Differential Contribution of Subunit Interfaces to α 9 α 10 Nicotinic Acetylcholine Receptor Function

Nicotinic acetylcholine receptors can be assembled from either homomeric or heteromeric pentameric subunit combinations. At the interface of the extracellular domains of adjacent subunits lies the acetylcholine binding site, composed of a principal component provided by one subunit and a complementary component of the adjacent subunit. Compared with neuronal nicotinic acetylcholine cholinergic receptors (nAChRs) assembled from α and β subunits, the α9α10 receptor is an atypical member of the family. It is a heteromeric receptor composed only of α subunits. Whereas mammalian α9 subunits can form functional homomeric α9 receptors, α10 subunits do not generate functional channels when expressed heterologously. Hence, it has been proposed that α10 might serve as a structural subunit, much like a β subunit of heteromeric nAChRs, providing only complementary components to the agonist binding site. Here, we have made use of site-directed mutagenesis to examine the contribution of subunit interface domains to α9α10 receptors by a combination of electrophysiological and radioligand binding studies. Characterization of receptors containing Y190T mutations revealed unexpectedly that both α9 and α10 subunits equally contribute to the principal components of the α9α10 nAChR. In addition, we have shown that the introduction of a W55T mutation impairs receptor binding and function in the rat α9 subunit but not in the α10 subunit, indicating that the contribution of α9 and α10 subunits to complementary components of the ligand-binding site is nonequivalent. We conclude that this asymmetry, which is supported by molecular docking studies, results from adaptive amino acid changes acquired only during the evolution of mammalian α10 subunits.Fil: Boffi, Juan Carlos. De la ganadería a la vitivinicultura : la consolidación de un nuevo orden

En los albores de 1880 y de la mano de la ‘Generación del 80’ se producirá el surgimiento del Estado Nacional, previa consolidación de los estados provinciales que articulaban en su interior lo político y la economía local. La presente ponencia pretende reconstruir el proceso desarrollado en la provincia de San Juan desde mediados de 1870, proceso de sustitución de una economía de exportación con destino a los mercados del Pacifico, por otra, destinada al naciente mercado interno. Esto es, el reemplazo del circuito comercial ganadero (y de agricultura subsidiaria) por una actividad agrícola intensiva, de oasis, basada en la vitivinicultura, poniendo particular atención a la identificación de los agentes que formaron parte de esa transformación. La ponencia se enmarca en una línea de trabajo que denominamos sociohistorica, y se propone indagar en las particularidades locales con el objetivo de contribuir al desarrollo de la sociología histórica provincial. El periodo transcurrido entre 1880 y 1930 se caracterizó por un proceso de expansión económica en el cual algunas regiones pudieron incorporarse al modelo de economía agroexportadora. En el caso de San Juan, como sostienen Peñaloza y Arias, los vinos y aguardientes que habían constituido el principal producto comercial de la provincia hasta la caída de Rosas, y relegados a un segundo plano después de Caseros, serán recuperados en el marco del proceso de Conformación del Estado nacional. Así “mientras la mayoría arrancaba vides cual maleza", los “pioneros, tuvieron fe" (Peñaloza y Arias, 1966). Este proceso que transforma la economía local dará lugar a la conformación de un sector de clase dominante que se constituye en dirigente hacia 1900. Existen algunos indicios que permiten pensar que este sector dirigencial se asocia a estos ‘pioneros’. Agentes que, relacionados al ‘comercio de exportación’ (Richard Jorba: 2001) van a experimentar un fuerte proceso de acumulación de capital. Pero también, van a incorporar a otros grupos que se fortalecen en el proceso de expansión y consolidación del mercado interno dando origen a una burguesía agroindustrial favorecida por las medidas tomadas por el Estado en vías de lograr la transformación de la estructura económica.Fil: Lirussi, María Gabriela. RobotPerf: An Open-Source, Vendor-Agnostic, Benchmarking Suite for Evaluating Robotics Computing System Performance

We introduce RobotPerf, a vendor-agnostic benchmarking suite designed to evaluate robotics computing performance across a diverse range of hardware platforms using ROS 2 as its common baseline. The suite encompasses ROS 2 packages covering the full robotics pipeline and integrates two distinct benchmarking approaches: black-box testing, which measures performance by eliminating upper layers and replacing them with a test application, and grey-box testing, an application-specific measure that observes internal system states with minimal interference. Our benchmarking framework provides ready-to-use tools and is easily adaptable for the assessment of custom ROS 2 computational graphs. Drawing from the knowledge of leading robot architects and system architecture experts, RobotPerf establishes a standardized approach to robotics benchmarking. As an open-source initiative, RobotPerf remains committed to evolving with community input to advance the future of hardware-accelerated robotics

Multi-organ point-of-care ultrasound for COVID-19 (PoCUS4COVID): international expert consensus

COVID-19 has caused great devastation in the past year. Multi-organ point-of-care ultrasound (PoCUS) including lung ultrasound (LUS) and focused cardiac ultrasound (FoCUS) as a clinical adjunct has played a significant role in triaging, diagnosis and medical management of COVID-19 patients. The expert panel from 27 countries and 6 continents with considerable experience of direct application of PoCUS on COVID-19 patients presents evidence-based consensus using GRADE methodology for the quality of evidence and an expedited, modified-Delphi process for the strength of expert consensus. The use of ultrasound is suggested in many clinical situations related to respiratory, cardiovascular and thromboembolic aspects of COVID-19, comparing well with other imaging modalities. The limitations due to insufficient data are highlighted as opportunities for future research

A Systematic Review and International Web-Based Survey of Randomized Controlled Trials in the Perioperative and Critical Care Setting: Interventions Reducing Mortality

The authors aimed to identify interventions documented by randomized controlled trials (RCTs) that reduce mortality in adult critically ill and perioperative patients, followed by a survey of clinicians’ opinions and routine practices to understand the clinicians’ response to such evidence. The authors performed a comprehensive literature review to identify all topics reported to reduce mortality in perioperative and critical care settings according to at least 2 RCTs or to a multicenter RCT or to a single-center RCT plus guidelines. The authors generated position statements that were voted on online by physicians worldwide for agreement, use, and willingness to include in international guidelines. From 262 RCT manuscripts reporting mortality differences in the perioperative and critically ill settings, the authors selected 27 drugs, techniques, and strategies (66 RCTs, most frequently published by the New England Journal of Medicine [13 papers], Lancet [7], and Journal of the American Medical Association [5]) with an agreement ≥67% from over 250 physicians (46 countries). Noninvasive ventilation was the intervention supported by the largest number of RCTs (n = 13). The concordance between agreement and use (a positive answer both to “do you agree” and “do you use”) showed differences between Western and other countries and between anesthesiologists and intensive care unit physicians. The authors identified 27 clinical interventions with randomized evidence of survival benefit and strong clinician support in support of their potential life-saving properties in perioperative and critically ill patients with noninvasive ventilation having the highest level of support.
