Margaret Fuller1 When Henry David Thoreau alludes to Samuel Taylor Coleridge\u27s Dejection: An Ode in his epigraph to Walden, he situates his Walden Pond experiment as a contribution to a transatlantic conversation concerning a central theme in Coleridge\u27s poem - namely, how the self makes meaning of its relation to the world.2 In fact, Coleridge\u27s corpus shaped Thoreau\u27s dedicated exploration of just this relation - that between the self and nature. For Thoreau, and for Coleridge before him, this relation involved a third integral category: spirit. While today we tend to separate the human self, the external world of nature, adn the world of spirit, Coleridge envisioned these categories as comprising a continuum accessible through the human mind. Hence, he wrote, Then it is, that Nature, like an individual spirit or fellow soul, seems to think and hold commune with us. 3 Thoreau expressed a similar interest in nature, spirit, and self: My desire to commune with the spirit of the universe - to be intoxicated even with the fumes, call it, of that divine nectar - to bear my head through atmospheres and over heights unknown to my feet - is perennial & constant. 4 Both writers pursued a notion of spirit as interrelated and integrated with the self and with the natural world

    From the very beginning of this project we have focused on taking a balanced approach to identifying the strengths and challenges of new immigrants in the Great Plains. Discussions of change in our world invariably focus on problems, and only occasionally on the strengths. We set out to look at new immigrants from a different perspective, in our view, a more realistic perspective: seeing the inherent strengths they possess as newcomers to our region and the gifts they bring, and examining the cultural assets the newcomers and the longer-term residents all can rely upon in working together to meet the inevitable challenges that an influx of new people brings to our world. What, then, have we learned about the new immigrants and the new immigrant families to the Heartland? What challenges do they face in their journey to create a safe, prosperous, and harmonious new home for themselves and their loved ones and friends? What strengths do they possess to accomplish these goals? What gifts do they bring to our region? Let\u27s go back to the original questions posed by the authors, and discuss what we have learned

    Spacecraft Orbital Debris Reentry: Aerothermal Analysis

    In the past 40 years, thousands of objects have been placed in Earth orbit and are being tracked. Space hardware reentry survivability must be evaluated to assess risks to human life and property on the ground. The objective of this paper is to present results of a study to determine altitude of demise (burn-up) or survivability of reentering objects. Two NASA/JSC computer codes - Object Reentry Survival Analysis Tool (ORSAT) and Miniature ORSAT (MORSAT) were used to determine trajectories, aerodynamic aerothermal environment, and thermal response of selected spacecraft components. The methodology of the two codes is presented, along with results of a parametric study of reentering objects modeled as spheres and cylinders. Parameters varied included mass, diameter, wall thickness, ballistic coefficient, length, type of material, and mode of tumbling/spinning. Two fragments of a spent Delta second stage undergoing orbital decay, stainless steel cylindrical propellant tank and titanium pressurization sphere, were evaluated with ORSAT and found to survive entry, as did the actual objects. Also, orbital decay reentry predictions of the Japanese Advanced Earth Observing Satellite (ADEOS) aluminum and nickel box-type components and the Russian COSMOS 954 satellite beryllium cylinders were made with MORSAT. These objects were also shown to survive reentry

    A life-course perspective of sex trafficking among the Bedia caste of India

    Thousands of Indian women and girls enter the commercial sex industry (CSI) annually based solely on membership in particular castes (e.g., Bedia, Nat). CSI-involved females bear the burden of sustaining entire family units on money earned in the sex trade; it is a life-long responsibility with negligible social status or personal indemnity. Based on the life-course developmental theory (Elder, Jr. 1994, 1998) this investigation was intended to examine trafficked women’s experiences within the commercial sex industry across time. Beyond the CSI, we were equally interested in experiences with factors that could promote well-being (i.e., social support) and normative developmental transitions including education attainment and motherhood. To that end, three questions were posed. First, to what extent do factors surrounding CSI entry and continued involvement differ through time among CSI-involved Bedia? Second, how do CSI-involved Bedia describe social network composition and perceived support through time? Finally, are differences detectable, through time, in CSI-involved Bedia women’s experiences with normative developmental transitions including education attainment and motherhood? Interview data were collected from 31 Bedia females (age range 17 – 65 years) residing in rural Madhya Pradesh, India. To examine change through time, participants were divided into cohorts based on age and time involved in the commercial sex industry. Data were then analyzed within and across cohorts with particular attention to cohort-related experiential differences. Policy implications and suggestions for continued research are presented

    Comparison of ORSAT and SCARAB Reentry Analysis Tools for a Generic Satellite Test Case

    Reentry analysis is essential to understanding the consequences of the full life cycle of a spacecraft. Since reentry is a key factor in spacecraft development, NASA and ESA have separately developed tools to assess the survivability of objects during reentry. Criteria such as debris casualty area and impact energy are particularly important to understanding the risks posed to people on Earth. Therefore, NASA and ESA have undertaken a series of comparison studies of their respective reentry codes for verification and improvements in accuracy. The NASA Object Reentry Survival Analysis Tool (ORSAT) and the ESA Spacecraft Atmospheric Reentry and Aerothermal Breakup (SCARAB) reentry analysis tools serve as standard codes for reentry survivability assessment of satellites. These programs predict whether an object will demise during reentry and calculate the debris casualty area of objects determined to survive, establishing the reentry risk posed to the Earth's population by surviving debris. A series of test cases have been studied for comparison and the most recent uses "Testsat," a conceptual satellite composed of generic parts, defined to use numerous simple shapes and various materials for a better comparison of the predictions of these two codes. This study is an improvement on the others in this series because of increased consistency in modeling techniques and variables. The overall comparison demonstrated that the two codes arrive at similar results. Either most objects modeled resulted in close agreement between the two codes, or if the difference was significant, the variance could be explained as a case of semantics in the model definitions. This paper presents the main results of ORSAT and SCARAB for the Testsat case and discusses the sources of any discovered differences. Discussion of the results of previous comparisons is made for a summary of differences between the codes and lessons learned from this series of tests

    Transdermal fluorescence detection of a dual fluorophore system for noninvasive point-of-care gastrointestinal permeability measurement

    The intestinal mucosal barrier prevents macromolecules and pathogens from entering the circulatory stream. Tight junctions in this barrier are compromised in inflammatory bowel diseases, environmental enteropathy, and enteric dysfunction. Dual sugar absorption tests are a standard method for measuring gastrointestinal integrity, however, these are not clinically amenable. Herein, we report on a dual fluorophore system and fluorescence detection instrumentation for which gastrointestinal permeability is determined in a rat small bowel disease model from the longitudinal measured transdermal fluorescence of each fluorophore. This fluorophore technology enables a specimen-free, noninvasive, point-of-care gastrointestinal permeability measurement which should be translatable to human clinical studies

    Diet, physical activity and behavioural interventions for the treatment of overweight or obese adolescents aged 12 to 17 years

    Adolescent overweight and obesity has increased globally, and can be associated with short- and long-term health consequences. Modifying known dietary and behavioural risk factors through behaviour changing interventions (BCI) may help to reduce childhood overweight and obesity. This is an update of a review published in 2009. To assess the effects of diet, physical activity and behavioural interventions for the treatment of overweight or obese adolescents aged 12 to 17 years. We performed a systematic literature search in: CENTRAL, MEDLINE, Embase, PsycINFO, CINAHL, LILACS, and the trial registers ClinicalTrials.gov and ICTRP Search Portal. We checked references of identified studies and systematic reviews. There were no language restrictions. The date of the last search was July 2016 for all databases. We selected randomised controlled trials (RCTs) of diet, physical activity and behavioural interventions for treating overweight or obesity in adolescents aged 12 to 17 years. Two review authors independently assessed risk of bias, evaluated the overall quality of the evidence using the GRADE instrument and extracted data following the guidelines of the Cochrane Handbook for Systematic Reviews of Interventions. We contacted trial authors for additional information. We included 44 completed RCTs (4781 participants) and 50 ongoing studies. The number of participants in each trial varied (10 to 521) as did the length of follow-up (6 to 24 months). Participants ages ranged from 12 to 17.5 years in all trials that reported mean age at baseline. Most of the trials used a multidisciplinary intervention with a combination of diet, physical activity and behavioural components. The content and duration of the intervention, its delivery and the comparators varied across trials. The studies contributing most information to outcomes of weight and body mass index (BMI) were from studies at a low risk of bias, but studies with a high risk of bias provided data on adverse events and quality of life.The mean difference (MD) of the change in BMI at the longest follow-up period in favour of BCI was -1.18 kg/m(2) (95% confidence interval (CI) -1.67 to -0.69); 2774 participants; 28 trials; low quality evidence. BCI lowered the change in BMI z score by -0.13 units (95% CI -0.21 to -0.05); 2399 participants; 20 trials; low quality evidence. BCI lowered body weight by -3.67 kg (95% CI -5.21 to -2.13); 1993 participants; 20 trials; moderate quality evidence. The effect on weight measures persisted in trials with 18 to 24 months' follow-up for both BMI (MD -1.49 kg/m(2) (95% CI -2.56 to -0.41); 760 participants; 6 trials and BMI z score MD -0.34 (95% CI -0.66 to -0.02); 602 participants; 5 trials).There were subgroup differences showing larger effects for both BMI and BMI z score in studies comparing interventions with no intervention/wait list control or usual care, compared with those testing concomitant interventions delivered to both the intervention and control group. There were no subgroup differences between interventions with and without parental involvement or by intervention type or setting (health care, community, school) or mode of delivery (individual versus group).The rate of adverse events in intervention and control groups was unclear with only five trials reporting harms, and of these, details were provided in only one (low quality evidence). None of the included studies reported on all-cause mortality, morbidity or socioeconomic effects.BCIs at the longest follow-up moderately improved adolescent's health-related quality of life (standardised mean difference 0.44 ((95% CI 0.09 to 0.79); P = 0.01; 972 participants; 7 trials; 8 comparisons; low quality of evidence) but not self-esteem.Trials were inconsistent in how they measured dietary intake, dietary behaviours, physical activity and behaviour. We found low quality evidence that multidisciplinary interventions involving a combination of diet, physical activity and behavioural components reduce measures of BMI and moderate quality evidence that they reduce weight in overweight or obese adolescents, mainly when compared with no treatment or waiting list controls. Inconsistent results, risk of bias or indirectness of outcome measures used mean that the evidence should be interpreted with caution. We have identified a large number of ongoing trials (50) which we will include in future updates of this review

    Association of fluorescent protein pairs and it's significant impact on fluorescence and energy transfer

    Fluorescent proteins (FPs) are commonly used in pairs to monitor dynamic biomolecular events through changes in proximity via distance dependent processes such as Förster resonance energy transfer (FRET). The impact of FP association is assessed by predicting dimerization sites in silico and stabilizing the dimers by bio‐orthogonal covalent linkages. In each tested case dimerization changes inherent fluorescence, including FRET. GFP homodimers demonstrate synergistic behavior with the dimer being brighter than the sum of the monomers. The homodimer structure reveals the chromophores are close with favorable transition dipole alignments and a highly solvated interface. Heterodimerization (GFP with Venus) results in a complex with ≈87% FRET efficiency, significantly below the 99.7% efficiency predicted. A similar efficiency is observed when the wild‐type FPs are fused to a naturally occurring protein–protein interface system. GFP complexation with mCherry results in loss of mCherry fluorescence. Thus, simple assumptions used when monitoring interactions between proteins via FP FRET may not always hold true, especially under conditions whereby the protein–protein interactions promote FP interaction

    The small molecule raptinal can simultaneously induce apoptosis and inhibit PANX1 activity

    Discovery of new small molecules that can activate distinct programmed cell death pathway is of significant interest as a research tool and for the development of novel therapeutics for pathological conditions such as cancer and infectious diseases. The small molecule raptinal was discovered as a pro-apoptotic compound that can rapidly trigger apoptosis by promoting the release of cytochrome c from the mitochondria and subsequently activating the intrinsic apoptotic pathway. As raptinal is very effective at inducing apoptosis in a variety of different cell types in vitro and in vivo, it has been used in many studies investigating cell death as well as the clearance of dying cells. While examining raptinal as an apoptosis inducer, we unexpectedly identified that in addition to its pro-apoptotic activities, raptinal can also inhibit the activity of caspase-activated Pannexin 1 (PANX1), a ubiquitously expressed transmembrane channel that regulates many cell death-associated processes. By implementing numerous biochemical, cell biological and electrophysiological approaches, we discovered that raptinal can simultaneously induce apoptosis and inhibit PANX1 activity. Surprisingly, raptinal was found to inhibit cleavage-activated PANX1 via a mechanism distinct to other well-described PANX1 inhibitors such as carbenoxolone and trovafloxacin. Furthermore, raptinal also interfered with PANX1-regulated apoptotic processes including the release of the 'find-me' signal ATP, the formation of apoptotic cell-derived extracellular vesicles, as well as NLRP3 inflammasome activation. Taken together, these data identify raptinal as the first compound that can simultaneously induce apoptosis and inhibit PANX1 channels. This has broad implications for the use of raptinal in cell death studies as well as in the development new PANX1 inhibitors
