    Spectral features of canthaxanthin in hcp2. A qm/mm approach

    The increased interest in sequencing cyanobacterial genomes has allowed the identifi-cation of new homologs to both the N-terminal domain (NTD) and C-terminal domain (CTD) of the Orange Carotenoid Protein (OCP). The N-terminal domain homologs are known as Helical Carotenoid Proteins (HCPs). Although some of these paralogs have been reported to act as singlet oxygen quenchers, their distinct functional roles remain unclear. One of these paralogs (HCP2) exclusively binds canthaxanthin (CAN) and its crystal structure has been recently characterized. Its absorption spectrum is significantly red-shifted, in comparison to the protein in solution, due to a dimerization where the two carotenoids are closely placed, favoring an electronic coupling interaction. Both the crystal and solution spectra are red-shifted by more than 50 nm when compared to canthaxanthin in solution. Using molecular dynamics (MD) and quantum mechanical/molecular mechanical (QM/MM) studies of HCP2, we aim to simulate these shifts as well as obtain insight into the environmental and coupling effects of carotenoid–protein interactions.Fil: Nixon, Kevin Clark.     A Digital Twin for Friction Prediction in Dynamic Rubber Applications with Surface Textures

    Surface texturing is an effective method to reduce friction without the need to change materials. In this study, surface textures were transferred to rubber samples in the form of dimples, using a novel laser surface texturing (LST)—based texturing during moulding (TDM) production process, developed within the European Project MouldTex. The rubber samples were used to experimentally determine texture-induced friction variations, although, due to the complexity of manufacturing, only a limited amount was available. The tribological friction measurements were hence combined with an artificial intelligence (AI) technique, i.e., Reduced Order Modelling (ROM). ROM allows obtaining a virtual representation of reality through a set of numerical strategies for problem simplification. The ROM model was created to predict the friction outcome under different operating conditions and to find optimised dimple parameters, i.e., depth, diameter and distance, for friction reduction. Moreover, the ROM model was used to evaluate the impact on friction when manufacturing deviations on dimple dimensions were observed. These results enable industrial producers to improve the quality of their products by finding optimised textures and controlling nominal surface texture tolerances prior to the rubber components production

    Novel population pharmacokinetic model for Linezolid in critically Ill patients and evaluation of the adequacy of the current dosing recommendation

    Antimicrobial treatment in critically ill patients remains challenging. The aim of this study was to develop a population pharmacokinetic model for linezolid in critically ill patients and to evaluate the adequacy of current dosing recommendation (600 mg/12 h). Forty inpatients were included, 23 of whom were subjected to continuous renal replacement therapies (CRRT). Blood and effluent samples were drawn after linezolid administration at defined time points, and linezolid levels were measured. A population pharmacokinetic model was developed, using NONMEM 7.3. The percentage of patients that achieved the pharmacokinetic/pharmacodynamic (PK/PD) targets was calculated (AUC24/MIC > 80 and 100% T>MIC). A two-compartment model best described the pharmacokinetics of linezolid. Elimination was conditioned by the creatinine clearance and by the extra-corporeal clearance if the patient was subjected to CRRT. For most patients, the standard dose of linezolid did not cover infections caused by pathogens with MIC ≥ 2 mg/L. Continuous infusion may be an alternative, especially when renal function is preserved

    Synthesis, structural characterization, and ligand replacement reactions of gem-dithiolato-bridged rhodium and iridium complexes

    The reaction of gem-dithiol compounds R2C(SH)2 (R = Bn (benzyl), iPr; R2 = −(CH2)4−) with dinuclear rhodium or iridium complexes containing basic ligands such as [M(μ-OH)(cod)]2 and [M(μ-OMe)(cod)]2, or the mononuclear [M(acac)(cod)] (M = Rh, Ir, cod = 1,5-cyclooctadiene) in the presence of a external base, afforded the dinuclear complexes [M2(μ-S2CR2)(cod)2] (1−4). The monodeprotonation of 1,1-dimercaptocyclopentane gave the mononuclear complex [Rh(HS2Cptn)(cod)] (5) that is a precursor for the dinuclear compound [Rh2(μ-S2Cptn)(cod)2] (6). Carbonylation of the diolefin compounds gave the complexes [Rh2(μ-S2CR2)(CO)4] (7−9), which reacted with P-donor ligands to stereoselectively produce the trans isomer of the disubstituted complexes [Rh2(μ-S2CR2)(CO)2(PR′3)2] (R′ = Ph, Cy (cyclohexyl)) (10−13) and [Rh2(μ-S2CBn2)(CO)2{P(OR′)3}2] (R′ = Me, Ph) (14−15). The substitution process in [Rh2(μ-S2CBn2)(CO)4] (7) by P(OMe)3 has been studied by spectroscopic means and the full series of substituted complexes [Rh2(μ-S2CBn2)(CO)4−n{P(OR)3}n] (n = 1, 4) has been identified in solution. The cis complex [Rh2(μ-S2CBn2)(CO)2(μ-dppb)] (16) was obtained by reaction of 7 with the diphosphine dppb (1,4-bis(diphenylphosphino)butane). The molecular structures of the diolefinic dinuclear complexes [Rh2(μ-S2CR2)(cod)2] (R = Bn (1), iPr (2); R2 = −(CH2)4− (6)) and that of the cis complex 16 have been studied by X-ray diffraction.The financial support from Ministerio de Educación y Ciencia (MEC/FEDER) Project CTQ2006-03973/BQU is gratefully acknowledged.     FastIC: a fast integrated circuit for the readout of high performance detectors

    This work presents the 8-channel FastIC ASIC developed in CMOS 65 nm technology suitable for the readout of positive and negative polarity sensors in high energy physics experiments, Cherenkov detectors and time-of-flight systems. The front-end can be configured to perform analog summation of up to 4 single-ended channels before discrimination in view of improving time resolution when segmenting a SiPM. The outputs encode the time-of-arrival information and linear energy measurement which captures the peak amplitude of the input signal in the 5 µA–25 mA input peak current range. Power consumption of the ASIC is 12 mW/ch with default settings. Measurements of single photon time resolution with a red-light laser source and a HPK SiPM S13360-3050CS are ≈140 ps FWHM

    A QM/MM approach for the study of monolayer-protected gold clusters

    We report the development and implementation of hybrid methods that combine quantum mechanics (QM) with molecular mechanics (MM) to theoretically characterize thiolated gold clusters. We use, as training systems, structures such as Au25(SCH2-R)18 and Au38(SCH2-R)24, which can be readily compared with recent crystallographic data. We envision that such an approach will lead to an accurate description of key structural and electronic signatures at a fraction of the cost of a full quantum chemical treatment. As an example, we demonstrate that calculations of the 1H and 13C NMR shielding constants with our proposed QM/MM model maintain the qualitative features of a full DFT calculation, with an order-of-magnitude increase in computational efficiency.Comment: Journal of Materials Science, 201

    Grape Resveratrol Increases Serum Adiponectin and Downregulates Inflammatory Genes in Peripheral Blood Mononuclear Cells: A Triple-Blind, Placebo-Controlled, One-Year Clinical Trial in Patients with Stable Coronary Artery Disease

We investigated dose-depending effects of a resveratrol-containing grape supplement on stable patients with coronary artery disease (CAD) treated according to currently accepted guidelines for secondary prevention of cardiovascular disease.[Methods] In a triple-blind, randomized, placebo-controlled, one-year follow-up, 3-arm pilot clinical trial, 75 stable-CAD patients received 350 mg/day of placebo, resveratrol-containing grape extract (grape phenolics plus 8 mg resveratrol) or conventional grape extract lacking resveratrol during 6 months, and a double dose for the following 6 months. Changes in circulating inflammatory and fibrinolytic biomarkers were analyzed. Moreover, the transcriptional profiling of inflammatory genes in peripheral blood mononuclear cells (PBMCs) was explored using microarrays and functional gene expression analysis.[Results] After 1 year, in contrast to the placebo and conventional grape extract groups, the resveratrol-containing grape extract group showed an increase of the anti-inflammatory serum adiponectin (9.6 %, p = 0.01) and a decrease of the thrombogenic plasminogen activator inhibitor type 1 (PAI-1) (−18.6 %, p = 0.05). In addition, 6 key inflammation-related transcription factors were predicted to be significantly activated or inhibited, with 27 extracellular-space acting genes involved in inflammation, cell migration and T-cell interaction signals presenting downregulation (p < 0.05) in PBMCs. No adverse effects were detected in relation to the study products.[Conclusions] Chronic daily consumption of a resveratrol-containing grape nutraceutical could exert cardiovascular benefits in stable-CAD patients treated according to current evidence-based standards, by increasing serum adiponectin, preventing PAI-1 increase and inhibiting atherothrombotic signals in PBMCs.This study was supported by public funds: Projects CICYT-BFU2007-60576 and Consolider-Ingenio 2010 (CSD2007-00063, Fun-C-Food) from the Spanish Ministry of Science and Innovation (MICINN) and GERM-06-04486 (Fundación Séneca, Murcia, Spain).     Epidemiology of imported malaria among children and young adults in Barcelona (1990-2008)

    <p>Abstract</p> <p>Background</p> <p>Increasing international travel and migration is producing changes in trends in infectious diseases, especially in children from many European cities. The objective of this study was to describe the epidemiology and determine the trends of imported malaria in patients under 20 years old in the city of Barcelona, Spain, during an 18-year period.</p> <p>Methods</p> <p>The study included malaria cases that were laboratory confirmed and reported to the malaria register at the Public Health Agency of Barcelona from 1990 to 2008, residing in Barcelona and less than 20 years old. Patients were classified as natives (born in Spain) or immigrants. Differences in the distribution of demographic, clinical characteristics, and incidence per 100,000 person-year evolution were analysed. Natives and immigrants were compared by logistic regression by calculating the <it>odds ratio </it>(OR) with a 95% confidence interval (CI) and Chi-square for a linear trend (p < 0.05).</p> <p>Results</p> <p>Of the total 174 cases, 143 (82.1%) were immigrants, 100 (57.5%) were female, 121 (69.5%) <it>Plasmodium falciparum</it>, and 108 (62.1%) were visiting friends and relatives (VFR) as the reason for travel. Among the immigrants, 99 (67.8%) were from Equatorial Guinea. Immigrant cases more frequently travelled to Africa than natives (p = 0.02). The factors associated with imported malaria among immigrant residents was travelling for VFR (OR: 6.2 CI 1.9-20.2) and age 15-19 (OR: 3.7 CI 1-13.3). The incidence increased from 1990 to 1999 (p < 0.001) and decreased from 2000 to 2008 (p = 0.01), although the global linear trend was not statistically significant (p = 0.41). The fatality rate was 0.5%.</p> <p>Conclusions</p> <p>The majority of cases of malaria in population less than 20 years in Barcelona were immigrants, travelling to Africa for VFR and <it>Plasmodium falciparum </it>was most frequently detected. The trend analysis of the entire study period did not show a statistically significant decline. It is recommended to be aware of malaria, especially among children of immigrants who travel to their parent's home country for VFR. Better access to pre travel advice should be provided.</p

    HRFlexToT: A High Dynamic Range ASIC for Time-of-Flight Positron Emission Tomography

    Time-of-Flight positron emission tomography scanners demand fast and efficient photo sensors and scintillators coupled to fast readout electronics. This article presents the high resolution flexible Time-over-Threshold (HRFlexToT), a 16-channel application-specific-integrated circuit for silicon photomultipliers (SiPM) anode readout manufactured using XFAB 0.18- \mu \text{m} CMOS technology. The main features of the HRFlexToT are a linear Time-over-Threshold with an extended dynamic range (10 bits) for energy measurement, low power consumption (≈ 3.5 mW/ch), and an excellent timing response. The experimental measurements show an energy linearity error of ≈ 3% and an energy resolution of about 12% at 511 keV. Single-photon time resolution measurements performed using an Fondazione Bruno Kessler (FBK) SiPM NUV-HD ( 4 \times 4 mm2 pixel, 40- \mu \text{m} cell) and a Hamamatsu SiPM S13360-3050CS are around 142 and 167 ps full width at half maximum (FWHM), respectively. Coincidence time resolution (CTR) measurements with small cross-section pixelated crystals (LSO:Ce,Ca 0.4%, 2 \times 2 \times 5 mm3) coupled to the same Hamamatsu S13360-3050CS and FBK NUV-HD sensors yield a CTR of 117 ps and 119 ps, respectively. Measurements performed with a large cross-section monolithic crystal (LFS crystal measuring 25 \times 25 \times 20 mm3) and a Hamamatsu SiPM array S13361-6050NE-04 show a CTR of 324 ps FWHM after time-walk and time-skew correction
