    RICORS2040 : The need for collaborative research in chronic kidney disease

    Chronic kidney disease (CKD) is a silent and poorly known killer. The current concept of CKD is relatively young and uptake by the public, physicians and health authorities is not widespread. Physicians still confuse CKD with chronic kidney insufficiency or failure. For the wider public and health authorities, CKD evokes kidney replacement therapy (KRT). In Spain, the prevalence of KRT is 0.13%. Thus health authorities may consider CKD a non-issue: very few persons eventually need KRT and, for those in whom kidneys fail, the problem is 'solved' by dialysis or kidney transplantation. However, KRT is the tip of the iceberg in the burden of CKD. The main burden of CKD is accelerated ageing and premature death. The cut-off points for kidney function and kidney damage indexes that define CKD also mark an increased risk for all-cause premature death. CKD is the most prevalent risk factor for lethal coronavirus disease 2019 (COVID-19) and the factor that most increases the risk of death in COVID-19, after old age. Men and women undergoing KRT still have an annual mortality that is 10- to 100-fold higher than similar-age peers, and life expectancy is shortened by ~40 years for young persons on dialysis and by 15 years for young persons with a functioning kidney graft. CKD is expected to become the fifth greatest global cause of death by 2040 and the second greatest cause of death in Spain before the end of the century, a time when one in four Spaniards will have CKD. However, by 2022, CKD will become the only top-15 global predicted cause of death that is not supported by a dedicated well-funded Centres for Biomedical Research (CIBER) network structure in Spain. Realizing the underestimation of the CKD burden of disease by health authorities, the Decade of the Kidney initiative for 2020-2030 was launched by the American Association of Kidney Patients and the European Kidney Health Alliance. Leading Spanish kidney researchers grouped in the kidney collaborative research network Red de Investigación Renal have now applied for the Redes de Investigación Cooperativa Orientadas a Resultados en Salud (RICORS) call for collaborative research in Spain with the support of the Spanish Society of Nephrology, Federación Nacional de Asociaciones para la Lucha Contra las Enfermedades del Riñón and ONT: RICORS2040 aims to prevent the dire predictions for the global 2040 burden of CKD from becoming true

    DNA mismatch repair gene methylation in gastric cancer in individuals from northern Brazil

    7 páginas, 3 tablas, 1 figura.-- et al.Gastric cancer is one of the most common malignancies. DNA methylation is implicated in DNA mismatch repair genes deficiency. In the present study, we evaluated the methylation status of MLH1, MSH2, MSH6 and PMS2 in 20 diffuse- and 26 intestinal-type gastric cancer samples and 20 normal gastric mucosal of gastric cancer patients from Northern Brazil. We found that none of the nonneoplastic samples showed methylation of any gene promoter and 50% of gastric cancer samples showed at least one methylated gene promoter. Methylation frequencies of MLH1, MSH2, MSH6 and PMS2 promoter were 21.74%, 17.39%, 0% and 28.26% respectively in gastric cancer samples. MLH1 and PMS2 methylation were associated with neoplastic samples compared to nonneoplastic ones. PMS2 methylation was associated with diffuse- and intestinal-type cancer compared with normal controls. Intestinal-type cancer showed significant association with MLH1 methylation. Diffuse-type cancer was significantly associated with MSH2 methylation. Our findings show differential gene methylation in tumoral tissue, which allows us to conclude that methylation is associated with gastric carcinogenesis. Methylation of mismatch repair genes was associated with gastric carcinogenesis and may be a helpful tool for diagnosis, prognosis and therapies.     A escola como lócus de formação docente: uma gestão bem-sucedida.

    O trabalho focaliza a escola como lócus de formação. Analisa o contexto de uma escola de sucesso da rede municipal de Campo Grande, Estado do Mato Grosso do Sul, descrevendo-o em suas características. Entre estas, destaca a gestão e o clima escolar. Para tanto, entrevista professores e gestores sobre a organização da escola, o estilo de gestão, bem como sobre os seus efeitos no trabalho do professor e no desempenho dos alunos. O exame das entrevistas permitiu construir algumas categorias que definem o ethos da instituição: o porte pequeno, o trabalho em equipe, as relações democráticas e o compromisso com os alunos. A análise destaca o profissionalismo da gestão e da sua visão prospectiva sobre a tarefa educativa, estimulando os professores a enfrentarem os desafios e compartilhando com eles as formas de superar as dificuldades. O estudo aponta a importância da rede de relações que se estabelece no contexto da escola como forma de socialização da profissão, construção da identidade docente e espaço de formação de professores a partir da prática

    Um método para determinação de resíduos de oxitetraciclina, tetraciclina e clortetraciclina em leite por cromatografia líquida de alta eficiência foi otimizado e validado. Ensaios intralaboratoriais empregando soluções padrão, brancos de amostras e amostras adicionadas de soluções padrão dos antimicrobianos foram realizados para avaliação da linearidade, seletividade, exatidão, precisão, limites de detecção e de quantificação do método. As amostras foram homogeneizadas, tratadas com solução tampão de succinato de sódio pH 4,0 e centrifugadas, descartando-se o precipitado. As tetraciclinas foram removidas do sobrenadante por quelação com íons cobre reversivelmente ligados à resina quelante e eluídas com tampão McIlvaine-EDTA pH 4,0. As médias de recuperação para as tetraciclinas foram de 76,2 a 104,9% com coeficiente de variação na faixa de 1,4 a 18,9%. Limites de detecção de 10, 15 e 40ng/mL e limites de quantificação de 10, 20 e 50ng/mL foram determinados para oxitetraciclina, tetraciclina e clortetraciclina, respectivamente.<br>A method for determination of oxytetracycline, tetracycline and chlortetracycline residues in milk by high performance liquid chromatography was optimized and validated. Linearity, selectivity, trueness, precision, limits of detection and quantitation were evaluated by in-house validation procedures employing reference materials, sample blanks and spiked samples. Milk samples were homogenized, treated with sodium succinate pH 4.0 buffer and centrifuged, discarding the precipitate. Tetracyclines were removed from the supernatant by chelating with metal ions bound to a chelating resin and eluted with McIlvaine-EDTA pH 4.0 buffer. Mean recoveries for tetracyclines ranged from 76.2 to 104.9% with coefficients of variation from 1.4 to 18.9%. Detection limits of 10, 15 and 40ng/mL and quantitation limits of 10, 20 and 50ng/mL were determined for oxytetracycline, tetracycline and chlortetracycline, respectively

    Diversity and Host Relationships of the Mycoparasite Sepedonium (Hypocreales, Ascomycota) in Temperate Central Chile

    We present the first major survey of regional diversity, distribution and host-association of Sepedonium. Whereas the rather scarce worldwide records of this mycoparasitic fungus suggested no specific distribution pattern of most species before, we provide new evidence of endemic and specific host-parasite guilds of Sepedonium in Southern South America, including the description of a new species. The corresponding inventory was performed in temperate central Chile. The regional landscape, a mosaic of exotic timber plantations and remnants of native Nothofagus forests, facilitates a unique combination of endemic and adventitious Boletales hosts. During a two-year survey, 35 Sepedonium strains were isolated and cultured from infected basidiomata of allochthonous Chalciporus piperatus, Paxillus involutus, Rhizopogon spp. and Suillus spp., as well as from the native Boletus loyita, B. loyo, B. putidus and Gastroboletus valdivianus. Taxonomic diagnosis included morphology of conidia and conidiophores, sequences of ITS, RPB2 and EF1 molecular markers and characteristics of in vitro cultures. Phylogenetic reconstructions were performed using Bayesian methods. Four Sepedonium species could be identified and characterized, viz.: S. ampullosporum, S. chrysospermum, S. laevigatum and the newly described species S. loyorum. The most frequent species on introduced Boletales was S. ampullosporum, followed by S. chrysospermum and S. laevigatum. S. loyorum sp. nov. was found exclusively on native boletacean hosts, separated from its closest relative S. chalcipori by micromorphological and molecular attributes. Species descriptions and identification keys are provided. Ecological and biogeographical aspects of endemic and allochthonous symbiotic units consisting of mycoparasite, ectomycorrhizal fungal host and respective mycorrhizal tree are discussed

    Foi padronizado e validado um método analítico para determinação de resíduos de nitrofurazona, furazolidona e nicarbazina em tecido muscular empregando-se extração com acetonitrila, purificação com cartuchos de sílica e C18 e detecção e quantificação por CLAE/UV. Nos ensaios com amostras fortificadas entre 5 e 200mg/kg as recuperações médias obtidas variaram de 73,6 a 95,6% com valores de C.V. entre 4,8 e 26,4%. O limite de detecção e quantificação do método foi de 5mg/kg, para cada um dos três resíduos estudados.<br>A method for determination of nitrofurazone, furazolidone and nicarbazin residues in muscle tissues was standardized and validated using acetonitrile extraction, clean-up with silica and C18 cartridges and detection and quantification by HPLC/UV. In the assays using spiked samples in levels varying from 5 to 200mg/kg the average recovery values ranged from 73.6 to 95.6% with C.V. values between 4.8 and 26.4%. The limits of detection and quantification of this method was 5mg/kg for each studied residue

    A formação do professor investigador na escola e as possibilidades da pesquisa colaborativa: um retrato sem retoques

    The focus of our research is on how teachers construct their knowledge. We also intend to depth our understanding of the factors that regulate teachers' action - either to maintain, either to modify their professional practice. In order to investigate the means by which teachers learn and the reasons that make them change or resist transforming their practice, we accomplished a task aiming teachers' formation, in a manner similar to a collaborative research design, which involved part of the staff of a school chosen among others which were part of a sample of a previous exploratory study. The subjects were ten teachers of elementary teaching (first stage) and three school administrators. The intervention occurred through our constant presence in the school's daily life, for we believe that empowerment and professional development must be a choice of teachers themselves, based on their own expressed needs toward the construction of their autonomy. A preliminary analysis of data shows that teachers sometimes resist to changes; so these are slow and difficult. Nevertheless, our intervention based on a collaborative research design enabled us to deal with these difficulties, contributing to form a kind of teacher who investigates its own practice
