    Preeclampsia is associated with compromized maternal synthesis of long chain polyunsaturated fatty acids leading to offspring deficiency

    Obesity and excessive lipolysis are implicated in preeclampsia (PE). Intrauterine growth restriction is associated with low maternal body mass index and decreased lipolysis. Our aim was to assess how maternal and offspring fatty acid metabolism is altered in mothers in the third trimester of pregnancy with PE (n=62) or intrauterine growth restriction (n=23) compared with healthy pregnancies (n=164). Markers of lipid metabolism and erythrocyte fatty acid concentrations were measured. Maternal adipose tissue fatty acid composition and mRNA expression of adipose tissue fatty acid–metabolizing enzymes and placental fatty acid transporters were compared. Mothers with PE had higher plasma triglyceride (21%, P<0.001) and nonesterified fatty acid (50%, P<0.001) concentrations than controls. Concentrations of major n−6 and n−3 long-chain polyunsaturated fatty acids in erythrocytes were 23% to 60% lower (all P<0.005) in PE and intrauterine growth restriction mothers and offspring compared with controls. Subcutaneous adipose tissue Δ−5 and Δ−6 desaturase and very long-chain fatty acid elongase mRNA expression was lower in PE than controls (respectively, mean [SD] control 3.38 [2.96] versus PE 1.83 [1.91], P=0.030; 3.33 [2.25] versus 1.03 [0.96], P<0.001; 0.40 [0.81] versus 0.00 [0.00], P=0.038 expression relative to control gene [square root]). Low maternal and fetal long-chain polyunsaturated fatty acid concentrations in PE may be the result of decreased maternal synthesis

    (Never) Mind your p's and q's: Von Neumann versus Jordan on the Foundations of Quantum Theory

    In two papers entitled "On a new foundation [Neue Begr\"undung] of quantum mechanics," Pascual Jordan (1927b,g) presented his version of what came to be known as the Dirac-Jordan statistical transformation theory. As an alternative that avoids the mathematical difficulties facing the approach of Jordan and Paul A. M. Dirac (1927), John von Neumann (1927a) developed the modern Hilbert space formalism of quantum mechanics. In this paper, we focus on Jordan and von Neumann. Central to the formalisms of both are expressions for conditional probabilities of finding some value for one quantity given the value of another. Beyond that Jordan and von Neumann had very different views about the appropriate formulation of problems in quantum mechanics. For Jordan, unable to let go of the analogy to classical mechanics, the solution of such problems required the identication of sets of canonically conjugate variables, i.e., p's and q's. For von Neumann, not constrained by the analogy to classical mechanics, it required only the identication of a maximal set of commuting operators with simultaneous eigenstates. He had no need for p's and q's. Jordan and von Neumann also stated the characteristic new rules for probabilities in quantum mechanics somewhat differently. Jordan (1927b) was the first to state those rules in full generality. Von Neumann (1927a) rephrased them and, in a subsequent paper (von Neumann, 1927b), sought to derive them from more basic considerations. In this paper we reconstruct the central arguments of these 1927 papers by Jordan and von Neumann and of a paper on Jordan's approach by Hilbert, von Neumann, and Nordheim (1928). We highlight those elements in these papers that bring out the gradual loosening of the ties between the new quantum formalism and classical mechanics.Comment: New version.     Relational Hidden Variables and Non-Locality

    We use a simple relational framework to develop the key notions and results on hidden variables and non-locality. The extensive literature on these topics in the foundations of quantum mechanics is couched in terms of probabilistic models, and properties such as locality and no-signalling are formulated probabilistically. We show that to a remarkable extent, the main structure of the theory, through the major No-Go theorems and beyond, survives intact under the replacement of probability distributions by mere relations.Comment: 42 pages in journal style.     Complex transboundary movements of marine megafauna in the Western Indian Ocean

    Transboundary marine species have an increased risk of overexploitation as management regimes and enforcement can vary among states. The complex geopolitical layout of exclusive economic zones (EEZs) in the Western Indian Ocean (WIO) introduces the potential for migratory species to cross multiple boundaries, consequently a lack of scientific data could complicate regional management. In the current study, we highlight both the relative lack of spatial data available in the WIO, and the prevalence of transboundary movements in species that have previously been studied in the region. Five tiger sharks Galeocerdo cuvier were tracked with near real‐time positioning (SPOT) satellite tags to determine individual shark movements relative to EEZs within the WIO. Concurrently, a literature search was performed to identify all satellite telemetry studies conducted to date in the WIO for marine megafaunal species, and the results compared to global satellite telemetry effort. Finally, the satellite tracks of all marine species monitored in the WIO were extracted and digitized to examine the scale of transboundary movements that occur in the region. Tiger sharks exhibited both coastal and oceanic movements, with one individual crossing a total of eight EEZs. Satellite telemetry effort in the WIO has not matched the global increase, with only 4.7% of global studies occurring in the region. Species in the WIO remained within the EEZ in which they were tagged in only three studies, while all other species demonstrated some level of transboundary movement. This study demonstrates the lack of spatial data available for informed regional management in an area where transboundary movements by marine megafauna are highly prevalent. Without more dedicated funding and research, the rich biodiversity of the WIO is at risk of overexploitation from the diverse threats present within the various political regions

    Real measurements and Quantum Zeno effect

    In 1977, Mishra and Sudarshan showed that an unstable particle would never be found decayed while it was continuously observed. They called this effect the quantum Zeno effect (or paradox). Later it was realized that the frequent measurements could also accelerate the decay (quantum anti-Zeno effect). In this paper we investigate the quantum Zeno effect using the definite model of the measurement. We take into account the finite duration and the finite accuracy of the measurement. A general equation for the jump probability during the measurement is derived. We find that the measurements can cause inhibition (quantum Zeno effect) or acceleration (quantum anti-Zeno effect) of the evolution, depending on the strength of the interaction with the measuring device and on the properties of the system. However, the evolution cannot be fully stopped.Comment: 3 figure

    Stability of flows associated to gradient vector fields and convergence of iterated transport maps

    In this paper we address the problem of stability of flows associated to a sequence of vector fields under minimal regularity requirements on the limit vector field, that is supposed to be a gradient. We apply this stability result to show the convergence of iterated compositions of optimal transport maps arising in the implicit time discretization (with respect to the Wasserstein distance) of nonlinear evolution equations of a diffusion type. Finally, we use these convergence results to study the gradient flow of a particular class of polyconvex functionals recently considered by Gangbo, Evans ans Savin. We solve some open problems raised in their paper and obtain existence and uniqueness of solutions under weaker regularity requirements and with no upper bound on the jacobian determinant of the initial datum

    Factors associated with knowledge and awareness of Hepatitis B in individuals of Chinese descent:Results from a mass point of care testing and outreach campaign in Toronto, Canada

    BACKGROUND: Migrants from hepatitis B virus (HBV) endemic regions are at high risk of having chronic infection. Despite this, HBV knowledge and awareness programming, and low-barrier screening methods such as point of care (POC) testing, among this group have yet to become routine. METHODS: We conducted a mass HBV POC screening and knowledge and awareness campaign for individuals of Chinese descent in Toronto, Canada. POC screening was administered, then participants completed a knowledge questionnaire. Logistic regression identified associations between demographic factors and participants’ level of HBV knowledge. RESULTS: From 2015 to 2018, 33 outreach events resulted in 891 individuals completing testing and the knowledge questionnaire. Individuals averaged 64.4 years old. Most, 62% (N = 552), were female, and 73.6% (N = 656) have been in Canada for &lt;30 years. The average questionnaire score was 70.7% correct, with 65.2% (N = 581) demonstrating a high level of HBV knowledge. Post-secondary education (OR: 2.19, 95% CI: 1.41, 3.39), income of 50,000 to <75,000 (OR: 2.74, 95% CI: 1.39, 5.43), and having familial history of HBV (OR: 1.72, 95% CI: 1.06, 2.78) were associated with high knowledge. The observed prevalence of HBV was 1.5%, with 13 individuals testing positive on the POC test and confirmatory laboratory testing. CONCLUSIONS: Improving knowledge and awareness of HBV is critical to empowering people, especially migrants who experience barriers to care, to pursue vaccination, testing, and treatment. Combining knowledge outreach and POC test campaigns, enabled discussion and screening for HBV with large numbers of people, and can be tailored for optimal effectiveness for specific groups.</p

    Efficacy of antiviral therapy and host–virus interactions visualised using serial liver sampling with fine-needle aspirates

    Background &amp; Aims: Novel therapies for chronic hepatitis B (CHB), such as RNA interference, target all viral RNAs for degradation, whereas nucleoside analogues are thought to block reverse transcription with minimal impact on viral transcripts. However, limitations in technology and sampling frequency have been obstacles to measuring actual changes in HBV transcription in the liver of patients starting therapy. Methods: We used elective liver sampling with fine-needle aspirates (FNAs) to investigate the impact of treatment on viral replication in patients with CHB. Liver FNAs were collected from patients with CHB at baseline and 12 and 24 weeks after starting tenofovir alafenamide treatment. Liver FNAs were subjected to single-cell RNA sequencing and analysed using the Viral-Track method. Results: HBV was the only viral genome detected and was enriched within hepatocytes. The 5â€Č sequencing technology identified protein-specific HBV transcripts and showed that tenofovir alafenamide therapy specifically reduced pre-genomic RNA transcripts with little impact on HBsAg or HBx transcripts. Infected hepatocytes displayed unique gene signatures associated with an immunological response to viral infection. Conclusions: Longitudinal liver sampling, combined with single-cell RNA sequencing, captured the dynamic impact of antiviral therapy on the replication status of HBV and revealed host–pathogen interactions at the transcriptional level in infected hepatocytes. This sequencing-based approach is applicable to early-stage clinical studies, enabling mechanistic studies of immunopathology and the effect of novel therapeutic interventions. Impact and Implications: Infection-dependent transcriptional changes and the impact of antiviral therapy on viral replication can be measured in longitudinal human liver biopsies using single-cell RNA sequencing data.</p
