczna pozwala również na uwypuklenie kolejnej luki w teorii akumulacji przez wywłaszczenie, czyli brak dostatecznego namysłu nad konstytutywnymi dla niej procesami produkcji podmiotowości

Odpowiadają za to zarówno jej wewnętrzne sprzeczności, jak też postępujące odpolitycznienie, które traktuję jako podstawowy cel nowej mutacji miasta przedsiębiorczego. Tym, co pomaga w ich przekroczeniu, są analizowane w drugiej i trzeciej części artykułu praktyki uwspólniania, gdzie zastanawiam się kolejno nad polityczną ontologią miejskich dóbr wspólnych oraz różnymi aspektami kolektywnego odzyskiwania miasta (na przykładzie ateńskich skłotów dla uchodźców). Artykuł uwypukla ich rolę w poszerzaniu dominującego myślenia o polityczności, co dokonuje się poprzez rewaluację takich pozornie niepolitycznych kwestii, jak zamieszkiwanie czy społeczna reprodukcja. Zwraca też uwagę na konieczność upowszechniania procesów uwspólniania, które, łącząc pragmatyzm codziennej walki

Probiotics in colon cancer prevention
Probiotics are live, selected microbial strains that have a beneficial effect on the human body and when introduced into the body, they colonize in the digestive tract, especially in the large intestine, exerting a beneficial effect on the health of the host. The microbial strains, so that they can be included in the probiotics, must be thoroughly tested and meet several conditions. These microorganisms multiply in the gastrointestinal tract and are competitive for pathogenic microorganisms that cause infection. Probiotic bacteria are found in natural yogurts, sour milk, sauerkraut, pickled cucumbers. Many studies show a positive correlation between the consumption of probiotics and the risk of developing certain cancers. Probiotics are most likely to reduce the risk of developing colorectal cancer.

A new strategy for silver deposition on Au nanoparticles with the use of peroxidase-mimicking DNAzyme monitored by Localized Surface Plasmon Resonance technique
Peroxidase-mimicking DNAzyme was applied as a catalyst of silver deposition on gold nanoparticles. This DNAzyme is formed when hemin binds to the G-quadruplex-forming DNA sequence. Such a system is able to catalyze a redox reaction with a one- or two-electron transfer. The process of silver deposition was monitored via a localized surface plasmon resonance technique (LSPR), which allows one to record scattering spectrum of a single nanoparticle. Our study showed that DNAzyme is able to catalyze silver deposition. The AFM experiments proved that DNAzyme induced the deposition of silver shells of approximately 20 nm thickness on Au nanoparticles (AuNPs). Such an effect is not observed when hemin is absent in the system. However, we noticed non-specific binding of hemin to the capture oligonucleotides on a gold NP probe that also induced some silver deposition, even though the capture probe was unable to form G-quadruplex. Analysis of SEM images indicated that the surface morphology of the silver layer deposited by DNAzyme is different from that obtained for hemin alone. The proposed strategy of silver layer synthesis on gold nanoparticles catalyzed by DNAzyme is an innovative approach and can be applied in bioanalysis (LSPR, electrochemistry) as well as in material sciences
