    A genetic variant of the sperm-specific SLO3 K+ channel has altered pH and Ca2+ sensitivities

    To fertilize an oocyte, sperm must first undergo capacitation in which the sperm plasma membrane becomes hyperpolarized via activation of potassium (K(+)) channels and resultant K(+) efflux. Sperm-specific SLO3 K(+) channels are responsible for these membrane potential changes critical for fertilization in mouse sperm, and they are only sensitive to pH i However, in human sperm, the major K(+) conductance is both Ca(2+)- and pH i -sensitive. It has been debated whether Ca(2+)-sensitive SLO1 channels substitute for human SLO3 (hSLO3) in human sperm or whether human SLO3 channels have acquired Ca(2+) sensitivity. Here we show that hSLO3 is rapidly evolving and reveal a natural structural variant with enhanced apparent Ca(2+) and pH sensitivities. This variant allele (C382R) alters an amino acid side chain at a principal interface between the intramembrane-gated pore and the cytoplasmic gating ring of the channel. Because the gating ring contains sensors to intracellular factors such as pH and Ca(2+), the effectiveness of transduction between the gating ring and the pore domain appears to be enhanced. Our results suggest that sperm-specific genes can evolve rapidly and that natural genetic variation may have led to a SLO3 variant that differs from wild type in both pH and intracellular Ca(2+) sensitivities.     Fusarium equiseti and F. semitectum represent the most abundant species in the Fusarium complex isolated from flowers, soybean pods and seeds in Argentina. The aim of the present study was to assess the production of major type A and type B trichothecenes (diacetoxyscirpenol, neosolaniol, T-2 toxin and HT-2 toxin, nivalenol, deoxynivalenol) and zearalenone by 40 F. equiseti and 22 F. semitectum isolates on rice culture. Mycotoxins were determined by HPLC with fluorescence detection after derivatisation with 1-anthronylnitrile for type A trichothecenes (i.e. diacetoxyscirpenol, neosolaniol, T-2 toxin and HT-2 toxin), by HPLC with UV detection for type B trichothecenes (i.e. nivalenol and deoxynivalenol) and by TLC for zearalenone. Twenty-two of 40 F. equiseti isolates produced diacetoxyscirpenol, nivalenol and ZEA alone or in combination, whereas only 2 of 20 F. semitectum isolates were nivalenol and ZEA producers. Both Fusarium species did not produce any deoxynivalenol, neosolaniol, T-2 toxin and HT-2 toxin. The variable retention in toxigenicity displayed by both fungal species suggests that these species have a saprophytic lifestyle in the soybean agroecosystem in Argentina.Fil: Barros, Germán Gustavo.     Non-linear models for black carbon exposure modelling using air pollution datasets

    Black carbon (BC) is a product of incomplete combustion, present in urban aerosols and sourcing mainly from road traffic. Epidemiological evidence reports positive associations between BC and cardiovascular and respiratory disease. Despite this, BC is currently not regulated by the EU Air Quality Directive, and as a result BC data are not available in urban areas from reference air quality monitoring networks in many countries. To fill this gap, a machine learning approach is proposed to develop a BC proxy using air pollution datasets as an input. The proposed BC proxy is based on two machine learning models, support vector regression (SVR) and random forest (RF), using observations of particle mass and number concentrations (N), gaseous pollutants and meteorological variables as the input. Experimental data were collected from a reference station in Barcelona (Spain) over a 2-year period (2018–2019). Two months of additional data were available from a second urban site in Barcelona, for model validation. BC concentrations estimated by SVR showed a high degree of correlation with the measured BC concentrations (R2 = 0.828) with a relatively low error (RMSE = 0.48 µg/m3). Model performance was dependent on seasonality and time of the day, due to the influence of new particle formation events. When validated at the second station, performance indicators decreased (R2 = 0.633; RMSE = 1.19 µg/m3) due to the lack of N data and PM2.5 and the smaller size of the dataset (2 months). New particle formation events critically impacted model performance, suggesting that its application would be optimal in environments where traffic is the main source of ultrafine particles. Due to its flexibility, it is concluded that the model can act as a BC proxy, even based on EU-regulatory air quality parameters only, to complement experimental measurements for exposure assessment in urban areas.The authors would like to acknowledge the support from the Generalitat de Catalunya (Dept.     Comparison of prestellar core elongations and large-scale molecular cloud structures in the Lupus 1 region

    Turbulence and magnetic fields are expected to be important for regulating molecular cloud formation and evolution. However, their effects on sub-parsec to 100 parsec scales, leading to the formation of starless cores, are not well understood. We investigate the prestellar core structure morphologies obtained from analysis of the Herschel-SPIRE 350 mum maps of the Lupus I cloud. This distribution is first compared on a statistical basis to the large-scale shape of the main filament. We find the distribution of the elongation position angle of the cores to be consistent with a random distribution, which means no specific orientation of the morphology of the cores is observed with respect to the mean orientation of the large-scale filament in Lupus I, nor relative to a large-scale bent filament model. This distribution is also compared to the mean orientation of the large-scale magnetic fields probed at 350 mum with the Balloon-borne Large Aperture Telescope for Polarimetry during its 2010 campaign. Here again we do not find any correlation between the core morphology distribution and the average orientation of the magnetic fields on parsec scales. Our main conclusion is that the local filament dynamics---including secondary filaments that often run orthogonally to the primary filament---and possibly small-scale variations in the local magnetic field direction, could be the dominant factors for explaining the final orientation of each core

A team of experts has been set-up to review a broad range of area including industrial plasma physics and pure plasma physics, astrophysical and solar-terrestrial areas. A set of experiments have been identified that can potentially provide access to new phenomena and to allow advances in several fields of plasma science. These experiments concern phenomena on spatial scale (102 to104 m) intermediate between what is achievable on ground experiment and usual solar system plasma observations

    Plasma kinetics issues in an ESA study for a plasma laboratory in space

    A study supported by the European Space Agency (ESA), in the context of its General Studies Programme, performed an investigation of the possible use of space for studies in pure and applied plasma physics, in areas not traditionally covered by ‘space plasma physics’. A set of experiments have been identified that can potentially provide access to new phenomena and to allow advances in several fields of plasma science. These experiments concern phenomena on a spatial scale (101–104 m) intermediate between what is achievable on the ground and the usual solar system plasma observations. Detailed feasibility studies have been performed for three experiments: active magnetic experiments, largescale discharges and long tether–plasma interactions. The perspectives opened by these experiments are discussed for magnetic reconnection, instabilities, MHD turbulence, atomic excited states kinetics, weakly ionized plasmas,plasma diagnostics, artificial auroras and atmospheric studies.     Detección de Rickettsia spp. en ectoparásitos de animales domésticos y silvestres de la Reserva Natural Privada Cerro Chucantí y comunidades aledañas, Panamá, 2007-2010

    Introduction. Ectoparasites are the main vectors of rickettsiosis. In Panama, have limited data on the arthropods that may be considered vectors or reservoirs.Objectives. The aim is to present data on the presence of Rickettsia in ectoparasites of wildlife and domestic animals in the Private Natural Reserve Cerro Chucantí and neighboring villages.Materials and methods. We evaluated 9 humans, 95 domestic mammals and 48 wild. From these, were 21 species of ectoparasites, including fleas, lice, ticks and mites, which were preserved in 95% ethanol. Genetic material was extracted from ticks and fleas to be analyzed by molecular techniques in the detection of Rickettsia.Results. A total of 425 were carried out PCR reactions, of which 270 were positive and 155 negative. The positive, 86 PCR amplified for the gltA gene (55% of positives) of these also amplified 41 (26%) for ompA. DNA of Rickettsia amblyommii was found in horses ticks (Amblyomma cajennense, Dermacentor nitens), dogs ticks (Rhipicephalus sanguineus) and free living nymph in forest. Adicionally, DNA of R. felis was found in dogs fleas Ctenocephalides felis.Conclusions. Detected the presence of R. amblyommii and R. felis in ticks and fleas of domestic animals of the villages near Cerro Chucanti, even if they were unable to find genetic material from Rickettsia in ectoparasites of wildlife.Introducción. Los ectoparásitos son los principales vectores de rickettsiosis. En Panamá se tienen escasos datos sobre los artrópodos que pudieran considerarse vectores o reservorios.Objetivos. Presentar datos sobre la presencia de Rickettsia spp. en ectoparásitos de fauna silvestre y animales domésticos en la Reserva Natural Privada Cerro Chucantí y poblados vecinos.Materiales y métodos. Se revisaron 9 personas, 95 mamíferos domésticos y 48 silvestres. Los animales domésticos se examinaron con anuencia del propietario, mientras que la fauna silvestre se capturó con trampas Sherman y Tomahawk. Se extrajeron 21 especies de ectoparásitos: pulgas, piojos, garrapatas y otros ácaros, los cuales se preservaron en etanol al 95 %. Se extrajo material genético de garrapatas y pulgas para ser analizado por técnicas moleculares en la detección de Rickettsia spp.Resultados. Se practicaron 425 reacciones de PCR, de las cuales, 270 resultaron negativas y 155 positivas. De las positivas, 86 amplificaron para el gen gltA (55 % de las positivas); de estos también amplificaron 41 (26 %) para ompA. Se encontró material genético de Rickettsia amblyommii, en garrapatas de caballos (Amblyomma cajennense, Dermacentor nitens), de perros (Rhipicephalus sanguineus) y ninfas de Amblyomma recolectadas en el bosque. Además, se detectó ADN de R. felis en pulgas Ctenocephalides felis de perros.Conclusiones.     The balloon-borne large-aperture submillimeter telescope for polarimetry: BLAST-Pol

    The Balloon-borne Large Aperture Submillimeter Telescope for Polarimetry (BLAST-Pol) is a suborbital mapping experiment designed to study the role played by magnetic fields in the star formation process. BLAST-Pol is the reconstructed BLAST telescope, with the addition of linear polarization capability. Using a 1.8 m Cassegrain telescope, BLAST-Pol images the sky onto a focal plane that consists of 280 bolometric detectors in three arrays, observing simultaneously at 250, 350, and 500 um. The diffraction-limited optical system provides a resolution of 30'' at 250 um. The polarimeter consists of photolithographic polarizing grids mounted in front of each bolometer/detector array. A rotating 4 K achromatic half-wave plate provides additional polarization modulation. With its unprecedented mapping speed and resolution, BLAST-Pol will produce three-color polarization maps for a large number of molecular clouds. The instrument provides a much needed bridge in spatial coverage between larger-scale, coarse resolution surveys and narrow field of view, and high resolution observations of substructure within molecular cloud cores.
