    Determination of Nuclear Dna Content in Orchids by Flow Cytometry

    Flow cytometric analysis was found useful for determining ploidy and DNA content values for orchid species and hybrids. Immature leaf tissue yielded the most reliable results for all orchids sampled. Leaf endopolyploidy was common, with levels as high as 16N found in mature D. gouldii leaves. DNA content values for 37 Dendrobium species, 11 Dendrobium hybrids, 33 additional orchid species and 8 hybrids were determined. Values for Dendrobium species ranged from 1.53 pg to 4.23 pg/2C nuclei. C-values for the remaining orchid species ranged from 1.91 pg to 15.19 pg/2C nuclei, with those of Cattleya alone ranging from 3.29 pg to 9.29 pg/2C nuclei. The highest variation within Cattleya corresponded to different specimens of the same species, suggesting polyploidy. An analysis of Dendrobium hybrids and parent species showed that DNA content values could be reliably predicted in cases where parentage was indefinite

    Valuing the benefits from preserving threatened native fauna and flora from invasive animal pests

    Invasive animal pests inflict many kinds of damage on the environment, and threaten native fauna and flora. We attempt to value the benefits from the extra biodiversity that is protected if these threats were removed. The NSW Rural Lands Protection Board is a major agency that undertakes pest control, and is organised into 48 districts across the state. A cross-sectional set of data on Board expenditures, pest abundance, and environmental and climatic characteristics, was compiled by district and analysed. The number of threatened native plant and animal species increases with pest abundance and with the total number of native species present in the district. But the number of threatened species decreases as Board expenditures on pest control increase. The value of preserving an extra species is derived from these changes in expenditure, following conventional economic principles. Then the potential gain in economic surplus is estimated if the threats to biodiversity were removed. The results so far suggest that the value of the total benefit of protecting an extra species is at least 44,250peryear,andthepotentialgaininsurplusforNewSouthWalesifthethreatswereremovedisatleast44,250 per year, and the potential gain in surplus for New South Wales if the threats were removed is at least 132m per year. This change in surplus is also the total economic loss because invasive pests threaten native flora and fauna. If only half the native species could be protected, the avoidable economic loss is at least $95.7m per year.     Evidence for a nurse-led protocol for removing urinary catheters: A scoping review

    Background: Catheter-associated urinary tract infections significantly contribute to hospital acquired complications globally, with adverse implications for patient outcomes, healthcare, and fiscal resources. Nurse-led protocols for early removal of urinary catheters to reduce the incidence of catheter-associated urinary tract infections have been trialled. Aim: To report the evidence for nurse-led practices of removing urinary catheters within the acute healthcare setting. Methods: Five databases (CINAHL, MEDLINE, SCOPUS, EMCARE, and INFORMIT) were systematically searched in a scoping review of all peer reviewed publications up to 12/03/2021. Findings: Thirteen studies met the inclusion criteria. Eleven studies described a reduction in catheter-associated urinary tract infections regardless of the type of intervention, one study did not demonstrate a change and one study reported an increase in catheter-associated urinary tract infections. Settings, study duration and sample size varied substantially between the included studies. Interventions were exclusive nurse-led protocol for removal of urinary catheters, computerised reminder systems, bundle approaches or comprehensive packages. Outcome measures and definitions of catheter-associated urinary tract infections were varied or absent. Discussion: The quality of evidence of included studies in this review was low, attributed to by a number of methodological issues related to sample size and statistical analyses. Whilst the introduction of nurse-led protocols showed some improvements, the methodological inconsistencies make it difficult to highlight a specific protocol. Conclusion: Given the quality of existing evidence, caution is required in translating these findings to policy and practice

    Final Results From the Circumarctic Lakes Observation Network (CALON) Project

    Since 2012, the physical and biogeochemical properties of ~60 lakes in northern Alaska have been investigated under CALON, a project to document landscape-scale variability of Arctic lakes in permafrost terrain. The network has ten nodes along two latitudinal transects extending inland 200 km from the Arctic Ocean. A meteorological station is deployed at each node and six representative lakes instrumented and continuously monitored, with winter and summer visits for synoptic assessment of lake conditions. Over the 4-year period, winter and summer climatology varied to create a rich range of lake responses over a short period. For example, winter 2012-13 was very cold with a thin snowpack producing thick ice across the region. Subsequent years had relatively warm winters, yet regionally variable snow resulted in differing gradients of ice thickness. Ice-out timing was unusually late in 2014 and unusually early in 2015. Lakes are typically well–mixed and largely isothermal, with minor thermal stratification occurring in deeper lakes during calm, sunny periods in summer. Lake water temperature records and morphometric data were used to estimate the ground thermal condition beneath 28 lakes. Application of a thermal equilibrium steady-state model suggests a talik penetrating the permafrost under many larger lakes, but lake geochemical data do not indicate a significant contribution of subpermafrost groundwater. Biogeochemical data reveal distinct spatial and seasonal variability in chlorophyll biomass, chromophoric dissolved organic carbon (CDOM), and major cations/anions. Generally, waters sampled beneath ice in April had distinctly higher concentrations of inorganic solutes and methane compared with August. Chlorophyll concentrations and CDOM absorption were higher in April, suggesting significant biological/biogeochemical activity under lake ice. Lakes are a positive source of methane in summer, and some also emit nitrous oxide and carbon dioxide. As part of the Indigenous Knowledge component,76 Iñupiat elders, hunters and berry pickers have been interviewed and over 75 hours of videotaped interviews produced.     Identifying factors associated with sedentary time after stroke. Secondary analysis of pooled data from nine primary studies.

    <p><b>Background</b>: High levels of sedentary time increases the risk of cardiovascular disease, including recurrent stroke.</p> <p><b>Objective</b>: This study aimed to identify factors associated with high sedentary time in community-dwelling people with stroke.</p> <p><b>Methods</b>: For this data pooling study, authors of published and ongoing trials that collected sedentary time data, using the activPAL monitor, in community-dwelling people with stroke were invited to contribute their raw data. The data was reprocessed, algorithms were created to identify sleep-wake time and determine the percentage of waking hours spent sedentary. We explored demographic and stroke-related factors associated with total sedentary time and time in uninterrupted sedentary bouts using unique, both univariable and multivariable, regression analyses.</p> <p><b>Results</b>: The 274 included participants were from Australia, Canada, and the United Kingdom, and spent, on average, 69% (SD 12.4) of their waking hours sedentary. Of the demographic and stroke-related factors, slower walking speeds were significantly and independently associated with a higher percentage of waking hours spent sedentary (p = 0.001) and uninterrupted sedentary bouts of <i>>30</i> and <i>>60 min</i> (p = 0.001 and p = 0.004, respectively). Regression models explained 11–19% of the variance in total sedentary time and time in prolonged sedentary bouts.</p> <p><b>Conclusion</b>: We found that variability in sedentary time of people with stroke was largely unaccounted for by demographic and stroke-related variables. Behavioral and environmental factors are likely to play an important role in sedentary behavior after stroke. Further work is required to develop and test effective interventions to address sedentary behavior after stroke.</p

    Feasibility of low-intensity psychological interventions for emotional and behavioural difficulties in children and young people with genetic conditions: a case series

    Background: Children with genetic conditions may experience significant mental health difficulties such as anxiety and challenging behaviour. However, understanding of the feasibility and effectiveness of psychological interventions for emotional and behavioural problems in the context of genetic conditions is limited. Low-intensity psychological interventions have demonstrated promise in paediatric populations and may be able to address their mental health difficulties. / Method: A case series design was used to assess the feasibility of low-intensity interventions for emotional and behavioural difficulties in children and young people with genetic conditions recruited from a mental health drop-in centre at a tertiary hospital. Participants received seven weekly sessions with a trained practitioner. The intervention was based on existing modular treatments and evidence-based self-help materials. Feasibility and treatment satisfaction were assessed, as well as measures of symptoms of anxiety and challenging behaviour, treatment goals, and quality of life, at baseline, during treatment, and six-month follow-up. / Results: Five participants received treatment for challenging behaviour, one for anxiety, and one for obsessive compulsive disorder. All participants completed treatment. Clinically significant change in the SDQ Total score was found in three participants. All participants demonstrated progress in goals and symptoms of emotional and behavioural difficulties over the course of treatment. / Conclusions: Low-intensity psychological interventions for emotional and behavioural difficulties in children and young people with genetic conditions is feasible, acceptable, and potentially beneficial. Further research is warranted to examine the effectiveness of the intervention and its use in clinical paediatric settings

    A thin layer angiogenesis assay: a modified basement matrix assay for assessment of endothelial cell differentiation

    BACKGROUND: Basement matrices such as Matrigel™ and Geltrex™ are used in a variety of cell culture assays of anchorage-dependent differentiation including endothelial cell tube formation assays. The volumes of matrix recommended for these assays (approximately 150 μl/cm(2)) are costly, limit working distances for microscopy, and require cell detachment for subsequent molecular analysis. Here we describe the development and validation of a thin-layer angiogenesis (TLA) assay for assessing the angiogenic potential of endothelial cells that overcomes these limitations. RESULTS: Geltrex™ basement matrix at 5 μl/cm(2) in 24-well (10 μl) or 96-well (2 μl) plates supports endothelial cell differentiation into tube-like structures in a comparable manner to the standard larger volumes of matrix. Since working distances are reduced, high-resolution single cell microscopy, including DIC and confocal imaging, can be used readily. Using MitoTracker dye we now demonstrate, for the first time, live mitochondrial dynamics and visualise the 3-dimensional network of mitochondria present in differentiated endothelial cells. Using a standard commercial total RNA extraction kit (Qiagen) we also show direct RNA extraction and RT-qPCR from differentiated endothelial cells without the need to initially detach cells from their supporting matrix. CONCLUSIONS: We present here a new thin-layer assay (TLA) for measuring the anchorage-dependent differentiation of endothelial cells into tube-like structures which retains all the characteristics of the traditional approach but with the added benefit of a greatly lowered cost and better compatibility with other techniques, including RT-qPCR and high-resolution microscopy.     A systematic review of the effect of reproductive intention screening in primary care settings on reproductive health outcomes.

    Purpose: No recommendations exist for routine reproductive intention screening in primary care. The objective of this systematic review is to assess the effect of reproductive intention screening in primary care on reproductive health outcomes (PROSPERO CRD42015019726). Methods: We performed a systematic search in Ovid Medline, PubMed, CINAHL, Embase, CDR/DARE databases, Web of Science, ISRCTN registry, Clinicaltrials.gov and Cochrane Library. Studies published in English between 2000 and 2017 and whose population was patients of reproductive age (15-49) were included. Studies without a comparison group were excluded. Two independent reviewers assessed eligibility, study quality and abstracted data. Results: Of 24 780 titles and/or abstracts reviewed, nine studies met inclusion criteria: four randomized controlled trials (RCTs) and five observational studies. Two RCTs and one quasi-experimental cohort study showed a statistically significant increase in knowledge related to healthier pregnancy, such as the benefits of folic acid supplementation, and increased risk profiles for those with chronic conditions. Among studies measuring contraceptive use, only one cohort study showed any increase while the RCT and retrospective cohort did not show a statistically significant effect. Neither of the two RCTs that assessed the provision of contraception by primary care providers for those not desiring pregnancy found increased access to contraception, although one found increased documentation of contraception in electronic medical records. Acceptability of reproductive intention screening was measured in seven studies, and participant satisfaction was high in all seven studies. Conclusions: More research is needed to determine whether routine inclusion of reproductive intention screening in primary care is warranted

    ‘What are you going to do, confiscate their passports?’ Professional perspectives on cross-border reproductive travel

    Objective: This article reports findings from a UK-based study which explored the phenomenon of overseas travel for fertility treatment. The first phase of this project aimed to explore how infertility clinicians and others professionally involved in fertility treatment understand the nature and consequences of cross-border reproductive travel. Background: There are indications that, for a variety of reasons, people from the UK are increasingly travelling across national borders to access assisted reproductive technologies. While research with patients is growing, little is known about how ‘fertility tourism’ is perceived by health professionals and others with a close association with infertility patients. Methods: Using an interpretivist approach, this exploratory research included focussed discussions with 20 people professionally knowledgeable about patients who had either been abroad or were considering having treatment outside the UK. Semi-structured interviews were recorded, transcribed verbatim and subjected to a thematic analysis. Results: Three conceptual categories are developed from the data: ‘the autonomous patient’; ‘cross-border travel as risk’, and ‘professional responsibilities in harm minimisation’. Professionals construct nuanced, complex and sometimes contradictory narratives of the ‘fertility traveller’, as vulnerable and knowledgeable; as engaged in risky behaviour and in its active minimisation. Conclusions: There is little support for the suggestion that states should seek to prevent cross-border treatment. Rather, an argument is made for less direct strategies to safeguard patient interests. Further research is required to assess the impact of professional views and actions on patient choices and patient experiences of treatment, before, during and after travelling abroad

    Slow progressors to type 1 diabetes lose islet autoantibodies over time, have few islet antigen-specific CD8+ T cells and exhibit a distinct CD95hi B cell phenotype

    ims/hypothesis The aim of this study was to characterise islet autoantibody profiles and immune cell phenotypes in slow progressors to type 1 diabetes. Methods Immunological variables were compared across peripheral blood samples obtained from slow progressors to type 1 diabetes, individuals with newly diagnosed or long-standing type 1 diabetes, and healthy individuals. Polychromatic flow cytometry was used to characterise the phenotypic attributes of B and T cells. Islet autoantigen-specific B cells were quantified using an enzyme-linked immunospot (ELISpot) assay and islet autoantigen-specific CD8+ T cells were quantified using peptide–HLA class I tetramers. Radioimmunoassays were used to detect islet autoantibodies. Sera were assayed for various chemokines, cytokines and soluble receptors via ELISAs. Results Islet autoantibodies were lost over time in slow progressors. Various B cell subsets expressed higher levels of CD95 in slow progressors, especially after polyclonal stimulation, compared with the corresponding B cell subsets in healthy donors (p < 0.05). The phenotypic characteristics of CD4+ and CD8+ T cells were similar in slow progressors and healthy donors. Lower frequencies of CD4+ T cells with a central memory phenotype (CD27int, CD127+, CD95int) were observed in slow progressors compared with healthy donors (mean percentage of total CD4+ T cells was 3.00% in slow progressors vs 4.67% in healthy donors, p < 0.05). Autoreactive B cell responses to proinsulin were detected at higher frequencies in slow progressors compared with healthy donors (median no. of spots was 0 in healthy donors vs 24.34 in slow progressors, p < 0.05) in an ELISpot assay. Islet autoantigen-specific CD8+ T cell responses were largely absent in slow progressors and healthy donors. Serum levels of DcR3, the decoy receptor for CD95L, were elevated in slow progressors compared with healthy donors (median was 1087 pg/ml in slow progressors vs 651 pg/ml in healthy donors, p = 0.06). Conclusions/interpretation In this study, we found that slow progression to type 1 diabetes was associated with a loss of islet autoantibodies and a distinct B cell phenotype, consistent with enhanced apoptotic regulation of peripheral autoreactivity via CD95. These phenotypic changes warrant further studies in larger cohorts to determine their functional implications
