    Entanglement in the quantum Ising model

    We study the asymptotic scaling of the entanglement of a block of spins for the ground state of the one-dimensional quantum Ising model with transverse field. When the field is sufficiently strong, the entanglement grows at most logarithmically in the number of spins. The proof utilises a transformation to a model of classical probability called the continuum random-cluster model, and is based on a property of the latter model termed ratio weak-mixing.     Structural and biological identification of residues on the surface of NS3 helicase required for optimal replication of the hepatitis C virus

    The hepatitis C virus (HCV) nonstructural protein 3 (NS3) is a multifunctional enzyme with serine protease and DEXH/D-box helicase domains. A crystal structure of the NS3 helicase domain (NS3h) was generated in the presence of a single-stranded oligonucleotide long enough to accommodate binding of two molecules of enzyme. Several amino acid residues at the interface of the two NS3h molecules were identified that appear to mediate a proteinprotein interaction between domains 2 and 3 of adjacent molecules. Mutations were introduced into domain 3 to disrupt the putative interface and subsequently examined using an HCV subgenomic replicon, resulting in significant reduction in replication capacity. The mutations in domain 3 were then examined using recombinant NS3h in biochemical assays. The mutant enzyme showed RNA binding and RNA-stimulated ATPase activity that mirrored wild type NS3h. In DNA unwinding assays under single turnover conditions, the mutant NS3h exhibited a similar unwinding rate and only ∼2-fold lower processivity than wild type NS3h. Overall biochemical activities of the mutant NS3h were similar to the wild type enzyme, which was not reflective of the large reduction in HCV replicative capacity observed in the biological experiment. Hence, the biological results suggest that the known biochemical properties associated with the helicase activity of NS3h do not reveal all of the likely biological roles of NS3 during HCV replication.     Epizootic Landscapes: Sheep Scab and Regional Environment in England in 1279–1280

    This essay looks at late-medieval rural landscapes of animal disease through the prism of sheep epizootics in England, caused by sheep scab, a highly acute and transmissive disease, whose first wave broke out in 1279–1280. The essay focuses on three regions in England: East Anglia, the Wiltshire-Hampshire Chalklands and Kent, each possessing distinct topographic and environmental features and exhibiting different rates of mortality. The study sets a theoretical model, based on the concept of ‘complexity theory’ and consisting of ten different principles, determining regional variances in dissemination of scab and in mortality patterns. A close analysis of the available statistical sources suggests that there was no ‘universal’ explanatory factor accounting for the correlation between regional geography and mortality rates, and that the situation varied not only from region to region, but from farm to farm, depending on a combination of several possible factors. It is only through a meticulous analysis of local, rather than regional, conditions that the complexity of the situation can begin to be appreciate

    The Majorana Neutrinoless Double-Beta Decay Experiment

    The proposed Majorana double-beta decay experiment is based on an array of segmented intrinsic Ge detectors with a total mass of 500 kg of Ge isotopically enriched to 86% in 76Ge. A discussion is given of background reduction by: material selection, detector segmentation, pulse shape analysis, and electro-formation of copper parts and granularity. Predictions of the experimental sensitivity are given. For an experimental running time of 10 years over the construction and operation of Majorana, a half-life sensitivity of ~4x10^27 y (neutrinoless) is predicted. This corresponds to an effective Majorana mass of the electron neutrino of ~0.03-0.04 eV, according to recent QRPA and RQRPA matrix element calculations.Comment: 10 pages, 7 figure

    Autoantibodies against type I IFNs in patients with life-threatening COVID-19

    Interindividual clinical variability in the course of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection is vast. We report that at least 101 of 987 patients with life-threatening coronavirus disease 2019 (COVID-19) pneumonia had neutralizing immunoglobulin G (IgG) autoantibodies (auto-Abs) against interferon-w (IFN-w) (13 patients), against the 13 types of IFN-a (36), or against both (52) at the onset of critical disease; a few also had auto-Abs against the other three type I IFNs. The auto-Abs neutralize the ability of the corresponding type I IFNs to block SARS-CoV-2 infection in vitro. These auto-Abs were not found in 663 individuals with asymptomatic or mild SARS-CoV-2 infection and were present in only 4 of 1227 healthy individuals. Patients with auto-Abs were aged 25 to 87 years and 95 of the 101 were men. A B cell autoimmune phenocopy of inborn errors of type I IFN immunity accounts for life-threatening COVID-19 pneumonia in at least 2.6% of women and 12.5% of men
