    Extracorporeal shockwave therapy for the treatment of lower limb intermittent claudication: Study protocol for a randomised controlled trial (the SHOCKWAVE 1 trial)

Background: Peripheral arterial disease (PAD) has a population prevalence of 4.6% with intermittent claudication (IC) presenting as one of the earliest and most common symptoms. PAD has detrimental effects on patients' walking ability in terms of maximum walking distance (MWD) and pain-free walking distance (PFWD). Research has suggested extracorporeal shockwave therapy (ESWT) may induce angiogenesis in treated tissue; therefore, our objective is to assess the tolerability and efficacy of ESWT as a novel treatment of intermittent claudication. Methods/design: Patients with unilateral claudication will be randomised to receive either ESWT (PiezoWave 2 shockwave system) or sham treatment to the calf muscle bulk three times per week for 3 weeks. All patients are blinded to treatment group, and all assessments will be performed by a masked assessor. Treatment tolerability using a visual analogue scale, ankle-brachial pressure index, MWD, PFWD and safety will all be formally assessed as outcome measures at baseline and at 4, 8 and 12 weeks follow-up. Discussion: This trial will be the first of its kind in terms of methodology in relation to ESWT for intermittent claudication. A double-masked randomised controlled trial will provide useful information about the potential for the use of ESWT as a non-invasive treatment option and the need for further robust research.     Gambling-Like Behavior in Pigeons: \u27Jackpot\u27 Signals Promote Maladaptive Risky Choice

    Individuals often face choices that have uncertain outcomes and have important consequences. As a model of this environment, laboratory experiments often offer a choice between an uncertain, large reward that varies in its probability of delivery against a certain but smaller reward as a measure of an individual’s risk aversion. An important factor generally lacking from these procedures are gambling related cues that may moderate risk preferences. The present experiment offered pigeons choices between unreliable and certain rewards but, for the Signaled group on winning choices, presented a ‘jackpot’ signal prior to reward delivery. The Unsignaled group received an ambiguous stimulus not informative of choice outcomes. For the Signaled group, presenting win signals effectively blocked value discounting for the large, uncertain outcome as the probability of a loss increased, whereas the Unsignaled group showed regular preference changes similar to previous research lacking gambling related cues. These maladaptive choices were further shown to be unaffected by more salient loss signals and resistant to response cost increases. The results suggest an important role of an individual’s sensitivity to outcome-correlated cues in influencing risky choices that may moderate gambling behaviors in humans, particularly in casino and other gambling-specific environments

    This article describes the execution of and reflections of a Theatre of the Oppressed workshop offered in 2017 to middle and high school groups visiting the Louisiana Old State Capitol Museum on half-day field trips. The workshops accompanied The Power of Children: Making a Difference exhibit, which features the stories and struggles of Ruby Bridges, Ryan White, and Anne Frank. Despite initial plans for an hour-long session, the workshop designed to connect youth participant experiences to the stories of the children featured in the exhibit was cut to only fifteen minutes. Our group of four facilitators discusses ways we modified Augusto Boal’s Image Theatre practices, the difficulties we faced leading short sessions in the setting of a school field trip, and our worries about maintaining emotional safety as youth tackled themes of power, powerlessness, and policing

    Subgenome Dominance in an Interspecific Hybrid, Synthetic Allopolyploid, and a 140-Year-Old Naturally Established Neo-Allopolyploid Monkeyflower

    Recent studies have shown that one of the parental subgenomes in ancient polyploids is generally more dominant, having retained more genes and being more highly expressed, a phenomenon termed subgenome dominance. The genomic features that determine how quickly and which subgenome dominates within a newly formed polyploid remain poorly understood. To investigate the rate of emergence of subgenome dominance, we examined gene expression, gene methylation, and transposable element (TE) methylation in a natural, \u3c140-year-old allopolyploid (Mimulus peregrinus), a resynthesized interspecies triploid hybrid (M. robertsii), a resynthesized allopolyploid (M. peregrinus), and progenitor species (M. guttatus and M. luteus). We show that subgenome expression dominance occurs instantly following the hybridization of divergent genomes and significantly increases over generations. Additionally, CHH methylation levels are reduced in regions near genes and within TEs in the first-generation hybrid, intermediate in the resynthesized allopolyploid, and are repatterned differently between the dominant and recessive subgenomes in the natural allopolyploid. Subgenome differences in levels of TE methylation mirror the increase in expression bias observed over the generations following hybridization. These findings provide important insights into genomic and epigenomic shock that occurs following hybridization and polyploid events and may also contribute to uncovering the mechanistic basis of heterosis and subgenome dominance

    The Discovery of Cometary Activity in Near-Earth Asteroid (3552) Don Quixote

    The near-Earth object (NEO) population, which mainly consists of fragments from collisions between asteroids in the main asteroid belt, is thought to include contributions from short-period comets as well. One of the most promising NEO candidates for a cometary origin is near-Earth asteroid (3552) Don Quixote, which has never been reported to show activity. Here we present the discovery of cometary activity in Don Quixote based on thermal-infrared observations made with the Spitzer Space Telescope in its 3.6 and 4.5 {\mu}m bands. Our observations clearly show the presence of a coma and a tail in the 4.5 {\mu}m but not in the 3.6 {\mu}m band, which is consistent with molecular band emission from CO2. Thermal modeling of the combined photometric data on Don Quixote reveals a diameter of 18.4 (-0.4/+0.3) km and an albedo of 0.03 (-0.01/+0.02), which confirms Don Quixote to be the third-largest known NEO. We derive an upper limit on the dust production rate of 1.9 kg s^-1 and derive a CO2 gas production rate of (1.1+-0.1)10^26 molecules s^-1. Spitzer IRS spectroscopic observations indicate the presence of fine-grained silicates, perhaps pyroxene rich, on the surface of Don Quixote. Our discovery suggests that CO2 can be present in near-Earth space over a long time.
