    Multi-Telescope Radio Observations for Low Frequency Gravitational Wave Astrophysics

    The North American Nanohertz Observatory for Gravitational Waves (NANOGrav) has the principal goal of detecting gravitational waves (GWs) in the nanohertz part of the spectrum using pulsar timing. This thesis presents results from radio campaigns at frequencies from 322 MHz to 10 GHz aimed at both multi-messenger constraints on GW sources and improving the timing sensitivity. The primary expected source of GWs at the nanohertz frequencies to which pulsar timing is sensitive are supermassive black hole (SMBH) binaries. We investigate a purported SMBH displaced from the galactic photocenter in NGC 3115. We explore the possibilities that the source is a SMBH binary or a post-merger recoiling SMBH. We place constraints on a possible SMBH companion using observations taken with the NRAO Very Large Array. If a companion SMBH can be confirmed, this system could be a future GW source detectable with pulsar timing. In order to detect such sources, our pulsar timing array must be as sensitive as possible, requiring the mitigation of all other astrophysical delays, including those from the interstellar medium (ISM). Using NANOGrav wideband multi-frequency observations obtained with the Green Bank Telescope and Arecibo Observatory, we characterize frequency-dependent dispersion. This effect is quantified by the dispersion measure (DM). We analyze trends in the DM time series, propose sources of these trends, and identify timescales over which the DM varies beyond measurement errors and therefore can no longer be modeled as constant in timing. Analyzing DM variations aids in characterizing properties of the ISM and informs our timing observation strategy. Multi-telescope observations around the globe and at complementary frequencies can be used to more sensitively constrain DMs. We compare DMs measured with dual-frequency observations obtained using the Giant Metrewave Radio Telescope (GMRT) to those calculated in the NANOGrav 11-year data release to assess the possible precision of frequency-dependent noise measurements with the GMRT. We discuss the possibility of incorporating the GMRT into international pulsar timing efforts and anticipated challenges in future data combination

    Transgenic Drosophila for Investigating DUX4 and FRG1, Two Genes Associated with Facioscapulohumeral Muscular Dystrophy (FSHD)

    Facioscapulohumeral muscular dystrophy (FSHD) is typically an adult onset dominant myopathy. Epigenetic changes in the chromosome 4q35 region linked to both forms of FSHD lead to a relaxation of repression and increased somatic expression of DUX4-fl (DUX4-full length), the pathogenic alternative splicing isoform of the DUX4 gene. DUX4-fl encodes a transcription factor expressed in healthy testis and pluripotent stem cells; however, in FSHD, increased levels of DUX4-fl in myogenic cells lead to aberrant regulation of target genes. DUX4-fl has proven difficult to study in vivo; thus, little is known about its normal and pathogenic roles. The endogenous expression of DUX4-fl in FSHD-derived human muscle and myogenic cells is extremely low, exogenous expression of DUX4-fl in somatic cells rapidly induces cytotoxicity, and, due in part to the lack of conservation beyond primate lineages, viable animal models based on DUX4-fl have been difficult to generate. By contrast, the FRG1 (FSHD region gene 1), which is linked to FSHD, is evolutionarily conserved from invertebrates to humans, and has been studied in several model organisms. FRG1 expression is critical for the development of musculature and vasculature, and overexpression of FRG1 produces a myopathic phenotype, yet the normal and pathological functions of FRG1 are not well understood. Interestingly, DUX4 and FRG1 were recently linked when the latter was identified as a direct transcriptional target of DUX4-FL. To better understand the pathways affected in FSHD by DUX4-fl and FRG1, we generated transgenic lines of Drosophila expressing either gene under control of the UAS/GAL4 binary system. Utilizing these lines, we generated screenable phenotypes recapitulating certain known consequences of DUX4-fl or FRG1 overexpression. These transgenic Drosophila lines provide resources to dissect the pathways affected by DUX4-fl or FRG1 in a genetically tractable organism and may provide insight into both muscle development and pathogenic mechanisms in FSHD

    PREDICTORS OF PELVIC ACCELERATION DURING TREADMILL RUNNING AT DIFFERENT STRIDE FREQUENCIES

    The aim of this study was to examine predictors of peak vertical and anteroposterior pelvic acceleration during treadmill running. Participants ran at 9 km∙h-1 at their preferred stride frequency and at ± 5% of their preferred stride frequency. Coordinate and acceleration data were collected using a motion capture system and inertial measurement units. Linear mixed models showed that for every one standard deviation increase in the anteroposterior displacement from knee to ankle at initial contact, vertical pelvic acceleration increased by 2.18 m∙s-2 (p = 0.046). Additionally, for every one standard deviation increase in stride frequency, peak anteroposterior pelvic acceleration increased by 0.68 m∙s-2 (p = 0.035). Runners who suffer from injuries or pain at the pelvis may benefit from decreasing the anteroposterior displacement from their knee to their ankle at initial contact and reducing their stride frequency

    Systematic and Stochastic Variations in Pulsar Dispersion Measures

    We analyze deterministic and random temporal variations in dispersion measure (DM) from the full three-dimensional velocities of pulsars with respect to the solar system, combined with electron-density variations on a wide range of length scales. Previous treatments have largely ignored the pulsar's changing distance while favoring interpretations involving the change in sky position from transverse motion. Linear trends in pulsar DMs seen over 5-10~year timescales may signify sizable DM gradients in the interstellar medium (ISM) sampled by the changing direction of the line of sight to the pulsar. We show that motions parallel to the line of sight can also account for linear trends, for the apparent excess of DM variance over that extrapolated from scintillation measurements, and for the apparent non-Kolmogorov scalings of DM structure functions inferred in some cases. Pulsar motions through atomic gas may produce bow-shock ionized gas that also contributes to DM variations. We discuss possible causes of periodic or quasi-periodic changes in DM, including seasonal changes in the ionosphere, annual variation of the solar elongation angle, structure in the heliosphere-ISM boundary, and substructure in the ISM. We assess the solar cycle's role on the amplitude of ionospheric and solar-wind variations. Interstellar refraction can produce cyclic timing variations from the error in transforming arrival times to the solar system barycenter. We apply our methods to DM time series and DM gradient measurements in the literature and assess consistency with a Kolmogorov medium.     Smoke-free homes: what are the barriers, motivators and enablers? A qualitative systematic review and thematic synthesis

    Objective: To thematically synthesise primary qualitative studies of the barriers, motivators and enablers of smoke-free homes (SFHs). Design: Systematic review and thematic synthesis. Data sources: Searches of MEDLINE, EBM Reviews (Cochrane Database of Systematic Reviews), PsycINFO, Global Health, CINAHL, Web of Science, Informit and EMBASE, combining terms for families, households and vulnerable populations; SFH and secondhand smoke; and qualitative research, were supplemented by searches of PhD theses, key authors, specialist journals and reference lists. Study selection: We included 22 articles, reporting on 18 studies, involving 646 participants. Inclusion criteria: peer-reviewed; English language; published from 1990 onwards (to week 3 of April 2014); used qualitative data collection methods; explored participants’ perspectives of home smoking behaviours; and the barriers, motivators and enablers to initiating and/or maintaining a SFH. Data extraction: 1 of 3 authors extracted data with checking by a second. Data synthesis: A thematic synthesis was performed to develop 7 core analytic themes: (1) knowledge, awareness and risk perception; (2) agency and personal skills/attributes; (3) wider community norms and personal moral responsibilities; (4) social relationships and influence of others; (5) perceived benefits, preferences and priorities; (6) addiction and habit; (7) practicalities. Conclusions: This synthesis highlights the complexity faced by many households in having a SFH, the practical, social, cultural and personal issues that need to be addressed and balanced by households, and that while some of these are common across study settings, specific social and cultural factors play a critical role in shaping household smoking behaviours. The findings can inform policy and practice and the development of interventions aimed at increasing SFHs. Trial registration number: CRD42014014115

    Trajectories of Childhood Weight Gain: The Relative Importance of Local Environment versus Individual Social and Early Life Factors

    Objective: To determine the association between local environmental factors with child weight status in a longitudinal study, using a semi-parametric, group-based method, while also considering social and early life factors. Methods: Standardized, directly measured BMI from 4-10 y of age, and group-based trajectory modeling (PROC TRAJ) were used to estimate developmental trajectories of weight change in a Québec birth cohort (n = 1,566). Associations between the weight trajectories and living location, social cohesion, disorder, and material and social deprivation were estimated after controlling for social and early life factors. Results: Four weight trajectory groups were estimated: low-increasing (9.7%); low-medium, accelerating (36.2%); medium-high, increasing (43.0%); and high-stable (11.1%). In the low-increasing and medium-high trajectory groups, living in a semi-urban area was inversely related to weight, while living in a rural area was positively related to weight in the high-stable group. Disorder was inversely related to weight in the low-increasing group only. Other important risk factors for high-stable weight included obesity status of the mother, smoking during pregnancy, and overeating behaviors. Conclusions: In this study, associations between local environment factors and weight differed by trajectory group. Early life factors appear to play a more consistent role in weight status. Further work is needed to determine the influence of place on child weight. © 2012 Carter et al

    A stem-cell-derived platform enables complete Cryptosporidium development in vitro and genetic tractability

    Despite being a frequent cause of severe diarrheal disease in infants and an opportunistic infection in immunocompromised patients, Cryptosporidium research has lagged due to a lack of facile experimental methods. Here, we describe a platform for complete life cycle development and long-term growth of C. parvum in vitro using air-liquid interface (ALI) cultures derived from intestinal epithelial stem cells. Transcriptomic profiling revealed that differentiating epithelial cells grown under ALI conditions undergo profound changes in metabolism and development that enable completion of the parasite life cycle in vitro. ALI cultures support parasite expansion \u3e 100-fold and generate viable oocysts that are transmissible in vitro and to mice, causing infection and animal death. Transgenic parasite lines created using CRISPR/Cas9 were used to complete a genetic cross in vitro, demonstrating Mendelian segregation of chromosomes during meiosis. ALI culture provides an accessible model that will enable innovative studies into Cryptosporidium biology and host interactions

    Plural policing in Europe:relationships and governance in contemporary security system

    References to ‘plural policing’, ‘policing beyond the police’ and the ‘extended policing family’ are now commonplace in many discussions of policing in late modern societies. There is a danger that claims about the dynamic and changing nature of plural policing themselves become a new orthodoxy and begin to lose a sense of local nuance and recognition of the importance of place-based specificity and context in understanding the particularities of policing. It is this need to unpack the complex ways in which contemporary plural policing is now configured at a local level within different national political environments that provides the underpinning rationale for this Special Issue. Focussing on aspects of relationships and governance in six jurisdictions across northern and western Europe, it provides important insights into how the policies, practices and narratives around plural policing reflect the influence of particular histories and geographies. The first three articles are focused primarily on the relationships which have emerged in the public sector through its own processes of pluralisation, in particular, through the introduction of policing auxiliaries or municipal policing in Scotland, England and The Netherlands. The fourth article considers both relationships and governance in pluralised policing in Paris, France. A detailed analysis of the governance of safety and security is taken up in the final two articles, examining the cases of Austria and Belgium. These articles clearly demonstrate that experiences of pluralised policing vary widely within Europe and call into question the assumed dominance of neo-liberal forces in this area

    An Examination of Commercial Aviation Accidents and Incidents Related to Integrated Vehicle Health Management

    The Integrated Vehicle Health Management (IVHM) Project is one of the four projects within the National Aeronautics and Space Administration's (NASA) Aviation Safety Program (AvSafe). The IVHM Project conducts research to develop validated tools and technologies for automated detection, diagnosis, and prognosis that enable mitigation of adverse events during flight. Adverse events include those that arise from system, subsystem, or component failure, faults, and malfunctions due to damage, degradation, or environmental hazards that occur during flight. Determining the causal factors and adverse events related to IVHM technologies will help in the formulation of research requirements and establish a list of example adverse conditions against which IVHM technologies can be evaluated. This paper documents the results of an examination of the most recent statistical/prognostic accident and incident data that is available from the Aviation Safety Information Analysis and Sharing (ASIAS) System to determine the causal factors of system/component failures and/or malfunctions in U.S. commercial aviation accidents and incidents
